为了研究鸡汤乳化液中多肽序列组成与乳化性能之间的关系,通过炖煮鸡骨架3h获得鸡白汤,离心得到稳定的乳化液层,经过除脂、微滤处理,得到乳化多肽溶液。分析发现多肽的平均长度为13个氨基酸残基,临界胶束浓度值为4mg/mL。乳化多肽溶液经3kDa超滤离心管过滤后,进行高效液相色谱-质谱法序列分析,获得1646条多肽序列。统计发现乳化多肽平均疏水度为4791.70kJ/mol,亲水性氨基酸和疏水性氨基酸组成比例接近,疏水性氨基酸或亲水性氨基酸占比很高的多肽数量较少,强亲水性氨基酸和强疏水性氨基酸在多肽序列中的占比不高,多肽两端亲水性氨基酸较多。该研究旨在为新型多功能多肽表面活性剂的开发提供理论依据。  相似文献

Rice genomic DNA was surveyed by polymerase chain reaction (PCR) to detect homeobox sequences. The PCR product (183 bp) was cloned and sequenced. The nucleotide sequence and the deduced amino acid sequence of a homeobox, which was isolated in this study, and designated OSIHI1, were obtained. Comparison of the encoded polypeptide sequence with other homeodomains reveals that OSIHI1 has 85% and 87% identity to that of Antp, and quail Quox1, respectively at the protein level. An alignment of the OSIHI1 amino acid sequence with homeodoma in sequences from varlous other eukaryotes shows that OSIHI1 homeodomain contains identical residues in the eight positions most conserved among homeodomains, and also contains the four invariant residues present in the putative recognition helix (helix3) Supported by the National Natural Science Foundation of China Yi Qingming: born in Apr. 1938. Isolated polypeptide chains of the T-cell antigen receptor complex are degraded or retained in the endoplasmic reticulum (ER). Assembly of the multisubunit complex allows the individual chains to escape retention in the ER and to be expressed on the cell surface. We engineered a series of deletions in the CD3 epsilon subunit of the human T-cell receptor in order to find the sequences responsible for its retention in the ER. Deletion of amino acids 171 to 180 in the cytosolic tail resulted in the cell-surface expression of the isolated chain. This sequence also promotes retention when it is appended to CD4, a plasma membrane protein. Mutagenesis of the 10-amino-acid CD3 epsilon sequence established that the tyrosine and serine residues are important for ER retention. This and other ER retention signals must be hidden when a complete T-cell receptor complex is assembled in order to allow its expression on the cell surface.  相似文献

Epidermal growth factor (EGF) has a profound effect on the differentiation of specific cells in vivo, and has been shown to be a potent mitogenic factor for a variety of cultured cells, of both ectodermal and mesodermal origin (see ref. 1 for review). This 53-amino acid polypeptide of known sequence contains six cysteine residues, which are thought to form three intrachain disulphide bonds. Urogastrone, a polypeptide bearing anti-gastric secretory activity isolated from human urine, which is presumably synthesized in submandibular and Brunner's glands, shares extensive sequence homology (70%) with EGF and may represent the human EGF equivalent. Here we present the sequence of a mouse EGF cDNA clone, which suggests that EGF is synthesized as a large protein precursor of 1,168 amino acids. Our data indicate that the discrepancy between EGF levels in male and female mouse submaxillary glands (MSGs) is due to different EGF mRNA levels in these tissues, and suggest that precursor EGF processing may differ from that described previously for other polypeptide hormones.  相似文献

用硫酸胺盐析、Sephadex G-100柱层析等方法,从人体精子中提纯得到电泳单点纯的顶体蛋白酶。用SDS-聚丙烯酰铵凝胶电泳法测定了人体精子顶体蛋白酶的相对分子质量(约为10470)。用N-末端分析和氨基酸组成分析等方法研究了这种酶的一些化学特征。实验结果表明,人体精子顶体蛋白酶由85个氨基酸残基组成,它是以亮氨酸为N-末端的单一多肽链。  相似文献

