利用甲基丙烯酸缩水甘油酯与乙二醇甲基丙烯酸酯经悬浮聚合反应合成亲水性亲和载体骨架,骨架上的不环氧基与氨水反应,继之与丁二酸酐反应,最后与配基联苯胺偶联制备亲和载体,配基密度为54μmol/g湿胶。亲和载体吸附激酶的最佳条件为PH4.6和2.5mol/LNaCl;测定了吸附等温线,算得离常数Kd为316IU/ml,了大吸附量qm为2.383×10^4U/mL湿胶,洗脱条件为含0.5mol/LNaCl  相似文献

聚苯乙烯载体亲和层析纯化尿激酶   总被引:2,自引:0,他引:2  
聚苯乙烯经磺化后连接手臂6-氨基己酸,然后偶联配基对氨基苯甲脒,制成亲水性的亲和载体。载体的最适配基密度为每克湿载体32μmol,最适吸附条件为pH7.5、0.05mol/L磷酸缓冲液(含NaCl0.5mol/L)。在pH4.0、0.1mol/L醋酸缓冲溶液(含NaCl0.5mol/L)中,采用0~1mol/L NaSCN梯度洗脱,可将比活8.33μmol·S-1·mg-1的粗酶纯化50倍,收率大于53%。  相似文献

用低浓度锌离子从人尿中沉淀尿激酶(UK),锌离子浓度为3mmol/L时,沉淀回收率为87%~93%,用pH10~10.5的20g/L硫酸铵按1∶4(g∶ml)溶解UK沉淀物,UK溶解得率为97%。溶解液用70%饱和度的硫酸铵盐析,UK盐析收率为91%,粗品纯度为4.08×105IU/g,固体效价为3.6×104IU/g,总收率在77%~82%之间。每吨尿(5.00×106IU计)可得UK粗品(3.8~4.1)×106IU。该粗品可用离子交换层析或亲和层析进一步纯化。  相似文献

将横纹东方豚卵巢与醋酸乙醇溶液混合匀浆,经离心,加热,沉淀,脱脂等处理,制得粗提液,活性炭吸附粗提液得到粗毒液;以壳聚糖为载体合成亲和微珠分别来分离纯化粗提液和粗毒液,用传统的小鼠生物法检测TTX毒素量.潮湿活性炭对TTX的吸附量约为2μg/g,潮湿亲和微珠吸附量约为10μg/g;亲和微珠吸附提取粗毒液,回收率达51%,直接吸附粗提液,回收率达到80%,同时蛋白也得到有效的去除.  相似文献

提出了两种不同用途的木聚糖酶的诱导合成方法。以里氏木霉为产酶菌,经适当处理后的玉米芯可诱导产生含纤维素酶(3.4IU/mL)的高活力木聚糖酶(54.4IU/mL)。以混有少量纤维素的粗木聚糖作碳源,通过分批补料及对培养条件的限制性控制里氏木霉可选择性合成木聚糖酶;选择性合成程度与碳源浓度有关,当碳源浓度为10g/L时木聚糖酶和纤维素酶活力分别为35.5IU/mL、0.2U/mL,两种酶活的比值达177.5。  相似文献

以壳聚糖为载体、肝素为配基和以琼脂糖4B为载体、肝素为配基分别合成2种亲和吸附剂,用以纯化猪凝血酶,并对两者的纯化效果进行比较.结果表明对于猪凝血酶的分离提纯,前者明显优于后者.前者的酶比活为1553.7U*mg-1,提纯11.5倍,收率为78.8%;后者的酶比活为1126.4U*mg-1,提纯8.8倍,收率为61.4%.前者无明显压床,流速均匀,而且前者的价格低廉,易于得到.  相似文献

含抗人尿激酶单克隆抗体的腹水经ProteinA-SepharoseCL4B柱纯化得到IgG纯品,分子量为150kD(重链55kD,轻链20kD),与CNBr-Sepharose4B偶联制成抗人尿激酶单克隆抗-体-Sepharose4B亲和层析介质。尿激酶粗品经单抗-Sepharose4B亲和层析柱纯化后,所得尿激酶制品比活为9×10-4~1.7×10-3mol/s·mg-1,纯化倍数为15~30倍,酶活性回收率在50%以上。  相似文献

啤酒废酵母(108-109细胞/ml)通气发酵后,经乙醇-氯仿沉淀,加热,调PH,(NH4)2SO4沉淀等除去杂蛋白质。最后经SephadexG-75层析得到纯化的CuZn-SOD,酶比活在1500单位/mg蛋白以上,酶的分子量为148000,由四个亚基组成,紫外最大吸收峰位于256um处。A256.4/280=1.22.  相似文献

The concept of nanopore analysis, using the pore-forming protein a-hemolysin to detect individual nucleic acids at a single-molecule level, was first proposed in 1996. Over the past two decades, tremendous progress has been made in the nanopore field, and nanopore analysis has become a label-free and high-throughput method for probing bio- molecules and other analytes with single-molecule sensi- tivity, especially holds the promising for "third generation" DNA sequencing. However, challenges still remain in the experimental strategies and the design of whole nanopore-based instruments. Here, we proudly present a special topic dedicated to the topic of "Nanopore Analysis", with 8 reviews/articles providing up to date coverage of the experimental strategies, theoretical calcu- lations and simulations, and instrument design. Reviews and articles on the experimental strategies cover control of DNA partitioning into a nanopore, detection of target DNA, and the advantages of nanopore-based DNA sequencing. The theoretical calculations and simulations discuss the translocation behavior of DNA, and an inte- grated measurement system and data analysis software are presented for instrument design.  相似文献

给出了利用双频观测值计算L3组合电离层延迟高阶项改正的方法,并与全球电离层延迟文件的改正效果进行对比.利用赤道附近的15个国际全球卫星导航定位系统服务组织(IGS)站的数据进行比较,结果表明:2种方法计算的电离层二阶项延迟互差最大不超过1 cm,三阶项延迟互差最大不超过5 mm;电离层高阶项改正后的观测值精密单点定位(PPP)解算结果N、E、U方向互差平均值分别为0.4、0.5、1.0mm,因此2种改正方法效果在同一水平.  相似文献
