以褐藻酸钠作为褐藻胶裂解酶产生菌初筛培养基唯一碳源,从鲍鱼养殖水样中分离获得一株产褐藻胶裂解酶的微生物,形态学和分子生物学16S rDNA鉴定结果显示,该菌株为交替假单胞菌Pseudoalteromonas sp.BYS-2.对该菌株进行产酶条件研究,获得最适产酶配方为:褐藻酸钠1 g·L~(-1),葡萄糖0.5 g·L~(-1),酵母膏10g·L~(-1),黄豆饼粉3 g·L~(-1),(NH_4)_2HPO_4 3 g·L~(-1),初始培养基pH8.0;最佳产酶条件为:接种量2%(体积分数),装液量50 m L/250 m L,发酵温度20℃,摇床转速200 r·min~(-1),在此条件下发酵24 h酶活力可达5.29 U·m L~(-1),比优化前(1.57 U·m L~(-1))提高2.37倍.  相似文献

为进一步探索高产褐藻胶裂解酶菌株,促进其工业化应用,以褐藻酸钠为唯一碳源筛选培养基,从鲍鱼内脏中筛选出一株高产褐藻胶裂解酶菌株B4,通过形态学观察和系统发育分析,鉴定为弧菌属细菌Vibrio sp.B4。通过单因素实验对B4菌株的发酵条件和发酵培养基进行优化,获得发酵培养基的最适碳源为褐藻酸钠10g/L,最适氮源为(NH4)2SO410g/L,最适发酵条件为温度30℃,接种量1%,培养基初始pH 6.0。在单因素实验的基础上,通过Plackett-Burman(PB)筛选出3个影响产酶活力的显著因素:(NH4)2SO4浓度、pH、接种量。通过响应面进一步分析优化,得到最佳的发酵培养基为褐藻酸钠10g/L,(NH_4)_2SO_4 10.91g/L,NaCl 10g/L,KH_2PO_4 1g/L,MgSO4·7H2O 0.5g/L,CaCl_2 0.2g/L,FeSO_4·7H_2O 0.02g/L,最佳发酵条件为温度30℃,接种量1.1%,起始pH 6.07。在最佳培养条件下,褐藻胶裂解酶活力可达19.09U/mL,比基础培养条件下提高了3.67倍。该菌经过产酶发酵条件的优化,酶活力得到较大幅度的提高,且其发酵产酶时间短,可为褐藻胶裂解酶的进一步研究应用提供参考。  相似文献

从淀粉加工厂附近土壤中筛选得到一株普鲁兰酶高产菌株,编号为Z-13,其初始酶活达到5.7 U/mL.通过对此菌株的16S rDNA比对,以及生理生化鉴定,鉴定此菌株为克雷伯氏菌(Klebsiella variicola),通过发酵条件优化,确定最佳发酵产酶条件为:玉米淀粉1.5%,蛋白胨2.0%,KH2PO40.05%,MgSO4·7H2O 0.01%.装液量为70 L/250 L,培养基最初pH为6.0,发酵温度30℃,摇床转速200 r/min,发酵时间48 h,优化后菌株产普鲁兰酶的酶活高达67.8 U/mL,是优化前菌株产普鲁兰酶活性的11.89倍.  相似文献

利用栀子苷培养基快速筛选β-葡萄糖苷酶产生菌,其中草酸青霉产酶水平较高.通过单因素实验、均匀设计及回归分析优化了发酵培养基配比和摇瓶产酶最佳条件:麸皮4%,牛肉膏0.2%,NaCl 0.1%,CuSO40.08%,EDTA 0.2%,KH2PO4 0.2%,初始pH 7.0,装液量20 mL/250 mL,温度30℃.发酵液上清中的酶活由最初的16.80 U/mL提高到52.18 U/mL.  相似文献

为提高几丁质酶产生菌C4的产酶效率,加强其对Bt(Bacillus thuringiensis)的增效作用,本研究通过培养基优化,筛选出了该菌发酵优化培养基,配方为:蛋白胨0.8%,酵母膏0.8% KNO3 0.8%,MgSO4 0.05%,KH2PO4 0.03%,pH 7.5. 28℃,180 r/min,培养48 h至对数期,加入1.0%的胶体几丁质,诱导C4菌产生几丁质酶.在此条件下,C4菌产酶周期由5 d缩短为4 d;几丁质酶活力由2.68 U/mL提高到5.95 U/mL.用此发酵菌液与B  相似文献

