以黄粉虫幼虫粉为原料,采用胰蛋白酶水解的方法,对原料中的蛋白质进行水解,对影响酶解反应的因素进行了选择和分析。适宜的反应条件为:黄粉虫与水的重量比为20∶60,酶作用量为黄粉虫粉重的0.5%,控制反应温度50℃,酶解时间8h,pH值为8.0。在此基础上,针对水解液有异味和缺乏Vc的问题,进行了水解液与果汁调配试验。结果表明:水解液与果汁按1∶1比例,添加饮料重量的6%蔗糖,0.2%黄原胶,0.3%CMC-Na,最终制得一种含果汁的复合氨基酸饮料。  相似文献

以浓缩乳清蛋白为原料,选择碱性蛋白酶(Alcalase)水解乳清蛋白。分析了pH、温度、酶和底物比(E/S)和反应时间等因素对乳清蛋白水解的影响。通过响应面法分析,确定了碱性蛋白酶(Alcalase)酶解乳清蛋白的最佳水解条件为:pH8.5、反应温度55℃、酶和底物比0.05。水解3.0h,水解度为19.34%。  相似文献

研究了几种蛋白酶对桑蛋蛹蛋白偻的水解情况,采用均匀设计法对桑蚕蛹蛋白粉的限制水解条件进行了选择,得到胰酶水解桑蚕蛹蛋白粉的优化条件为:酶用量占底物的质量分数为4.50%,底物含量的质量体积分数为1.5%,反应温度为40℃,水解时间为6.0h,水解P来8.0,在此条件下水解,酸溶性肽得率可达92.37%。  相似文献

克鲁维酵母y-85菊粉酶水解菊粉的研究及其中试   总被引:1,自引:0,他引:1  
克鲁维酵母(Kluveromycessp.)Y-85在菊粉提取液培养基中生长产菊粉酶,其细胞和胞外的菊粉酶活力分别约占菊粉酶总活力的76%和24%.试验中以酶发酵液作酶液.该酶的S/I值为15.2.酶液在50,55和60℃下保温1h,活力分别残留100%,96%和65%;在8℃下放置7d,14d,酶活力分别残留98%和96%.在pH3.6~7.0内,酶活性十分稳定.5mmolL-半胱氨酸和Fe+2分别可使酶活力提高10.4%和41.2%.y-85菊粉酶水解菊粉的适宜条件是:底物为总糖浓度10%~15%菊粉提取液,pH5.0,温度为55℃,酶用量为底物每克糖加酶800u(以蔗糖为底物测酶活力)或26.3u(以菊糖为底物测酶活力)、搅拌酶解6h.在上述条件上,底物降解率最高可达98.5%.酶解液中,果糖占总还原糖量的85%.用1100L罐进行酶解中试,5批试验,底物降解率平均达94.2%.  相似文献

紫孢侧耳、糙皮侧耳和佛罗里达侧耳的原生质体制备与再生的最佳条件为:用1%溶壁酶+0.5%蜗牛酶的混合酶液酶解培养3~4天的菌丝体,1.5小时后,原生质体产量均可达10 ̄(10)个/L;三种侧耳的最佳再生培养基分别为SM、MP和RP,再生率分别为0.61%、0.67%和0.76%。随着紫外线对原生质体处理时间的加长,再生率逐渐下降,当处理4min时,再生率接近零。  相似文献

对糖化酶在工业生产中影响酶活力的几个主要因素进行了试验研究,获得培养基最适浓度为20.25%,准确控制发酵培养基的标准体积、浓度及其适宜通风量(前期控制在1:0.40~0.47,中期为1:0.55~0.65,后期为1:0.5~0.55)是糖化酶生产中稳产高产的关键技术措施。结果表明:采取该措施提高酶活力47.32%,经济效益显著。  相似文献

