应用RAPD标记构建马尾松单株树遗传连锁图谱   总被引:5,自引:0,他引:5  
利用马尾松(Pinusmassoniana)崇义群体C-16号单株上40粒种子的胚乳为作图群体材料,用随机扩增多态性DNA(RAPD)方法构建马尾松单株遗传连锁图谱,从被步筛选的80个引物中,得到16个具有多态性产物的引物,对这16种引物进行Mendelian1:1分离检测(卡方检验,P<0.05),我们得到符合Mendelian1:1分离比例的RAPDs标记49个.通过对49个标记的连锁关系进行分析,其中29个标记分布在12个连锁群上,总图距为483.3cM,平均图距为28.42cM,20个标记没有连锁到任何连锁群上,需要继续进行大量标记的连续分析,构建完善的高密度的遗传图谱,使之能够对数量性状位点QTLs进行定位,从而进行分子标记辅助选择育种(MAS).  相似文献

以随机扩增多态性DNA(RAPD)技术为手段,对生野鲫,雄性异育银鲫及其杂交FI代(选择性状接近父本的5%F1代),父本回交F1你进行了遗传多态分析,从150个随机序列引物中筛选得到9种引物,并用它们可得到36个用作遗传标记的RAPD多态性条带,其中的22条父本显性条带均遗传子代,而14条母本性条带则均未被遗传子代,对两亲本和子代中的RAPD标记条带的分布情况及相似系数分析,以及聚类分析结果均支持  相似文献

将RAPD标记转换为SCAR标记是更到更为可靠而有有标记的有效方法,应用集群分离体分析(BSA)方法,筛选了900个100个碱基随机引物,得到了2个与单显性大豆花叶病毒抗性基因Rsa连锁的显性RAPD标记OPAS-061800和OPW-05600,将与Rsa连锁距离较近的OPW-05660克隆并测定DNA序列,根据这个序列的两端设计了1对特异的寡聚核苷酸引物,用这对引物进行扩增,在抗感亲本间观察到  相似文献

选用40个随机引物对杉木属的两个种杉木(CunninghamialanceolataHook)和德昌杉木(CunninghamiaunicanaliculataD.Y.WangetH.L.Liu)进行了RAPD分析,在两个杉木种中共获得了182个RAPD位点,其中98个为多态位点.多态位点达到53.84%.两个杉木种的遗传相似度S=0.64418,它们的遗传距离为0.355802.杉木与德昌杉木的遗传相似度略低于我们所作的大陆其它杉木种源区间的平均遗传相似度.从两个种间的遗传相似度和遗传距离来看,将德昌杉木单独定为一个新种是不确切的,作者赞成将德昌杉木认定为一个地方居群的观点.  相似文献

采用随机扩增多态性DNA技术对中国特有科特有属植物大血藤(Sargentodoxacuneata)进行分析,探讨植物地带性分化的分子差异.从100个随机引物中筛选出11个有效的10碱基对寡聚核糖核苷酸作引物,用于对取自5省区的大血藤植物基因组DNA进行PCR扩增;用UPGMA法对各居群间的Jacard相似性系数和遗传距离进行聚类分析.RAPD分析结果表明:分布于各省区(地区)的大血藤居群,尽管其形态特征的变异尺度较为一致,但在分子水平上存在明显差异,这种差异与地理范围的变化相关,表现出一定的地带性分化规律.  相似文献

应用RAPD技术分析了猕猴属(Macaca)种间和亚种间6个分类群之间的遗传多态性,共筛选出16个随机引物每个分类群平均获得77个遗传标记,根据遗传距离构建了系统发育进化树,结果显示从RAPD数据分析得到的6个分类群的相互关系与形态、同功酶、mtDNA的研究结果基本一致.表明RAPD技术适用于猕猴属中近缘种间和亚种间的分类分析.  相似文献

以四倍体彩色马铃薯(‘黑美人’,‘MIN-021’)杂交F1代单株无性株系群体为材料,利用SSR分子标记技术及作图软件(TetraploidMap)进行了彩色马铃薯分子遗传连锁图谱的构建研究.结果表明:从255对SSR引物中筛选出34对条带清晰稳定、多态性丰富的适宜引物,对‘黑美人’בMIN-021’杂种F_1的210个单株无性系及其双亲的基因组DNA进行PCR扩增共得到201个多态性标记.用这201个SSR标记对双亲分别作图,首次构建了四倍体彩色马铃薯的分子遗传连锁框架图谱.母本图谱含129个标记,12个连锁群总长度为1 167cM,标记间平均距离为9.04cM;父本图谱含131个标记,12个连锁群总长度为1 187cM,标记间平均距离为9.06cM.  相似文献

利用随机扩增多态生DNA(RAPD)技术对裙带菜9个品系的种质类进行分析,共筛选出18个引物扩增出221条清晰可重复的带。进一步分析表明,9个裙带菜品都保持着一定的遗传离距,且有6个品系具有可作为遗传标记的特征带。上述结果在NDA水平上提供了裙带菜品进行杂交组配时远缘亲本选择的理论依据,并初步证明了利用RAPD技术对裙带菜进行种质来源分析和种质差异鉴定的有效性。  相似文献

