【目的】气味结合蛋白(odorant binding proteins,OBPs)是一种嗅觉相关的蛋白,该蛋白参与大多数气味分子的识别过程,并与气味分子相结合。获得杨小舟蛾[Micromelalopha troglodyta (Graeser)]OBPs以明确其特性。【方法】选择羽化后健康的杨小舟蛾触角为模板,通过反转录聚合酶链反应(RT-PCR)技术克隆杨小舟蛾MtroOBP1基因,利用生物信息学分析软件研究其基因序列和蛋白结构,运用实时荧光定量PCR(qPCR)技术研究MtroOBP1的组织表达模式。【结果】利用RT-PCR技术从杨小舟蛾触角总RNA中扩增得到MtroOBP1基因(GenBank登录号:MN056510),序列分析表明,MtroOBP1开放阅读框为777 bp,一共编码了258个氨基酸残基,且翻译的氨基酸序列仅含有4个保守的半胱氨酸位点,表明得到的OBP基因的编码蛋白不属于典型气味结合蛋白家族,而是属于Minus-C家族。蛋白质理化性质预测显示:MtroOBP1蛋白的分子量为29 664.57 u,等电点为6.23,有18个潜在的磷酸化位点,没有明显的跨膜区,疏水指数为-2.011~3.078,有一个由19个氨基酸组成的信号肽,说明为分泌型蛋白。组织表达模式表明MtroOBP1在杨小舟蛾的各部位都表达,但在触角中表达量最高。【结论】首次克隆得到杨小舟蛾MtroOBP1基因,在触角中高表达,推测其蛋白具有运输气味分子的功能,在杨小舟蛾的嗅觉识别中起着至关重要的作用。  相似文献

从麻疯树基因组中克隆到一个CBF2基因(命名为JcCBF2).运用生物信息学的分析手段对该基因序列及其编码的蛋白质序列进行了分析,结果表明JcCBF2蛋白中不止包含保守的AP2/EREBP结合结构域,还具有CBF转录因子特征序列PKK/RPAGRxKFxETRHP、DSAWR.实时荧光定量PCR结果显示,JcCBF2基因主要在麻疯树叶片内转录表达,对低温胁迫极其敏感,但对干旱及盐胁迫不敏感,初步研究表明JcCBF2基因是低温诱导型转录因子.  相似文献

为初步了解南亚实蝇对引诱剂的感受机制,笔者基于南亚实蝇转录组数据进行了气味结合蛋白(odorant-binding protein,简称OBP)家族基因的筛选和克隆验证.共筛选出OBPs基因32条,成功克隆出26条,对其编码蛋白的结构分析表明,典型OBPs 21条,"Dimer"OBPs 1条,Minus-OBPs、"Plus-C"OBPs各2条.碱基序列分析和蛋白进化分析表明,这26条基因与瓜实蝇OBPs高度相似,其编码的蛋白质与瓜实蝇亲缘关系最近,有16条与瓜实蝇聚类数值达90以上.这为下一步南亚实蝇OBPs高级结构和功能的研究提供了基础数据,也为实蝇引诱机制的探讨提供了理论依据.  相似文献

从麻疯树胚乳cDNA文库中克隆得到一个C2H2型锌指蛋白基因,JcZFP1,全长931bp,开放阅读框全长735bp,编码244个氨基酸残基,等电点pI=7.0,分子量PA=27.38kD.C2H2型锌指结构位于第81至103个氨基酸残基中,包括植物锌指蛋白特有的保守氨基酸序列QALGGH.在该基因的顺式作用元件上发现了与生长发育和逆境胁迫相关的启动元件.荧光定量PCR(qPCR)检测发现,JcZFP1基因在麻疯树各器官中的表达量依次为:绿果皮花叶根茎种仁,且幼嫩器官表达量高于成熟器官;对麻疯树幼苗进行低温、干旱胁迫时,JcZFP1基因的表达量明显增加,表明该基因可能在麻疯树的生长发育及逆境胁迫应答过程中起重要作用.  相似文献

铜蓝蛋白(Ceruloplasmin,Cp)是一种重要的铜转运蛋白,合成于肝脏并参与生物体铁的代谢,在医学上是各种炎症、感染、中毒及癌症疾病的标志性蛋白.铜蓝蛋白的研究已在多种真骨鱼类中被报道,文中第一次在稀有鮈鲫(Gobiocypris rarus)中报道此基因.采用cDNA末端快速扩增技术(rapid amplification of cDNA ends,RACE)克隆了稀有鮈鲫铜蓝蛋白基因,使用荧光定量PCR的方法构建了该基因组织表达谱.序列分析表明稀有鮈鲫铜蓝蛋白基因包含3 264bp全长编码序列,该序列编码1 087个氨基酸,其核苷酸和氨基酸序列与斑马鱼同源性最高(分别为88.1%和90.3%).理论相对分子质量和等电点分别为124 429.1D和6.41.荧光定量PCR检测表明该基因在肝脏和脾脏中相对表达量最高,在肌肉和鳃中相对表达量最低.使用氨基酸序列进行蛋白结构保守域分析,结果表明铜蓝蛋白基因在脊椎动物中是相对保守的,推测其功能也与其他物种相似.这为进一步研究稀有鮈鲫该基因的功能及其应用奠定了基础.  相似文献

