大鼠胆管阻塞性肝纤维化模型的建立   总被引:8,自引:0,他引:8  
目的 建立大鼠胆管阻塞性肝纤维化模型 ,为肝纤维化机制和治疗研究提供基础。方法 雌性Wistar大鼠进行胆管内逆行性注入N 丁基 2 氰基丙烯酸盐加胆管结扎导致胆管阻塞 ,建立胆管阻塞性肝纤维化模型。观察手术后第 2 8d和 5 6d不同时间相关指标的改变。结果 胆管阻塞性肝纤维化模型ALT、AST、HA、LN均明显升高 ;病理组织学可见弥漫增生的胆小管 ,大量的胶原沉积在胆小管周围 ,肝细胞与增生的胆小管相比明显减少。结论 胆管阻塞性肝纤维化模型造模周期短 ,自发逆转率低 ,肝纤维化形成稳定 ,是研究肝纤维化的理想模型  相似文献

近年来,国内外学者应用胆管阻塞型肝纤维化模型进行了肝纤维化可逆性及药物抗纤维化作用等的研究。现综述如下。1胆管阻塞型肝纤维化模型的建立及其形成机制1·1胆管阻塞型肝纤维化模型的建立及其特点胆管阻塞(BDL)包括胆管结扎、胆管结扎后切断和胆管粘堵(如逆行性注入组织胶)等方法。研究证实胆管粘堵的方法较其它两种方法稳定,胆管再通率低于单纯胆管结扎与胆管结扎后切断。阴宏[1,2]等总结了胆管阻塞型肝纤维化模型的特点:(1)炎症反应轻,肝细胞坏死不明显;(2)胆管阻塞术后纤维化形成快,术后7d血清肝纤维化指标(板层素、透明质酸等)升…  相似文献

目的研究汉丹肝乐抗大鼠免疫性肝纤维化的作用.方法猪血清腹腔内注射复制大鼠免疫损伤性肝纤维化模型,用汉丹肝乐治疗,研究肝脏细胞外基质的变化及中药汉丹肝乐对其的影响.结果模型组Ⅰ、Ⅲ型胶原及层粘蛋白的表达较正常组明显,汉丹肝乐治疗组Ⅰ、Ⅲ型胶原及层粘蛋白的表达明显较模型组轻,差异有显著性意义(P<0.05).结论采用猪血清复制大鼠免疫性肝纤维化模型效果较为理想,汉丹肝乐能有效防治免疫损伤性肝纤维化.  相似文献

目的:观察虎金丸的抗肝纤维化作用及对肝纤维化的免疫组化和超微结构的影响。方法:采用CCl4 后腿皮下注射致大鼠肝纤维化模型。结果:模型组大鼠病理片呈肝纤维化变,FSC增生活跃,有较大量的IV型胶原沉积。超微结构可见大量的胶原纤维生成和成纤维细胞。结论:虎金丸能明显改善上述组织形态学的变化,抑制肝纤维化的形成,改善肝细胞的超微结构。  相似文献

目的观察黄甲软肝颗粒对人血白蛋白( human serum albumin,HSA)所致的Wistar大鼠免疫性肝纤维化的保护作用。方法采用体内实验,将72只雄性大鼠随机分成阴性对照组、模型组、阳性药组、黄甲软肝高、中以及低剂量组剂量组,建立HSA致大鼠免疫性肝纤维化模型,计算肝、脾系数,测定血清中肝纤维化标志物含量、丙氨酸氨基酸转移酶( ALT) 、天门冬氨酸转移酶( AST)等相关生化指标,并检查肝脏病理形态改变。结果与模型组比较,黄甲软肝三个剂量组大鼠血清透明质酸( HA)含量显著降低;高剂量组血清层粘连蛋白( LN) 、Ⅲ型前胶原( PCⅢ )水平、ALT 和 AST 显著降低;高剂量组及中剂量组肝纤维化病变明显减轻。 结论 黄甲软肝颗粒能显著改善 HSA 所致的免疫性肝纤维化大鼠的肝纤维化程度。  相似文献

