用透射电镜、紫外-可见光谱研究银纳米粒子与血清白蛋白的相互作用及其相关效应,透射电镜观测到血清白蛋白对银纳米粒子的包覆,表面等离子共振吸收峰的研究表明,静电力和亲水力是血清白蛋白与银纳米粒子之间的主要作用力;血清白蛋白对锒纳米粒子最低包覆比在10^-5~10^-6之间。  相似文献

用荧光光谱、紫外吸收光谱及圆二色光谱技术研究Fe3O4磁性纳米粒子与透明质酸酶(Hyaluronidase)的相互作用,结果表明Fe3O4磁性纳米粒子对透明质酸酶的荧光猝灭机理为静态猝灭;二者结合常数K在104数量级,说明Fe3O4磁性纳米粒子与透明质酸酶有较强的结合;二者相互作用过程中各热力学常数的变化表明Fe3O4与透明质酸酶的作用过程是一个自发过程,而且他们之间的结合力为疏水作用力;紫外吸收光谱研究发现Fe3O4能改变透明质酸酶的空间结构;圆二色谱数据表明Fe3O4磁性纳米粒子能够引起透明质酸酶的二级结构发生变化。  相似文献

采用荧光光谱、紫外光谱(UV-vis)、圆二色谱(CD)、透射电镜(TEM)、原子力显微镜(AFM)、琼脂糖凝胶电泳等技术研究Cd S-NH2-EcoRI复合物与DNA的相互作用.研究发现:Cd S-NH2纳米粒子与p BR322DNA结合后会延迟EcoRI的酶切反应.DNA的曲率和纳米粒子的粒径都是影响结合作用的因素,曲率较大的环状DNA比线性DNA能更好地与纳米粒子结合,小粒径的Cd S-NH2纳米粒子则更易结合到DNA上.并研究了DNA与Cd S-NH2纳米粒子之间的作用机理.  相似文献

在体外模拟人体生理环境,采用光谱法并结合分子对接技术,研究了安普那韦与人血清白蛋白(HSA)之间的相互作用.荧光光谱表明:安普那韦可以增强HSA的荧光强度.通过对结合常数和热力学常数的计算可知,安普那韦和HSA之间的作用力主要是疏水和静电作用力;紫外-可见光谱显示:安普那韦能够增加HSA的吸收峰强度,表明两者之间发生了相互作用.三维荧光光谱也显示:加入安普那韦后,HSA的斯托克斯位移从62 nm增加到了86 nm,说明安普那韦可以使HSA的微环境极性增加,疏水性减弱;从圆二色光谱的研究结果可知:随着安普那韦的加入,HSA的α-螺旋结构含量从18.38%增加到了63.62%,说明安普那韦的存在改变了HSA的二级结构.分子对接研究结果表明:安普那韦与HSA的作用位点位于HSA的ⅡA结构域,而同步荧光光谱的分析进一步证明了这一点.研究结果为进一步研究安普那韦在体内与血清白蛋白的相互作用提供了一定的理论参考.  相似文献

在生理条件(pH7.43)下,采用时间扫描圆二色光谱(CD)研究了Ni2 诱导人血清白蛋白(HSA)或牛血清白蛋白(BSA)产生的构象变化.拟合计算远紫外CD光谱.结果表明,血清白蛋白在结合Ni2 过程中产生了鲜见的N→R态和R→T态两步构象变化.在结合的最初几分钟,血清白蛋白中的部分α-螺旋结构分别转变为了β-折叠、β-转角和无规卷曲,其含量从66%(N态)降低到约64%(R态).随后发生了相反的变化,达到结合平衡时,α-螺旋结构含量上升到68.7%(T态).近紫外CD光谱结果表明,血清白蛋白在结合Ni2 的过程中,二硫键及芳香氨基酸残基附近的微环境发生了缓慢扰动.CD光谱观察到该构象变化持续约3 h才达到平衡.对Ni2 诱导血清白蛋白产生两步构象变化的生理意义做了讨论,推测该构象变化能暴露结合位点,提供合适的立体配位空间.有助于血清白蛋白结合Ni2 .  相似文献

