利用Opton显微注射系统(Microinjectionsystem)将不同分子量的荧光分子探针分别注射到不同生育期的紫竹梅雄蕊花丝毛细胞内,在OlympusBH—2型荧光显微镜下观察探针在细胞内的转移情况,取得以下结果,花蕾和开放花的雄蕊花丝毛细胞的胞间连丝允许LRB(559Da),FITC-(Ala)6(870.3Da)通过,而FITC-Leu-Arg-Arg-Ala-Ser-Val-Ala(1161.3Da)难以通过.花蕾和开放花的雄蕊花丝毛细胞间的胞间连丝的通透性差异不大.且允许通过的分子探针不超过1161.3Da.在花蕾和开放花的雄蕊花丝毛细胞间不能通过的分子探针FITC-Leu-Arg-Arg-Ala-Ser-Val-Ala(1161.3Da),FITC-InsulinAChain(2921.0Da)在衰老花雄蕊花丝毛细胞间可以转移,但分子量为4400.0De和9400.0Da的FITC-Dertran探针却难以转移.胞间连丝的通透性能,随着植物的生长发育发生相应的变化.  相似文献

首次从木通科猫儿屎(DecaisneafargesiiFranch)中分离鉴定了11种皂甙化合物.经光谱分析及化学方法鉴定,其中5种化合物为新化合物,分别命名为:DecaisosideA、B、C、D、G;另5种化合物为SaponinPG、DipsacosideB、Kalopanax-saponinB、Saponin11、HederasaponinB和saponinPJ3.猫儿屎对小鼠移植性肿瘤S180(小白鼠肉瘤)、Hepa(肝癌)、Ec(艾氏病)的平均抑制率分别为47.80%、41.48%、44.71%,具有一定的抗肿瘤活性.  相似文献

参照苜宿银纹夜蛾核型多角体病毒(AcMNPV)衣壳蛋白基因vp39的序列设计引物,采用PCR技术扩增了甜菜夜蛾核型多角体病毒的约1.3kb片段,通过将片段亚克隆至原核表达载体pET-28构建出重组表达质粒pET-Se39,以pET-Se39转化E.coli BL21。经IPTG诱导后,SeMNPVvp39基因高效表达,SDS-PAGE分析显示表达产物的分子量为39KD,并且表达量在IPTG诱导4h达到最高水平。/  相似文献

Two large molecular weight fucoidan fractions F-A and F-B were obtained by water extraction and anion-exchange chromatography and then L-A and L-B with low molecular weight and uronic acid content were prepared by radical process degradation of F-A and F-B. The antioxidant activities of the fucoidan fractions on two oxygen free radicals and Cu2 -induced low-density lipoprotein (LDL) oxidation were studied. The results show that all the fucoidan fractions had excellent scavenging capacities on superoxide radical and hypochlorous acid, except the highly sulfated fraction L-B. In LDL oxidation system, low molecular weight fractions L-A and L-B exhibited great inhibitory effects on LDL oxidation induced by Cu2 , however F-A and F-B had little inhibitory effects in this system due to their large molecular weights. The present results suggest that fucoidan from Laminaria japonica is an excellent natural antioxidant and has great potential for preventing the free radical-mediated diseases.  相似文献

以丙烯酸钠和丙烯酰胺为单体,采用反相悬浮聚合法制备了超高分子量的聚丙烯酸钠(NaPA).研究了引发剂浓度、抗交联剂及其他助剂对合成产物聚丙烯酸钠性能的影响.结果表明,(NH4)2S2O8的最佳用量是0.15%(质量分数);随着CO(NH2)2用量的增加分子量提高明显;在聚合体系中加入甲基丙烯酸N,N-二甲氨基乙酯(DMAEMA)可提高分子量但用量应控制在9.4×10-4%~15.6×10-4%之间.同时用抗交联剂防止交联反应,结合使用醋酸钠和异丙醇这两种分子量调节剂不仅能提高分子量而且溶解性也得到改善.最终得到了分子量高达3.0×107的产物,其分子量和溶解性能较前人研究成果有明显提高.  相似文献

采用恒速型毛细管流变仪研究分子量双峰分布聚乙烯PE-SP2520的高速挤出性能.结果表明:因分子量分布的特殊性,PE-SP2520的结晶度和熔峰温度均低于对比样品LLDPE-7047.高速挤出时PE-SP2520剪切黏度较高,非牛顿流动性较弱,入口压力降和熔体强度较高.与对比样品比较,PE-SP2520挤出稳定性好:挤出压力稳定,流动曲线连续,挤出畸变主要为鲨鱼皮畸变.流动稳定性提高的重要原因在于分子量双峰分布使分子链缠结能力下降,熔体/管壁边界应力集中效应弱.  相似文献

Fruit specific promoter (2A12) from Lycopersicom esculentum and cDNA of isopentenyl-transferase (ipt) from Ti plasmid of Agrobacterium tumerfaciens C58 were cloned by PCR procedure respectively. Two plant expression vectors with 2A12/gus or 2A12/ipt were respectively constructed. These two chimeric genes were transferred into tomato by Agrobacterium mediated procedure. The results of Southern hybridization showed that the fusion genes had been integrated into tomatoes. The result of gus histochemical staining showed that 2A12 had high fruit specific expressive capability in transgenic tomato. The ipt expression resulted in accumulation of high level of cytokinins (CTKs) in fruit lead to developmental changes in fruits and seeds. The fruit of ipt transformed tomato had the hyperplastic placenta with very few seeds or even seedless. The shelf life of transgenic fruits elongated for 1–2 weeks. The ratio of fruit set, the dry weight of fruit and the crude protein content in fruit were increased, while the soluble sugar of fruits decreased.  相似文献

以银杏叶和绞股蓝为研究对象,利用超滤法对所提取物分子量大小可控的特点,从单味中药提取和双中药提取两个方面考察了银杏叶、绞股蓝两味中药中黄酮的提取效果,并优化超滤提取条件.结果表明:单味银杏叶提取物黄酮含量为4.83%,单味绞股蓝提取物黄酮含量为3.58%,银杏叶和绞股蓝双中药提取物中黄酮含量为5.76%,双中药提取物中黄酮含量明显高于单中药提取物中黄酮含量.本论文的研究为制备高含量黄酮提取物以及制备药效更好的抗心血管疾病中药提供理论基础.  相似文献

用数量遗传学的原理和方法,对20个常用自选和引入的玉米自交系进行数量性状遗传参数的田间试验研究。结果表明:穗粗、行粒数、百粒重与单株产量的遗传相关达到正向极显著吐丝期、株高、穗行数、单株叶片数、单株产量的传力较高,受环境影响小,宜早代选择;单株产量的遗传进度较其余性状高,获得的遗传增量大,直接选择效果较为理想;穗粗、行粒数、百粒重以及它们所构成的选择指数对单株产量的相关遗传进度都较高,通过这3个性  相似文献

分别采用高效凝胶渗透色谱(HLGPC)法和多角度激光光散射一凝胶渗透色谱法(MALLS/GPC)法测定了分子量为10万以上氯化聚丙烯(CPP)的分子量及其分布,并据此对两种测定高聚物分子量的方法进行了初步对比分析.结果表明,两种测定方法得到的高聚物分子量相差小于1%,分布相差约2%.与传统的GPC法相比,MALLS法可获得重均分子量的绝对值,因而与GPC联用可获得更加符舍样品实际分子量分布的结果.  相似文献