Somatomedins (SM) or insulin-like growth factors (IGF) constitute a heterogeneous group of peptides with important growth-promoting effects in vitro as well as in vivo. Amino acid sequences have been determined for only two of them, IGF-I and IGF-II, which are highly homologous. IGF-I, which is identical with SM-C, is composed of 70 amino acid residues and IGF-II contains 73 amino acids and may be identical with SM-A. Other peptides with different charge properties but with similar SM-like or insulin-like behaviour in biological and receptor assays, have been described but have not yet been fully characterized. The liver is known to be a major site of production of these peptides, but many other tissues--especially in the fetus--may synthesize them as well. We report here the nucleotide sequence of a human liver cDNA encoding the complete amino acid sequence of IGF-I. The IGF-I coding region is flanked by sequences encoding an amino-terminal peptide of at least 25 amino acid residues and a carboxyl-terminal peptide of 35 amino acids. This provides evidence that IGF-I is synthesized as a precursor protein and that formation of IGF-I from this precursor requires proteolytic processing at both ends.  相似文献

对云芝等7种真菌的菌丝体和发酵液分别测定17种氨基酸的含量,测定结果表明这些菌丝体和发酵液中含有丰富的人体8种必需氨基酸,支链氨基酸及精氨酸,而芳香族氨基酸和含硫氨基酸量却很低。7种真菌的菌丝体和它们的子实体,三种食用蛋白(α—酪蛋白,卵白蛋白,大豆球蛋白)及4种粮食产品(大米,小麦,小麦精粉,玉米)的氨基酸进行了比较,其结果是菌丝体的必需氨基氨含量,支链氨基酸及精氨酸含量一般都高于子实体。菌丝体必需氨基酸相对百分含量高于或接近于三种食用蛋白,菌丝体支链氨基酸和精氨酸相对百分含量均高于三种食用蛋白,菌丝体芳香族氨基酸相对百分含量全低于三种食用蛋白。令人注目的是7种真菌菌丝的支链氨基酸与芳香族氨基酸的比值全大于3种食用蛋白和4种粮食产品。这些数据为发酵培养开发利用真菌资源提供了新资料。  相似文献

In this study, an amphioxus cDNA, AmphiSDHD, encoding the cytochrome b small subunit in mitochondrial succinate-ubiquinone oxidoreductase, was isolated from the gut cDNA library of amphioxus Branchiostoma belcheri tsingtauense. It is 1429 bp in length, with an open reading frame of 465 bp coding for a protein of 154 amino acids. The deduced protein contains a mitochondrial targeting presequence of 65 amino acids rich in basic residues like arginine and hydroxy residues such as serine and threonine. Alignment of the amino acid sequences of AmphiSDHD and other eukaryotic SDHD proteins showed that AmphiSDHD has three transmembrane segments, and includes two histidine residues in the second transmembrane segment that are the putative binding sites for the heme b molecule. The phylogenetic tree constructed suggests that AmphiSDHD appears more closely related to vertebrate SDHD proteins than invertebrate ones. Northern blotting demonstrated that AmphiSDHD is ubiquitously expressed in amphioxus, being in line with the fact that SDHD is a house-keeping protein.  相似文献

A gene encoding a cysteine proteinase was isolated from senescent leave of cotton (Gossypium hirsutum) cv liaomian No. 9 by utilizing rapid amplification of cDNA end spolymerase chain reaction (RACE-PCR), and a set of consensus oligonucleotide primers was designed to anneal the conserved sequences of plant cysteine protease genes. The cDNA, which designated Ghcysp gene, contained 1368 bp terminating in a poly(A)^ trail, and included a putative 5‘(98 bp) and a 3‘(235 bp) non-coding region. The opening reading frame (ORF) encodes polypeptide 344 amino acids with the predicted molecular mass of 37.88 kD and theoretical pl of 4.80. A comparison of the deduced amino acid sequence with the sequence in the GenBank database has shown considerable sequence similarity to a novel family of plant cysteine proteases. This putative cotton Ghcysp protein shows from 67% to 82% identity to the other plants. All of them share catalytic triad of residues, which are highly conserved in three regions. Hydropaths analysis of the amino acid sequence shows that the Ghcysp is a potential membrane protein and localizes to the vacuole, which has a transmembrane helix between resides 7-25. A characteristic feature of Ghcysp is the presence of a putative vacuole-targeting signal peptide of 19-amino acid residues at the N-terminal region. The expression of Ghcysp gene was determined using northern blot analysis. The Ghcysp mRNA levels are high in development senescent leaf but below the limit of detection in senescent root, hypocotyl, faded flower, 6 d post anthesis ovule, and young leaf.  相似文献