The discovery of the prolific Ordovician Red River reservoirs in 1995 in southeastern Saskatchewan was the catalyst for extensive exploration activity which resulted in the discovery of more than 15 new Red River pools. The best yields of Red River production to date have been from dolomite reservoirs. Understanding the processes of dolomitization is, therefore, crucial for the prediction of the connectivity, spatial distribution and heterogeneity of dolomite reservoirs.The Red River reservoirs in the Midale area consist of 3~4 thin dolomitized zones, with a total thickness of about 20 m, which occur at the top of the Yeoman Formation. Two types of replacement dolomite were recognized in the Red River reservoir: dolomitized burrow infills and dolomitized host matrix. The spatial distribution of dolomite suggests that burrowing organisms played an important role in facilitating the fluid flow in the backfilled sediments. This resulted in penecontemporaneous dolomitization of burrow infills by normal seawater. The dolomite in the host matrix is interpreted as having occurred at shallow burial by evaporitic seawater during precipitation of Lake Almar anhydrite that immediately overlies the Yeoman Formation. However, the low δ18O values of dolomited burrow infills (-5.9‰~ -7.8‰, PDB) and matrix dolomites (-6.6‰~ -8.1‰, avg. -7.4‰ PDB) compared to the estimated values for the late Ordovician marine dolomite could be attributed to modification and alteration of dolomite at higher temperatures during deeper burial, which could also be responsible for its 87Sr/86Sr ratios (0.7084~0.7088) that are higher than suggested for the late Ordovician seawaters (0.7078~0.7080). The trace amounts of saddle dolomite cement in the Red River carbonates are probably related to "cannibalization" of earlier replacement dolomite during the chemical compaction.  相似文献

本文叙述了对海南岛及其毗邻大陆边缘白垩纪到第四纪地层岩石进行古地磁研究的全部工作过程。通过分析岩石中剩余磁矢量的磁偏角及磁倾角的变化,提出海南岛白垩纪以来经历的构造演化模式如下:早期伴随顺时针旋转而向南迁移,后期伴随逆时针转动并向北运移。联系该地区及邻区的地质、地球物理资料,对海南岛上述的构造地体运动提出以下认识:北部湾内早期有一拉张作用,主要是该作用使湾内地壳显著伸长减薄,形成北部湾盆地。从而导致了海南岛的早期构造运动,而海南岛后期的构造运动则主要是受南海海底扩张的影响。海南地体运动规律的阐明对于了解北部湾油气盆地的形成演化有重要的理论和实际意义。  相似文献

The explosive growth of the Internet and database applications has driven database to be more scalable and available, and able to support on-line scaling without interrupting service. To support more client's queries without downtime and degrading the response time, more nodes have to be scaled up while the database is running. This paper presents the overview of scalable and available database that satisfies the above characteristics. And we propose a novel on-line scaling method. Our method improves the existing on-line scaling method for fast response time and higher throughputs. Our proposed method reduces unnecessary network use, i.e. , we decrease the number of data copy by reusing the backup data. Also, our on-line scaling operation can be processed parallel by selecting adequate nodes as new node. Our performance study shows that our method results in significant reduction in data copy time.  相似文献

R-Tree is a good structure for spatial searching. But in this indexing structure,either the sequence of nodes in the same level or sequence of traveling these nodes when queries are made is random. Since the possibility that the object appears in different MBR which have the same parents node is different, if we make the subnode who has the most possibility be traveled first, the time cost will be decreased in most of the cases. In some case, the possibility of a point belong to a rectangle will shows direct proportion with the size of the rectangle. But this conclusion is based on an assumption that the objects are symmetrically distributing in the area and this assumption is not always coming into existence. Now we found a more direct parameter to scale the possibility and made a little change on the structure of R-tree, to increase the possibility of founding the satisfying answer in the front sub trees. There are numerous geometric objects stored in the spatial databases. An importance function in a spatial database is that users can browse the geometric objects as a map efficiently. Thus the spatial database should display the geometric objects users concern about swiftly onto the display window. This process includes two operations:retrieve data from database and then draw them onto screen. Accordingly, to improve the efficiency, we should try to reduce time of both retrieving object and displaying them. The former can be achieved with the aid of spatial index such as R-tree, the latter require to simplify the objects. Simplification means that objects are shown with sufficient but not with unnecessary detail which depend on the scale of browse. So the major problem is how to retrieve data at different detail level efficiently. This paper introduces the implementation of a multi-scale index in the spatial database SISP (Spatial Information Shared Platform) which is generalized from R-tree. The difference between the generalization and the R-tree lies on two facets: One is that every node and geometric object in the generalization is assigned with a importance value which denote the importance of them, and every vertex in the objects are assigned with a importance value,too. The importance value can be use to decide which data should be retrieve from disk in a query. The other difference is that geometric objects in the generalization are divided into one or more sub-blocks, and vertexes are total ordered by their importance value. With the help of the generalized R-tree, one can easily retrieve data at different detail levels.Some experiments are performed on real-life data to evaluate the performance of solutions that separately use normal spatial index and multi-scale spatial index. The results show that the solution using multi-scale index in SISP is satisfying.  相似文献

The geographic information service is enabled by the advancements in general Web service technology and the focused efforts of the OGC in defining XML-based Web GIS service. Based on these models, this paper addresses the issue of services chaining,the process of combining or pipelining results from several interoperable GIS Web Services to create a customized solution. This paper presents a mediated chaining architecture in which a specific service takes responsibility for performing the process that describes a service chain. We designed the Spatial Information Process Language (SIPL) for dynamic modeling and describing the service chain, also a prototype of the Spatial Information Process Execution Engine (SIPEE) is implemented for executing processes written in SIPL. Discussion of measures to improve the functionality and performance of such system will be included.  相似文献