芽孢杆菌(Bacillus sp. )DPE7产嗜热蛋白酶的适宜条件,较好的碳源是0.1%葡萄糖,最佳氮源为酵母粉加硫酸铵,在pH 6~11范围内产酶均良好.在65℃,175 r/min,pH 9的产酶培养基中发酵24 h,产酶量可达23.4 U/mL.0.01% MnCl2,0.15% CaCl2或0.15%吐温存在时能刺激菌株产酶.  相似文献

研究实验室自主分离的2株纤溶酶高产菌株发酵条件,采用4因素、3水平的正交试验设计方法对液体发酵培养基的碳源、氮源、无机盐进行了优化,并对培养基初始pH、发酵时间、发酵温度对产酶量的影响进行了测定.结果表明:菌株1液体发酵合适的产酶培养基配方为大豆粉的质量分数为6%,葡萄糖2%,CaCl2 0.06%,MgSO4 0.07%;菌株2液体发酵合适的产酶培养基配方为大豆粉的质量分数为6%,玉米淀粉2%,CaCl2 0.06%,MgSO4 0.07%.培养基初始pH均为6.5时酶活性最高;发酵温度均以38℃最好;而菌株1和菌株2的适宜发酵时间分别为48~84 h和60~84 h.在优化条件下,菌株1、菌株2液体发酵最高酶活分别为306.46 IU/mL和319.76 IU/mL。  相似文献

对桔绿木霉(Trichoderma citrinoviride)XE-13产木聚糖酶发酵条件进行优化.采用单因素法,研究固体发酵条件下最适培养基组成、料水比以及培养温度、培养基初始p H、培养时间、接种量等条件对产酶的影响.确定培养基组成为:碳源(m(麸皮)∶m(玉米芯)=4 g∶6 g);氮源(NH_4)_2SO_4;碳∶氮=(m(C)∶m(N)=1 g∶0.05 g);料∶水=1 g∶1.5 mL.最适合的培养条件为:培养温度30℃、培养基初始pH值6.0、培养时间72 h、接种量1.0×10~7个/mL孢子悬液/培养基干料=0.3 mL/10 g.在此条件下,木聚糖酶活力可达391.5 U/g.  相似文献

分别以多孔陶瓷和浮石为载体吸附法固定重组大肠杆菌(Escherichia coli JM 109-pLF3)表达β-葡聚糖酶.研究了载体量、转速、温度、pH等条件对固定化细胞产酶的影响.结果表明,以多孔陶瓷和浮石为载体固定化细胞发酵可以大大提高产酶效率,二者均可有效吸附重组大肠杆菌,发酵液的酶活分别达到97 U/mL和73 U/mL,比悬浮液体发酵提高了2~3倍;载体量、转速、温度对产酶的影响较大,多孔陶瓷和浮石的最佳装载量分别为20 g/100 mL培养基和8g/100 mL培养基,最佳转速为200 r/min,最佳培养温度为37℃;发酵液pH值对细胞产酶的影响较小,pH值6.0~8.0之间发酵液的酶活力变化不大.重复批次发酵结果表明,固定化发酵具有良好的重复使用能力,在连续10批次实验中,多孔陶瓷载体和浮石载体固定化发酵的酶活力分别不低于91 U/mL和72 U/mL.  相似文献

