靶向基因疫苗pcDNA3/MDC-VP1的构建及免疫效果   总被引:1,自引:0,他引:1  
用RT-PCR扩增小鼠巨噬细胞源趋化因子(macrophage-derived chemokine,MDC)基因,与柯萨奇病毒B组3型(CVB3)VP1基因通过一个编码10个氨基酸残基的接头序列(Gly4Ser)2相连,形成融合基因MDC-VP1,构建真核表达质粒pcDNA3/MDC-VP1,作为疫苗免疫BALB/c小鼠.3次免疫后,pcDNA3/MDC-VP1组小鼠血清中和抗体滴度明显高于pcDNA3/VP1组;而病毒滴度低于pcDNA3/VP1组.用10 LD50CVB3攻击,pcDNA3/MDC-VP1组小鼠生存率为50%,用Kaplan-Meier法进行生存分析,生存率高于其他各组.  相似文献

MEV-GDⅦ株脑内接种小鼠5~7天收获的脑悬液,经胰酶处理可获得高滴度HA抗原。本病毒接种BHK-21细胞后5天有细胞病变,而且病毒抗原阳性细胞可达90%。据此建立了HI和玻片EIA法检测GDⅦ血清抗体的方法。已应用于实验小鼠和豚鼠的常规检查。  相似文献

目的: 为建立脊灰病毒Sabin株I型特异的单克隆抗体.方法:以Vero细胞增殖病毒并用蔗糖密度梯度离心法纯化后,用病毒免疫SPF级Balb/c小鼠.取小鼠免疫脾淋巴细胞与SP2/0-Ag14骨髓瘤细胞进行细胞融合,通过间接 ELISA 法筛选抗体阳性的杂交瘤细胞,并用有限稀释法克隆化.用细胞培养法制备脊灰病毒单抗,并以中和试验测定单抗效价.结果:获得了8株分泌抗脊灰病毒Sabin株I型单克隆抗体的杂交瘤细胞株,其中1株G8G7能够稳定分泌抗体.间接ELISA 法测得单抗的效价为1∶8,中和试验测得中和抗体的效价为1∶58.结论:G8G7分泌的抗体为抗脊灰病毒Sabin株I型特异的单克隆抗体.  相似文献

脉瑞康大剂量对异丙肾上腺素(Isoprenaline,Isop)增加小鼠心肌耗氧有明显抑制作用,可显著延长小鼠耐缺氧存活时间(P<0.05);脉瑞康大剂量可致心肌损伤大鼠血清活性升高的天门冬氨酸氨基转换酶AST、磷酸肌酸激酶CPK显著降低(P<0.05),对活性升高的α-羟丁酸脱氢酶(α-HBDH)、乳酸脱氢酶(LDH)有一定降低作用,但无显著意义,组织病理学检查也显示心肌病理损伤减轻.  相似文献

将基因枪介导的DNA疫苗用于小鼠腹部免疫,并用免疫组化法检测gD抗原在接种部位的表达,利用ELISA检测血清中的抗gD抗体,并采用病毒攻击检测DNA疫苗对动物的保护效果。结果表明,基因枪可有效地将DNA质粒运送至小鼠皮肤组织,DNA疫苗携带的抗原保护基因gD2糖蛋白能在真皮和肌肉组织中高效表达。同时,免疫小鼠体内产生高滴度的中和抗体,能有效抵抗HSV2病毒的攻击。  相似文献

目的:观察全氟辛酸对小鼠肝脏相关酶系的影响。方法:将小鼠随机分为4组,分别为空白组、1/16LD50、1/8LD50和1/4LD50组,染毒14 d后采用试剂盒法测定肝组织匀浆中琥珀酸脱氢酶(SDH)、血清中乳酸脱氢酶(LDH)和血清中单胺氧化酶(MAO)含量的变化。结果:染毒组肝组织中SDH活性显著低于空白组(P0.01),随染毒剂量增加呈递减趋势;染毒组血清中LDH和MAO活性显著高于空白组(P0.01)。随染毒剂量增加呈递增趋势。结论:全氟辛酸通过损伤肝细胞线粒体,破坏能量代谢,进而损伤肝组织,可致肝坏死。  相似文献

目的:探讨油橄榄叶提取物(OLE)对铅中毒小鼠心肌乳酸脱氢酶活性和抗氧化能力的影响.方法:选健康小鼠,每日灌胃醋酸铅溶液的同时灌胃不同剂量的OLE进行治疗,连续用药30 d,检测心肌乳酸脱氢酶(LDH)、超氧化物歧化酶(SOD)、谷胱苷肽过氧化物酶(GSH-PX)活性和丙二醛(MDA)含量.结果:与模型对照组相比,小鼠灌胃OLE后心肌SOD和GSH-PX活性升高,LDH活性和MDA含量降低.结论:OLE对铅中毒小鼠有一定的疗效,能影响心肌ATP酶活性和抗氧化能力,阻止铅对心肌的毒性.  相似文献