利用环糊精(CD)在碱性溶液中原位还原硝酸银制得环糊精包覆的银纳米粒子,并通过透射电镜、红外光谱和紫外分光光度计对其进行表征。实验结果表明,在适当的pH和环糊精浓度下制备得到的银纳米粒子的粒径很均一,但是碱性过强(pH12)或环糊精浓度太低(cCD0.025mmol/L)会导致银纳米粒子团聚。在对硝基苯酚的还原实验中发现环糊精包覆的银纳米粒子具有很好的催化活性,而且β-环糊精和γ-环糊精包覆的银纳米粒子的催化活性高于α-环糊精包覆的银纳米粒子。  相似文献

采用柠檬酸钠还原H2PtCl6制备得到浅黄棕色负电铂纳米颗粒.研究发现,该纳米颗粒使α,β,γ,δ-四(N-甲基-3-吡啶基)卟啉(TMPyP-3)发生J-型聚集,使其特征的Soret带和Q带吸收降低,荧光猝灭、散射信号增强,圆二色光谱及暗场成像发生变化.当铂纳米粒子浓度发生变化时,TMPyP-3聚集程度不同,其光学信号的变化强度也有差异,为进一步阐明卟啉类物质聚集体形成的过程及形态奠定了基础.  相似文献

用荧光光谱、紫外光谱和圆二色谱法研究了抗凝血药物华发灵钠与人血清白蛋白(HSA)的相互作用.初步确定华法灵钠对人血清白蛋白的荧光产生猝灭现象,猝灭方式为静态,并计算了二者形成化合物的结合常数K和结合位点数n.根据不同温度下的热力学函数,确定了华发灵钠与人血清白蛋白的相互作用力类型为氢键和范德华力.发现华法灵钠的存在明显改变人血清白蛋白的构象,并讨论了华发灵钠使人血清白蛋白构象发生变化的可能原因.  相似文献

以α-淀粉酶盐酸胍变性作用为对照,利用紫外差光谱、荧光光谱和圆二色谱法研究了α-淀粉酶、脱钙α-淀粉酶和牛血清白蛋白碳酸胍及碳酸钠的变性作用。光谱数据所反映的酶分子构象变化和酶活性的变化表明,碳酸胍阴离子CO_3~(2-)与酶蛋白间存在着复杂的作用,可能是碳酸胍强变性作用的重要原因之一。  相似文献

There are numerous geometric objects stored in the spatial databases. An importance function in a spatial database is that users can browse the geometric objects as a map efficiently. Thus the spatial database should display the geometric objects users concern about swiftly onto the display window. This process includes two operations:retrieve data from database and then draw them onto screen. Accordingly, to improve the efficiency, we should try to reduce time of both retrieving object and displaying them. The former can be achieved with the aid of spatial index such as R-tree, the latter require to simplify the objects. Simplification means that objects are shown with sufficient but not with unnecessary detail which depend on the scale of browse. So the major problem is how to retrieve data at different detail level efficiently. This paper introduces the implementation of a multi-scale index in the spatial database SISP (Spatial Information Shared Platform) which is generalized from R-tree. The difference between the generalization and the R-tree lies on two facets: One is that every node and geometric object in the generalization is assigned with a importance value which denote the importance of them, and every vertex in the objects are assigned with a importance value,too. The importance value can be use to decide which data should be retrieve from disk in a query. The other difference is that geometric objects in the generalization are divided into one or more sub-blocks, and vertexes are total ordered by their importance value. With the help of the generalized R-tree, one can easily retrieve data at different detail levels.Some experiments are performed on real-life data to evaluate the performance of solutions that separately use normal spatial index and multi-scale spatial index. The results show that the solution using multi-scale index in SISP is satisfying.  相似文献