利用乙肝病毒表面抗原转基因(HBs-Tg)小鼠与γδT细胞缺陷型(TCRδ~(-/-))小鼠杂交,筛选出HBs-Tg-TCRδ~(-/-)小鼠.采用腹腔连续注射四氯化碳(CCl_4)的方式建立了小鼠肝纤维化模型,探讨了γδT细胞在HBs-Tg小鼠肝纤维化模型中的作用.结果表明:与HBsTg小鼠相比,HBs-Tg-TCRδ~(-/-)小鼠反映肝损伤和肝纤维化的相关指标ALT及α-SMA,TIMP1,COl1a1基因表达水平显著增高——肝纤维化更为严重——γδT细胞在HBs-Tg小鼠肝纤维化模型中具有保护作用.为今后γδT细胞对肝纤维化的治疗奠定了一定基础.  相似文献

目的:观察红景天苷对四氯化碳(CCl4)致大鼠肝纤维化模型血清学指标、肝组织羟脯氨酸(Hyp)和肝组织学的影响,探讨红景天苷抗肝纤维化作用。方法:实验大鼠分为5组,制备CCl4肝纤维化模型,12周后,测定血清总蛋白(TP)、白蛋白(ALB)、丙氨酸转氨酶(GPT)、天冬氨酸转氨酶(GOT)、透明质酸(HA)、层粘蛋白(LN)I、II型前胶原(PCIII)I、V型胶原(IV-C)和肝组织Hyp,并用光镜观察肝组织形态学变化。结果:模型大鼠与正常大鼠比较,血清GPT、GOT、HA、LN、PCIII和IV-C升高,血清TP、ALB和肝组织Hyp降低,呈现肝纤维化组织形态学变化;红景天苷可明显降低模型大鼠血清GPT、GOT、HA、LN、PCIII和IV-C水平(P〈0.05或P〈0.01),升高血清TP、ALB和肝组织Hyp(P〈0.05或P〈0.01),实验性肝纤维化组织形态学得到改善。结论:红景天苷具有抗实验性肝纤维化作用。  相似文献

水稻黄酮对免疫性肝纤维化的保护作用   总被引:2,自引:0,他引:2  
目的:观察水稻黄酮对免疫性肝纤维化大鼠模型的保护作用,并从自由基的产生及清除方面探讨其机理.方法:腹腔注射异源血清诱导的大鼠肝纤维化模型,以γ-干扰素作对照,测定相应肝功和肝纤维化指标,及肝组织中GSH-PX、MDA、SOD活力.结果:γ-干扰素和水稻黄酮可显著改善肝功与肝纤维化,降低MDA含量,提高SOD、GSH-PX活力.结论:γ-干扰素和水稻黄酮,对肝纤维化有明显的抑制作用;而水稻黄酮还可显著改善肝功能,提高肝组织中自由基清除系统的功能,加强肝组织清除自由基及MDA,减轻自由基对肝组织的损害.  相似文献

在中日友好医院临床及病理确诊的45例慢性乙型肝炎病例中,随机抽取9例通过血清肝纤维化指标进行基于等价矩阵的模糊聚类,判定肝纤维化程度。并按2000年全国传染病防治方案制定的慢性乙型肝炎病理诊断标准进行纤维化分期。模糊聚类结果与肝穿病理纤维化程度基本一致。依此建立慢性乙型肝炎纤维化预报模型,采用3例待预报样本进行验证,证实本模型利用无损伤的方法对判定肝纤维化程度具有一定的实用性。  相似文献

The discovery of the prolific Ordovician Red River reservoirs in 1995 in southeastern Saskatchewan was the catalyst for extensive exploration activity which resulted in the discovery of more than 15 new Red River pools. The best yields of Red River production to date have been from dolomite reservoirs. Understanding the processes of dolomitization is, therefore, crucial for the prediction of the connectivity, spatial distribution and heterogeneity of dolomite reservoirs.The Red River reservoirs in the Midale area consist of 3~4 thin dolomitized zones, with a total thickness of about 20 m, which occur at the top of the Yeoman Formation. Two types of replacement dolomite were recognized in the Red River reservoir: dolomitized burrow infills and dolomitized host matrix. The spatial distribution of dolomite suggests that burrowing organisms played an important role in facilitating the fluid flow in the backfilled sediments. This resulted in penecontemporaneous dolomitization of burrow infills by normal seawater. The dolomite in the host matrix is interpreted as having occurred at shallow burial by evaporitic seawater during precipitation of Lake Almar anhydrite that immediately overlies the Yeoman Formation. However, the low δ18O values of dolomited burrow infills (-5.9‰~ -7.8‰, PDB) and matrix dolomites (-6.6‰~ -8.1‰, avg. -7.4‰ PDB) compared to the estimated values for the late Ordovician marine dolomite could be attributed to modification and alteration of dolomite at higher temperatures during deeper burial, which could also be responsible for its 87Sr/86Sr ratios (0.7084~0.7088) that are higher than suggested for the late Ordovician seawaters (0.7078~0.7080). The trace amounts of saddle dolomite cement in the Red River carbonates are probably related to "cannibalization" of earlier replacement dolomite during the chemical compaction.  相似文献