运用mRNA差异显示技术(DDRT-PCR),并结合反向Northern杂交筛选麻疯树(Jatropha.curcus)干旱胁迫下差异表达基因,最终从麻疯树叶片中分离出8条干旱胁迫诱导下表达上调的差异片段.经克隆、测序和基因信息分析后发现其中4个片段与GenBank中已知序列有较高同源性.对这4个差异表达片段的序列分析表明:JcDD-3和JcDD-13分别与1-磷酸-肌醇合成酶(MIPS)基因及甜菜碱醛脱氢酶(BADH)基因序列有较高同源性;JcDD-16与白杨中度抗旱叶片中提取的表达序列标签(CU230383)核酸序列同源;JcDD-25与植物富含亮氨酸重复序列型类受体蛋白激酶(LRR-RLK)基因序列同源.  相似文献

利用抑制消减杂交法从芸香科柑橘属不耐寒植物佛手中分离得到了一个表达序列标签(EST)片段,结合RACE技术克隆获得该基因的1 669 bp序列,编码区长1 236 bp,编码413个氨基酸.通过Blastn同源序列比对分析,结果显示该基因与拟南芥已知的GRAS基因同源性较高;与GenBank数据库比对分析,表明该基因具有GRAS特有的保守结构域.因此,命名该基因为:CmsGRAS(GenBank登录号:JF440647).用荧光定量聚合酶链式反应(qPCR)的方法研究了该基因在低温胁迫处理下的表达特性,结果显示该基因在低温胁迫后的表达量有明显的变化.  相似文献

在日本鳗鲡(Anguilla japonica)中克隆获得2个γ-干扰素诱导的溶酶体巯基还原酶(GILT)基因AjGILT-1和AjGILT-2,并对其序列进行生物信息学分析,进而利用实时荧光定量PCR分别检测2个基因在不同组织器官中的表达水平,及其在脂多糖(LPS)、聚肌胞苷酸(PolyI:C)刺激和迟缓爱德华氏菌(Edwardsiella tarda)感染后的表达变化.结果显示:2个基因编码的蛋白序列中均含有GILT家族的特征序列CQHGX2ECX2NX4C、活性位点CXXC、2个糖基化位点和11个保守的半胱氨酸残基,基因组序列均含7个外显子和6个内含子.2个基因启动子中都存在多个转录因子结合位点,如γ-干扰素活化位点以及特化蛋白1结合位点;不同的是AjGILT-1启动子区存在多个核因子-1、激活蛋白-1结合位点及干扰素刺激反应元件,而在AjGILT-2启动子区未发现.AjGILT-1在鳃和肠中表达量最高,而AjGILT-2在性腺、头肾和脾脏中表达量最高;除肠道外,同一组织中AjGILT-2的表达量均高于AjGILT-1.LPS、PolyI:C刺激和E.tarda感染后,AjGILT-1与AjGILT-2的表达在鳃中仅有少数变化,而在头肾和中肾中则变化较明显.综上所述,AjGILT-1和AjGILT-2基因可能参与了日本鳗鲡受病毒和细菌感染后的免疫应答.  相似文献

对四川猪链球菌2型(S.suis 2)强毒株05ZYH33溶血素样蛋白(hemolysin-like protein,HLP)编码基因hlp进行序列信息分析、分子克隆表达及溶血活性检测,深入探讨HLP的致病机制.用Blast和Clustal X程序对HLP进行基因同源性分析,用Signal P 4.1和TMHMM Server 2.0分析HLP氨基酸序列.设计合成hlp引物进行PCR扩增,先后将hlp克隆入p MD-18T载体和p ET30a表达载体中,构建p ET30a::hly原核表达质粒.将重组质粒转化至E.coli BL21中诱导表达,并对重组HLP蛋白进行纯化和溶血活性检测.同源性分析表明HLP与多种具有溶血活性的蛋白相似性较高.氨基酸序列分析发现HLP具有信号肽和跨膜区,且由DUF21、CBS和Cor CHly C 3部分组成.序列测定显示hly片段全长1 335 bp,编码445个氨基酸.重组质粒经诱导表达和纯化后电泳显示,HLP分子量约为70 k Da,具有溶血活性.结果表明,HLP是一个膜结合蛋白,不同于能够分泌到细胞外的溶血素Suilysin.重组HLP具有一定的溶血效价,此可能与致病相关.  相似文献

本研究采用RT-PCR技术克隆了药材甲热激蛋白60(Heat shock protein 60,Hsp60)基因的cDNA全长序列,命名为SpHsp60(GenBank登录号:KX778623).氨基酸序列分析表明SpHsp60具有Hsp60基因家族高度保守的基序,实时定量PCR技术检测了不同温度胁迫后该基因的表达量,结果表明:-5℃和0℃低温处理2h,药材甲成虫体内的SpHsp60的表达量均显著高于对照组,且42℃高温处理2h后的表达量也显著高于对照组.说明SpHSP60与药材甲应对极端温度胁迫相关.  相似文献