从菊芋根际土壤中分离出一株菊粉酶(InulinaseE.C.3.2.1.7)产生菌,经物理和化学诱变得高酶活黑曲霉突变株AJ1958.该菌株在培养基(0.07g/mL菊芋提取液1000mL,豆饼粉5g,麸皮5g,KCI0.7g,FeSO4·7H2O0.01g,MgSO4·7H2O0.5g,PH5.0)中,于30℃振荡培养3d,酶活力可达64μmol·min-1.酶水解反应的最适PH为3.0~4.0,最适温度为60℃,在PH2.5~5.5的范围内和75℃以下较稳定.适宜条件下,10h内0.05g/mL的菊粉溶液中之底物几乎100%被酶水解.产物糖中果糖质量分数为93%,葡萄糖质量分数为5.6%.该突变株产生的菊粉酶具有较满意的水解性和热稳定性.  相似文献

以水解度和总氮回收率为指标,研究了单酶水解中国毛虾的工艺条件,分析了酶解温度、加酶量、料水比、酶解时间四因素对中国毛虾蛋白水解的影响。通过正交试验,确定最佳水解条件。综合考虑,木瓜蛋白酶水解中国毛虾的最适条件为:温度55℃,加酶量0.5%,料水比1:4,水解时间3h。在此条件下,中国毛虾的水解度为30.69%,总氮回收率为68.82%。  相似文献

为从紫菜蛋白中制备降血压肽,采用8种商业蛋白酶进行筛选,确定了以中性蛋白酶作为水解用酶和相应的酶解条件.研究了超声波处理紫菜蛋白后对酶解过程的影响,超声波处理后紫菜蛋白水溶液中游离氨基酸、酸溶性肽的质量浓度未变化,紫菜蛋白的紫外可见差示光谱也未出现变化;超声波处理的紫菜蛋白经酶解的产物,与未超声处理紫菜蛋白在同样条件酶解的产物相比,水解度增加1.1倍,IC50值降低29.5%.结果表明,超声处理没有破坏紫菜蛋白的分子结构,可能使它的分子空间构象发生变化,更加容易与酶分子结合,促进了酶解的进程.  相似文献

The discovery of the prolific Ordovician Red River reservoirs in 1995 in southeastern Saskatchewan was the catalyst for extensive exploration activity which resulted in the discovery of more than 15 new Red River pools. The best yields of Red River production to date have been from dolomite reservoirs. Understanding the processes of dolomitization is, therefore, crucial for the prediction of the connectivity, spatial distribution and heterogeneity of dolomite reservoirs.The Red River reservoirs in the Midale area consist of 3~4 thin dolomitized zones, with a total thickness of about 20 m, which occur at the top of the Yeoman Formation. Two types of replacement dolomite were recognized in the Red River reservoir: dolomitized burrow infills and dolomitized host matrix. The spatial distribution of dolomite suggests that burrowing organisms played an important role in facilitating the fluid flow in the backfilled sediments. This resulted in penecontemporaneous dolomitization of burrow infills by normal seawater. The dolomite in the host matrix is interpreted as having occurred at shallow burial by evaporitic seawater during precipitation of Lake Almar anhydrite that immediately overlies the Yeoman Formation. However, the low δ18O values of dolomited burrow infills (-5.9‰~ -7.8‰, PDB) and matrix dolomites (-6.6‰~ -8.1‰, avg. -7.4‰ PDB) compared to the estimated values for the late Ordovician marine dolomite could be attributed to modification and alteration of dolomite at higher temperatures during deeper burial, which could also be responsible for its 87Sr/86Sr ratios (0.7084~0.7088) that are higher than suggested for the late Ordovician seawaters (0.7078~0.7080). The trace amounts of saddle dolomite cement in the Red River carbonates are probably related to "cannibalization" of earlier replacement dolomite during the chemical compaction.  相似文献