There are numerous geometric objects stored in the spatial databases. An importance function in a spatial database is that users can browse the geometric objects as a map efficiently. Thus the spatial database should display the geometric objects users concern about swiftly onto the display window. This process includes two operations:retrieve data from database and then draw them onto screen. Accordingly, to improve the efficiency, we should try to reduce time of both retrieving object and displaying them. The former can be achieved with the aid of spatial index such as R-tree, the latter require to simplify the objects. Simplification means that objects are shown with sufficient but not with unnecessary detail which depend on the scale of browse. So the major problem is how to retrieve data at different detail level efficiently. This paper introduces the implementation of a multi-scale index in the spatial database SISP (Spatial Information Shared Platform) which is generalized from R-tree. The difference between the generalization and the R-tree lies on two facets: One is that every node and geometric object in the generalization is assigned with a importance value which denote the importance of them, and every vertex in the objects are assigned with a importance value,too. The importance value can be use to decide which data should be retrieve from disk in a query. The other difference is that geometric objects in the generalization are divided into one or more sub-blocks, and vertexes are total ordered by their importance value. With the help of the generalized R-tree, one can easily retrieve data at different detail levels.Some experiments are performed on real-life data to evaluate the performance of solutions that separately use normal spatial index and multi-scale spatial index. The discovery of the prolific Ordovician Red River reservoirs in 1995 in southeastern Saskatchewan was the catalyst for extensive exploration activity which resulted in the discovery of more than 15 new Red River pools. The best yields of Red River production to date have been from dolomite reservoirs. Understanding the processes of dolomitization is, therefore, crucial for the prediction of the connectivity, spatial distribution and heterogeneity of dolomite reservoirs.The Red River reservoirs in the Midale area consist of 3~4 thin dolomitized zones, with a total thickness of about 20 m, which occur at the top of the Yeoman Formation. Two types of replacement dolomite were recognized in the Red River reservoir: dolomitized burrow infills and dolomitized host matrix. The spatial distribution of dolomite suggests that burrowing organisms played an important role in facilitating the fluid flow in the backfilled sediments. This resulted in penecontemporaneous dolomitization of burrow infills by normal seawater. The dolomite in the host matrix is interpreted as having occurred at shallow burial by evaporitic seawater during precipitation of Lake Almar anhydrite that immediately overlies the Yeoman Formation. However, the low δ18O values of dolomited burrow infills (-5.9‰~ -7.8‰, PDB) and matrix dolomites (-6.6‰~ -8.1‰, avg. -7.4‰ PDB) compared to the estimated values for the late Ordovician marine dolomite could be attributed to modification and alteration of dolomite at higher temperatures during deeper burial, which could also be responsible for its 87Sr/86Sr ratios (0.7084~0.7088) that are higher than suggested for the late Ordovician seawaters (0.7078~0.7080). The trace amounts of saddle dolomite cement in the Red River carbonates are probably related to "cannibalization" of earlier replacement dolomite during the chemical compaction.  相似文献

Future mobile communication systems aim at providing very high data transmission rates, even in high-mobility scenarios such as high-speed wheel-track trains, maglev trains, highway vehicles, airplanes, guided missiles or spacecraft. A particularly important commercial application is the strong and increasing worldwide demand for high- speed broadband wireless communications (up to 574.8 km/ h test speeds or 380 km/h commercial speeds) in railways, providing data, voice and video services for applications such as onboard entertainment services to passengers, train control, train dispatch, train sensor status handling and sur- veillance. In such high-mobility scenarios, there are a number of communication challenges, including fast hand- over, location updating, high-speed channel modeling, estimation and equalization, anti-Doppler spreading tech- niques, fast power control, and dedicated network architec- ture. 本文叙述了对海南岛及其毗邻大陆边缘白垩纪到第四纪地层岩石进行古地磁研究的全部工作过程。通过分析岩石中剩余磁矢量的磁偏角及磁倾角的变化,提出海南岛白垩纪以来经历的构造演化模式如下:早期伴随顺时针旋转而向南迁移,后期伴随逆时针转动并向北运移。联系该地区及邻区的地质、地球物理资料,对海南岛上述的构造地体运动提出以下认识:北部湾内早期有一拉张作用,主要是该作用使湾内地壳显著伸长减薄,形成北部湾盆地。从而导致了海南岛的早期构造运动,而海南岛后期的构造运动则主要是受南海海底扩张的影响。海南地体运动规律的阐明对于了解北部湾油气盆地的形成演化有重要的理论和实际意义。  相似文献

Being the primary media of geographical information and the elementary objects manipulated, almost all of maps adopt the layer-based model to represent geographic information in the existent GIS. However, it is difficult to extend the map represented in layer-based model. Furthermore, in Web-Based GIS, It is slow to transmit the spatial data for map viewing. In this paper, for solving the questions above, we have proposed a new method for representing the spatial data. That is scale-based model. In this model we represent maps in three levels: scale-view, block, and spatial object, and organize the maps in a set of map layers, named Scale-View, which associates some given scales.Lastly, a prototype Web-Based GIS using the proposed spatial data representation is described briefly.  相似文献