Epidermal growth factor (EGF) and transforming growth factor type I (TGF) are polypeptides of 53 and 50 amino acid residues, respectively. Both bind to EGF receptor, a 1,200-residue transmembranous glycoprotein, leading to phosphorylation of the receptor, enhancement of its tyrosine-specific kinase activity and ultimately to stimulation of cell growth. We report here that a 140-residue polypeptide encoded by one of the early genes of vaccinia virus (VV) is related closely to EGF and TGF. The presence of putative signal and transmembranous sequences further suggests that the viral protein might be an integral membrane protein, but that, as in the case of EGF itself, the membrane-associated form may be the precursor of a soluble growth factor. Production of EGF-like growth factors by virally infected cells could account for the proliferative diseases associated with members of the poxvirus family such as Shope fibroma virus, Yaba tumour virus, and molluscum contagiosum virus (MCV).  相似文献

分析了4种植物蛋白的氨基酸组成特征和营养学特性。玉米蛋白粉、大豆粉、棉籽蛋白粉、花生饼粉蛋白质质量分数分别为61.60%,38.18%,51.49%,43.15%。大豆粉氨基酸组成比较均衡;棉籽蛋白粉、花生饼粉中多种必需氨基酸组成与大豆蛋白接近,但蛋氨酸含量较低;玉米蛋白粉富含亮氨酸、异亮氨酸、缬氨酸等支链氨基酸,但其中赖氨酸、色氨酸含量明显低于其他3种植物蛋白。建议根据蛋白资源的氨基酸特色组成,合理搭配互补,以提高谷物蛋白质营养价值。  相似文献

The polypeptide chain of the acid protease penicillo pepsin folds via an 18-stranded mixed beta-sheet into two distinct lobes separated by a 30-A long groove which is the extended substrate binding site. The catalytic residues Asp-32 and Asp-215 are located in this groove and their carboxyl groups are in intimate contact. Positively charged amino acids have been shown to be important elements in targeting-peptides that direct proteins into mitochondria, nuclei, and the secretory pathways of both prokaryotic and eukaryotic cells. The 'positive-inside' rule, which observes that regions of polytopic (multi-spanning) membrane proteins facing the cytoplasm are generally enriched in arginyl and lysyl residues whereas translocated regions are largely devoid of these residues, implies that the distribution of positively charged amino acids may also be a major determinant of the transmembrane topology of integral membrane proteins. If this is indeed the case, it should be possible to predictably alter the topology of a polytopic protein by site-directed insertions and/or deletions of positively charged residues in critical locations. I now describe a derivative of Escherichia coli leader peptidase, a polytopic inner-membrane protein, that switches from sec-gene-dependent membrane insertion with a Nout-Cout transmembrane topology to sec-gene-independent insertion with a Nin-Cin topology in response to the addition of four positively charged lysines to its N terminus.  相似文献

Knowledge of the tertiary structure of the proteinase from human immunodeficiency virus HIV-1 is important to the design of inhibitors that might possess antiviral activity and thus be useful in the treatment of AIDS. The conserved Asp-Thr/Ser-Gly sequence in retroviral proteinases suggests that they exist as dimers similar to the ancestor proposed for the pepsins. Although this has been confirmed by X-ray analyses of Rous sarcoma virus and HIV-1 proteinases, these structures have overall folds that are similar to each other only where they are also similar to the pepsins. We now report a further X-ray analysis of a recombinant HIV-1 proteinase at 2.7 A resolution. The polypeptide chain adopts a fold in which the N- and C-terminal strands are organized together in a four-stranded beta-sheet. A helix precedes the single C-terminal strand, as in the Rous sarcoma virus proteinase and also in a synthetic HIV-1 proteinase, in which the cysteines have been replaced by alpha-aminobuytric acid. The structure reported here provides an explanation for the amino acid invariance amongst retroviral proteinases, but differs from that reported earlier in some residues that are candidates for substrate interactions at P3, and in the mode of intramolecular cleavage during processing of the polyprotein.  相似文献