本文叙述了对海南岛及其毗邻大陆边缘白垩纪到第四纪地层岩石进行古地磁研究的全部工作过程。通过分析岩石中剩余磁矢量的磁偏角及磁倾角的变化,提出海南岛白垩纪以来经历的构造演化模式如下:早期伴随顺时针旋转而向南迁移,后期伴随逆时针转动并向北运移。联系该地区及邻区的地质、地球物理资料,对海南岛上述的构造地体运动提出以下认识:北部湾内早期有一拉张作用,主要是该作用使湾内地壳显著伸长减薄,形成北部湾盆地。从而导致了海南岛的早期构造运动,而海南岛后期的构造运动则主要是受南海海底扩张的影响。海南地体运动规律的阐明对于了解北部湾油气盆地的形成演化有重要的理论和实际意义。  相似文献

The explosive growth of the Internet and database applications has driven database to be more scalable and available, and able to support on-line scaling without interrupting service. To support more client's queries without downtime and degrading the response time, more nodes have to be scaled up while the database is running. This paper presents the overview of scalable and available database that satisfies the above characteristics. And we propose a novel on-line scaling method. Our method improves the existing on-line scaling method for fast response time and higher throughputs. Our proposed method reduces unnecessary network use, i.e. , we decrease the number of data copy by reusing the backup data. Also, our on-line scaling operation can be processed parallel by selecting adequate nodes as new node. Our performance study shows that our method results in significant reduction in data copy time.  相似文献

R-Tree is a good structure for spatial searching. But in this indexing structure,either the sequence of nodes in the same level or sequence of traveling these nodes when queries are made is random. Since the possibility that the object appears in different MBR which have the same parents node is different, if we make the subnode who has the most possibility be traveled first, the time cost will be decreased in most of the cases. In some case, the possibility of a point belong to a rectangle will shows direct proportion with the size of the rectangle. But this conclusion is based on an assumption that the objects are symmetrically distributing in the area and this assumption is not always coming into existence. Now we found a more direct parameter to scale the possibility and made a little change on the structure of R-tree, to increase the possibility of founding the satisfying answer in the front sub trees. There are numerous geometric objects stored in the spatial databases. An importance function in a spatial database is that users can browse the geometric objects as a map efficiently. Thus the spatial database should display the geometric objects users concern about swiftly onto the display window. This process includes two operations:retrieve data from database and then draw them onto screen. Accordingly, to improve the efficiency, we should try to reduce time of both retrieving object and displaying them. The former can be achieved with the aid of spatial index such as R-tree, the latter require to simplify the objects. Simplification means that objects are shown with sufficient but not with unnecessary detail which depend on the scale of browse. So the major problem is how to retrieve data at different detail level efficiently. This paper introduces the implementation of a multi-scale index in the spatial database SISP (Spatial Information Shared Platform) which is generalized from R-tree. The difference between the generalization and the R-tree lies on two facets: One is that every node and geometric object in the generalization is assigned with a importance value which denote the importance of them, and every vertex in the objects are assigned with a importance value,too. The importance value can be use to decide which data should be retrieve from disk in a query. The other difference is that geometric objects in the generalization are divided into one or more sub-blocks, and vertexes are total ordered by their importance value. With the help of the generalized R-tree, one can easily retrieve data at different detail levels.Some experiments are performed on real-life data to evaluate the performance of solutions that separately use normal spatial index and multi-scale spatial index. The results show that the solution using multi-scale index in SISP is satisfying.  相似文献

The geographic information service is enabled by the advancements in general Web service technology and the focused efforts of the OGC in defining XML-based Web GIS service. Based on these models, this paper addresses the issue of services chaining,the process of combining or pipelining results from several interoperable GIS Web Services to create a customized solution. This paper presents a mediated chaining architecture in which a specific service takes responsibility for performing the process that describes a service chain. We designed the Spatial Information Process Language (SIPL) for dynamic modeling and describing the service chain, also a prototype of the Spatial Information Process Execution Engine (SIPEE) is implemented for executing processes written in SIPL. Discussion of measures to improve the functionality and performance of such system will be included.  相似文献