The discovery of the prolific Ordovician Red River reservoirs in 1995 in southeastern Saskatchewan was the catalyst for extensive exploration activity which resulted in the discovery of more than 15 new Red River pools. The best yields of Red River production to date have been from dolomite reservoirs. Understanding the processes of dolomitization is, therefore, crucial for the prediction of the connectivity, spatial distribution and heterogeneity of dolomite reservoirs.The Red River reservoirs in the Midale area consist of 3~4 thin dolomitized zones, with a total thickness of about 20 m, which occur at the top of the Yeoman Formation. Two types of replacement dolomite were recognized in the Red River reservoir: dolomitized burrow infills and dolomitized host matrix. The spatial distribution of dolomite suggests that burrowing organisms played an important role in facilitating the fluid flow in the backfilled sediments. This resulted in penecontemporaneous dolomitization of burrow infills by normal seawater. The dolomite in the host matrix is interpreted as having occurred at shallow burial by evaporitic seawater during precipitation of Lake Almar anhydrite that immediately overlies the Yeoman Formation. However, the low δ18O values of dolomited burrow infills (-5.9‰~ -7.8‰, PDB) and matrix dolomites (-6.6‰~ -8.1‰, avg. -7.4‰ PDB) compared to the estimated values for the late Ordovician marine dolomite could be attributed to modification and alteration of dolomite at higher temperatures during deeper burial, which could also be responsible for its 87Sr/86Sr ratios (0.7084~0.7088) that are higher than suggested for the late Ordovician seawaters (0.7078~0.7080). The trace amounts of saddle dolomite cement in the Red River carbonates are probably related to "cannibalization" of earlier replacement dolomite during the chemical compaction.  相似文献

本文叙述了对海南岛及其毗邻大陆边缘白垩纪到第四纪地层岩石进行古地磁研究的全部工作过程。通过分析岩石中剩余磁矢量的磁偏角及磁倾角的变化,提出海南岛白垩纪以来经历的构造演化模式如下:早期伴随顺时针旋转而向南迁移,后期伴随逆时针转动并向北运移。联系该地区及邻区的地质、地球物理资料,对海南岛上述的构造地体运动提出以下认识:北部湾内早期有一拉张作用,主要是该作用使湾内地壳显著伸长减薄,形成北部湾盆地。从而导致了海南岛的早期构造运动,而海南岛后期的构造运动则主要是受南海海底扩张的影响。海南地体运动规律的阐明对于了解北部湾油气盆地的形成演化有重要的理论和实际意义。  相似文献

The explosive growth of the Internet and database applications has driven database to be more scalable and available, and able to support on-line scaling without interrupting service. To support more client's queries without downtime and degrading the response time, more nodes have to be scaled up while the database is running. This paper presents the overview of scalable and available database that satisfies the above characteristics. And we propose a novel on-line scaling method. Our method improves the existing on-line scaling method for fast response time and higher throughputs. Our proposed method reduces unnecessary network use, i.e. , we decrease the number of data copy by reusing the backup data. Also, our on-line scaling operation can be processed parallel by selecting adequate nodes as new node. Our performance study shows that our method results in significant reduction in data copy time.  相似文献

R-Tree is a good structure for spatial searching. But in this indexing structure,either the sequence of nodes in the same level or sequence of traveling these nodes when queries are made is random. Since the possibility that the object appears in different MBR which have the same parents node is different, if we make the subnode who has the most possibility be traveled first, the time cost will be decreased in most of the cases. In some case, the possibility of a point belong to a rectangle will shows direct proportion with the size of the rectangle. But this conclusion is based on an assumption that the objects are symmetrically distributing in the area and this assumption is not always coming into existence. Now we found a more direct parameter to scale the possibility and made a little change on the structure of R-tree, to increase the possibility of founding the satisfying answer in the front sub trees. The geographic information service is enabled by the advancements in general Web service technology and the focused efforts of the OGC in defining XML-based Web GIS service. Based on these models, this paper addresses the issue of services chaining,the process of combining or pipelining results from several interoperable GIS Web Services to create a customized solution. This paper presents a mediated chaining architecture in which a specific service takes responsibility for performing the process that describes a service chain. We designed the Spatial Information Process Language (SIPL) for dynamic modeling and describing the service chain, also a prototype of the Spatial Information Process Execution Engine (SIPEE) is implemented for executing processes written in SIPL. Advances in wireless technologies and positioning technologies and spread of wireless devices, an interest in LBS (Location Based Service) is arising. To provide location based service, tracking data should have been stored in moving object database management system (called MODBMS) with proper policies and managed efficiently. So the methods which acquire the location information at regular time intervals then, store and manage have been studied. In this paper, we suggest tracking data management techniques using topology that is corresponding to the moving path of moving object. In our techniques, we update the MODBMS when moving object arrived at a street intersection or a curved road which is represented as the node in topology and predict the location at past and future with attribute of topology and linear function. In this technique, location data that are corresponding to the node in topology are stored, thus reduce the number of update and amount of data. Also in case predicting the location,because topology are used as well as existing location information, accuracy for prediction is increased than applying linear function or spline function.  相似文献