The discovery of the prolific Ordovician Red River reservoirs in 1995 in southeastern Saskatchewan was the catalyst for extensive exploration activity which resulted in the discovery of more than 15 new Red River pools. The best yields of Red River production to date have been from dolomite reservoirs. Understanding the processes of dolomitization is, therefore, crucial for the prediction of the connectivity, spatial distribution and heterogeneity of dolomite reservoirs.The Red River reservoirs in the Midale area consist of 3~4 thin dolomitized zones, with a total thickness of about 20 m, which occur at the top of the Yeoman Formation. Two types of replacement dolomite were recognized in the Red River reservoir: dolomitized burrow infills and dolomitized host matrix. The spatial distribution of dolomite suggests that burrowing organisms played an important role in facilitating the fluid flow in the backfilled sediments. This resulted in penecontemporaneous dolomitization of burrow infills by normal seawater. The dolomite in the host matrix is interpreted as having occurred at shallow burial by evaporitic seawater during precipitation of Lake Almar anhydrite that immediately overlies the Yeoman Formation. However, the low δ18O values of dolomited burrow infills (-5.9‰~ -7.8‰, PDB) and matrix dolomites (-6.6‰~ -8.1‰, avg. -7.4‰ PDB) compared to the estimated values for the late Ordovician marine dolomite could be attributed to modification and alteration of dolomite at higher temperatures during deeper burial, which could also be responsible for its 87Sr/86Sr ratios (0.7084~0.7088) that are higher than suggested for the late Ordovician seawaters (0.7078~0.7080). The trace amounts of saddle dolomite cement in the Red River carbonates are probably related to "cannibalization" of earlier replacement dolomite during the chemical compaction.  相似文献

Future mobile communication systems aim at providing very high data transmission rates, even in high-mobility scenarios such as high-speed wheel-track trains, maglev trains, highway vehicles, airplanes, guided missiles or spacecraft. A particularly important commercial application is the strong and increasing worldwide demand for high- speed broadband wireless communications (up to 574.8 km/ h test speeds or 380 km/h commercial speeds) in railways, providing data, voice and video services for applications such as onboard entertainment services to passengers, train control, train dispatch, train sensor status handling and sur- veillance. In such high-mobility scenarios, there are a number of communication challenges, including fast hand- over, location updating, high-speed channel modeling, estimation and equalization, anti-Doppler spreading tech- niques, fast power control, and dedicated network architec- ture. 本文叙述了对海南岛及其毗邻大陆边缘白垩纪到第四纪地层岩石进行古地磁研究的全部工作过程。通过分析岩石中剩余磁矢量的磁偏角及磁倾角的变化,提出海南岛白垩纪以来经历的构造演化模式如下:早期伴随顺时针旋转而向南迁移,后期伴随逆时针转动并向北运移。联系该地区及邻区的地质、地球物理资料,对海南岛上述的构造地体运动提出以下认识:北部湾内早期有一拉张作用,主要是该作用使湾内地壳显著伸长减薄,形成北部湾盆地。从而导致了海南岛的早期构造运动,而海南岛后期的构造运动则主要是受南海海底扩张的影响。海南地体运动规律的阐明对于了解北部湾油气盆地的形成演化有重要的理论和实际意义。  相似文献

Being the primary media of geographical information and the elementary objects manipulated, almost all of maps adopt the layer-based model to represent geographic information in the existent GIS. However, it is difficult to extend the map represented in layer-based model. Furthermore, in Web-Based GIS, It is slow to transmit the spatial data for map viewing. In this paper, for solving the questions above, we have proposed a new method for representing the spatial data. That is scale-based model. In this model we represent maps in three levels: scale-view, block, and spatial object, and organize the maps in a set of map layers, named Scale-View, which associates some given scales.Lastly, a prototype Web-Based GIS using the proposed spatial data representation is described briefly.  相似文献