目的了解龙葵浓缩汁对小鼠细胞免疫功能的影响.方法用3种浓度的龙葵口服液,即高剂量(3 000 mg/kg体质量),中剂量(1 500 mg/kg体质量),低剂量(750 mg/kg体质量)连续灌胃不同组别小鼠,28 d后分别采用Jerne改良玻片法进行小鼠的抗体生成细胞检测试验;MTT法进行小鼠的淋巴细胞转化实验;乳酸脱氢酶测定进行小鼠NK细胞活性测定试验.结果小鼠抗体生成细胞数、NK细胞活性在高剂量组与对照组间比较有显著性差异(P<0.05),中、低剂量组与对照组间比较无显著性差异(P>0.05);吞噬率、吞噬指数及淋巴细胞的增殖能力在低、中、高剂量组与对照组间比较均无显著性差异(P>0.05).结论高剂量的龙葵浓缩汁能明显增加小鼠的抗体生成细胞数,增强小鼠NK细胞的活性;而龙葵浓缩汁对小鼠巨噬细胞吞噬功能及对ConA诱导的小鼠淋巴细胞转化的能力无影响.  相似文献

Language is a means of verbal communication. People use language to communicate with each other. In the society, no two speakers are exactly alike in the way of speaking. Some differences are due to age, gender, statue and personality. Above all, gender is one of the obvious reasons. The writer of this paper tries to describe the features of women's language from these perspectives: pronunciation, intonation, diction, subjects, grammar and discourse. From the discussion of the features of women's language, more attention should be paid to language use in social context. The discovery of the prolific Ordovician Red River reservoirs in 1995 in southeastern Saskatchewan was the catalyst for extensive exploration activity which resulted in the discovery of more than 15 new Red River pools. The best yields of Red River production to date have been from dolomite reservoirs. Understanding the processes of dolomitization is, therefore, crucial for the prediction of the connectivity, spatial distribution and heterogeneity of dolomite reservoirs.The Red River reservoirs in the Midale area consist of 3~4 thin dolomitized zones, with a total thickness of about 20 m, which occur at the top of the Yeoman Formation. Two types of replacement dolomite were recognized in the Red River reservoir: dolomitized burrow infills and dolomitized host matrix. The spatial distribution of dolomite suggests that burrowing organisms played an important role in facilitating the fluid flow in the backfilled sediments. This resulted in penecontemporaneous dolomitization of burrow infills by normal seawater. The dolomite in the host matrix is interpreted as having occurred at shallow burial by evaporitic seawater during precipitation of Lake Almar anhydrite that immediately overlies the Yeoman Formation. However, the low δ18O values of dolomited burrow infills (-5.9‰~ -7.8‰, PDB) and matrix dolomites (-6.6‰~ -8.1‰, avg. -7.4‰ PDB) compared to the estimated values for the late Ordovician marine dolomite could be attributed to modification and alteration of dolomite at higher temperatures during deeper burial, which could also be responsible for its 87Sr/86Sr ratios (0.7084~0.7088) that are higher than suggested for the late Ordovician seawaters (0.7078~0.7080). The trace amounts of saddle dolomite cement in the Red River carbonates are probably related to "cannibalization" of earlier replacement dolomite during the chemical compaction.  相似文献

理论推导与室内实验相结合,建立了低渗透非均质砂岩油藏启动压力梯度确定方法。首先借助油藏流场与电场相似的原理,推导了非均质砂岩油藏启动压力梯度计算公式。其次基于稳定流实验方法,建立了非均质砂岩油藏启动压力梯度测试方法。结果表明:低渗透非均质砂岩油藏的启动压力梯度确定遵循两个等效原则。平面非均质油藏的启动压力梯度等于各级渗透率段的启动压力梯度关于长度的加权平均;纵向非均质油藏的启动压力梯度等于各渗透率层的启动压力梯度关于渗透率与渗流面积乘积的加权平均。研究成果可用于有效指导低渗透非均质砂岩油藏的合理井距确定,促进该类油藏的高效开发。  相似文献

There are numerous geometric objects stored in the spatial databases. An importance function in a spatial database is that users can browse the geometric objects as a map efficiently. Thus the spatial database should display the geometric objects users concern about swiftly onto the display window. This process includes two operations:retrieve data from database and then draw them onto screen. Accordingly, to improve the efficiency, we should try to reduce time of both retrieving object and displaying them. The former can be achieved with the aid of spatial index such as R-tree, the latter require to simplify the objects. Simplification means that objects are shown with sufficient but not with unnecessary detail which depend on the scale of browse. So the major problem is how to retrieve data at different detail level efficiently. This paper introduces the implementation of a multi-scale index in the spatial database SISP (Spatial Information Shared Platform) which is generalized from R-tree. The difference between the generalization and the R-tree lies on two facets: One is that every node and geometric object in the generalization is assigned with a importance value which denote the importance of them, and every vertex in the objects are assigned with a importance value,too. The importance value can be use to decide which data should be retrieve from disk in a query. The other difference is that geometric objects in the generalization are divided into one or more sub-blocks, and vertexes are total ordered by their importance value. With the help of the generalized R-tree, one can easily retrieve data at different detail levels.Some experiments are performed on real-life data to evaluate the performance of solutions that separately use normal spatial index and multi-scale spatial index. The results show that the solution using multi-scale index in SISP is satisfying.  相似文献