There are numerous geometric objects stored in the spatial databases. An importance function in a spatial database is that users can browse the geometric objects as a map efficiently. Thus the spatial database should display the geometric objects users concern about swiftly onto the display window. This process includes two operations:retrieve data from database and then draw them onto screen. Accordingly, to improve the efficiency, we should try to reduce time of both retrieving object and displaying them. The former can be achieved with the aid of spatial index such as R-tree, the latter require to simplify the objects. Simplification means that objects are shown with sufficient but not with unnecessary detail which depend on the scale of browse. So the major problem is how to retrieve data at different detail level efficiently. This paper introduces the implementation of a multi-scale index in the spatial database SISP (Spatial Information Shared Platform) which is generalized from R-tree. The difference between the generalization and the R-tree lies on two facets: One is that every node and geometric object in the generalization is assigned with a importance value which denote the importance of them, and every vertex in the objects are assigned with a importance value,too. The importance value can be use to decide which data should be retrieve from disk in a query. The other difference is that geometric objects in the generalization are divided into one or more sub-blocks, and vertexes are total ordered by their importance value. With the help of the generalized R-tree, one can easily retrieve data at different detail levels.Some experiments are performed on real-life data to evaluate the performance of solutions that separately use normal spatial index and multi-scale spatial index. 本文叙述了对海南岛及其毗邻大陆边缘白垩纪到第四纪地层岩石进行古地磁研究的全部工作过程。通过分析岩石中剩余磁矢量的磁偏角及磁倾角的变化,提出海南岛白垩纪以来经历的构造演化模式如下:早期伴随顺时针旋转而向南迁移,后期伴随逆时针转动并向北运移。联系该地区及邻区的地质、地球物理资料,对海南岛上述的构造地体运动提出以下认识:北部湾内早期有一拉张作用,主要是该作用使湾内地壳显著伸长减薄,形成北部湾盆地。从而导致了海南岛的早期构造运动,而海南岛后期的构造运动则主要是受南海海底扩张的影响。海南地体运动规律的阐明对于了解北部湾油气盆地的形成演化有重要的理论和实际意义。  相似文献

The explosive growth of the Internet and database applications has driven database to be more scalable and available, and able to support on-line scaling without interrupting service. To support more client's queries without downtime and degrading the response time, more nodes have to be scaled up while the database is running. This paper presents the overview of scalable and available database that satisfies the above characteristics. And we propose a novel on-line scaling method. Our method improves the existing on-line scaling method for fast response time and higher throughputs. Our proposed method reduces unnecessary network use, i.e. , we decrease the number of data copy by reusing the backup data. Also, our on-line scaling operation can be processed parallel by selecting adequate nodes as new node. Our performance study shows that our method results in significant reduction in data copy time.  相似文献

R-Tree is a good structure for spatial searching. But in this indexing structure,either the sequence of nodes in the same level or sequence of traveling these nodes when queries are made is random. Since the possibility that the object appears in different MBR which have the same parents node is different, if we make the subnode who has the most possibility be traveled first, the time cost will be decreased in most of the cases. In some case, the possibility of a point belong to a rectangle will shows direct proportion with the size of the rectangle. But this conclusion is based on an assumption that the objects are symmetrically distributing in the area and this assumption is not always coming into existence. Now we found a more direct parameter to scale the possibility and made a little change on the structure of R-tree, to increase the possibility of founding the satisfying answer in the front sub trees. The geographic information service is enabled by the advancements in general Web service technology and the focused efforts of the OGC in defining XML-based Web GIS service. Based on these models, this paper addresses the issue of services chaining,the process of combining or pipelining results from several interoperable GIS Web Services to create a customized solution. This paper presents a mediated chaining architecture in which a specific service takes responsibility for performing the process that describes a service chain. We designed the Spatial Information Process Language (SIPL) for dynamic modeling and describing the service chain, also a prototype of the Spatial Information Process Execution Engine (SIPEE) is implemented for executing processes written in SIPL. Discussion of measures to improve the functionality and performance of such system will be included.  相似文献
