使用不同浓度的氯化镉(CdCl2)处理体外培养的条斑星鲽卵巢(BFO)细胞系细胞,利用毒理学和细胞生物学方法研究了重金属镉对BFO细胞的细胞毒性及其作用机理。毒性作用研究结果显示,BFO细胞对CdCl2敏感,浓度大于10ixmol/L的CdCl2对细胞的生长具有显著的抑制作用,能引起细胞内超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GST—Px)活性的持续降低和丙二醛(MDA)含量的持续升高,且具有浓度依赖性;机理研究结果显示.浓度大于40μmol/L的CdCl2可引起BFO细胞出现典型的凋亡细胞形态,BFO细胞在AO/EB荧光双染色中的质膜通透性显著提高,在单细胞凝胶电泳中出现明显的彗尾,且细胞凋亡率、彗尾的长度和亮度随CdCl2浓度的增加而逐渐升高,并具有浓度依赖性。可见,镉对BFO细胞具有显著的毒性作用,其毒性作用是通过诱导细胞凋亡来实现的,为利用BFO细胞系研究镉等重金属的细胞毒性及其作用机理奠定了基础。  相似文献

镉是一种致癌物质,其致癌作用与Cd^2 有着直接或间接的作用.用一定浓度的CdCl2液处理质粒pUC119和pCPP9,通过琼脂糖凝胶电泳分析,研究了镉对质粒DNA结构的影响.结果表明,CdCl2溶液2.5μg/mL作用12h或20μg/mL作用1h后,能导致质粒pCPP9的DNA损伤;浓度40μg/mL以上的CdCl2作用12h后,引起质粒pUC119的DNA损伤;且随着CdCl2浓度增大和作用时间的延长,对质粒DNA的损伤作用增强.  相似文献

目的观察依地福新对体外培养的Jurkat细胞的生长抑制作用,并进一步探讨其作用机制。方法应用MTT法检测细胞增殖活性;流式细胞仪检测细胞周期分布;Annexin V—FITC染色法检测细胞凋亡率。结果不同浓度依地福新处理Jurkat细胞24~96h后,细胞增殖显著受到抑制,并呈现浓度及时间依赖性;1.0μmol/L、5.0ymol/L、10.0μmol/L依地福新处理72h后,Jurkat细胞G0/G1期细胞数量显著增加,S期细胞数量显著降低(P〈0.01);各浓度组细胞凋亡率均显著增加(P〈0.01)。结论依地福新对Jurkat细胞具有生长抑制作用,其机制与阻滞细胞周期及诱导凋亡有关。  相似文献

培养的人喉癌细胞 Hep‐2及肝癌细胞HepG‐2,分别用不同浓度姜黄素(CUR ,0~50μmol/L)和紫杉醇(PTX ,0~5 nmol/L )单独处理或联合处理(5μmol/L CUR+0.5 nmol/L PTX、10μmol/L CUR+0.5 nmol/L PTX).处理48 h后,用WST‐1细胞增殖试剂盒检测细胞增殖情况,再用结晶紫染色检测活细胞的密度,最后用Hoechst 33258凋亡染色试剂盒检测细胞凋亡情况.结果显示:姜黄素或紫杉醇单独处理对 Hep‐2细胞和 H epG‐2细胞的增殖均有抑制作用,并呈现出一定剂量效应关系;姜黄素与紫杉醇联合处理后细胞的增殖速率要比紫杉醇单独处理的明显降低;同时,姜黄素与紫杉醇联合处理后细胞的凋亡要比紫杉醇单独处理的更为明显.上述研究结果说明,姜黄素对紫杉醇抑制肿瘤细胞H ep‐2和H epG‐2的增殖有促进作用,并能促进紫杉醇诱导细胞的凋亡.  相似文献

将离体培养的大叶冬青幼叶外植体在含有TDZ(噻吩隆,浓度0.1~5.0μmol/L)与IAA(浓度0.1~5.0μmol/L)的wPM培养基上培养,能够产生愈伤组织。当TDZ浓度不变时,5.0μmol/LIAA处理产生的愈伤组织块直径最大;愈伤组织块转培到含有不同浓度TDZ与IAA的WPM培养基上,在5.0μmol/LTDZ 3.0μmol/LIAA的培养基上出现较多不定芽苗(每一愈伤组织块平均产生11.2株);几种细胞分裂素作用比较研究表明:在BA、TDZ与ZT(均为15.0μmol/L)处理中,较好的芽苗增殖系数分别为3.7,6.0和7.4,BA与2-ip(均为3.0μmol/L)处理时芽苗高生长可达34mm和45mm。  相似文献

以CdCl2作为诱导剂,用不同浓度的CdCl2对扁藻进行诱导培养,随着CdCl2浓度的升高,扁藻的生长率逐渐降低。扁藻不能耐受1mmol/L以上的CdCl2.对MTL提取液进行紫外吸收、原子吸收等测定,结果表明扁藻类金属硫蛋白的合成随着CdCl2浓度的升高而增大,至0.25mmol/L达到最大值,每克湿藻能获得1.98mgMTI,诱导效果显著。  相似文献

目的观察慢性心衰患者血尿酸浓度,为临床判断慢性心衰疗效及预后提供帮助。方法(1)检测70例心功能Ⅰ~Ⅱ级,50例心功能Ⅲ级,73例心功能Ⅳ级患者血尿酸浓度,对其比较;(2)对193例心功能Ⅰ-Ⅳ级病因分别为冠心病、扩张性心肌病、高血压心脏病、风心病组患者观测其预后;(3)对例慢性心力衰竭患者治疗前后血尿酸浓度比较。结果(1)心功能Ⅰ~Ⅱ级组血尿酸浓度(209±29)μmol/L,HI级组血尿酸浓度(448.05±54)μmol/L,Ⅳ级组血尿酸浓度(526.11±102.8)μmol/l,心功能Ⅰ-Ⅱ级组分别与Ⅲ级组、Ⅳ级组间比较差异有统计学意义(P〈O.05),Ⅲ级组与Ⅳ级组间差异有统计学意义(P〈0.05);(2)心功能Ⅰ-Ⅱ级组治疗前(209±29)μmol/L,治疗后(178±36)μmol/L,差异无统计学意义(P〉0.05),Ⅲ级组治疗前后分别为(448.05±54)μmol/L、(276±98)μmol/L,Ⅳ级组治疗前后分别为(526.11±102.8)μmol/L、(324±106)μmol/L,Ⅲ级组与Ⅳ级组治疗前后差异有统计学意义(P〈0.05)。结论心衰越严重血尿酸浓度越高,预后越差。心功能改善后血清尿酸浓度明显下降,动态检测血清尿酸浓度是判断心衰的预后的一个有意义的生化指标。  相似文献

目的:实验结果表明4HPR抑制Hela细胞生长的机理之一是诱导该细胞凋亡。本文用流式细胞仪检测不同浓度的4HPR作用不同时间Hela细胞凋亡的百分比及细胞周期变化。方法:体外细胞培养及流式细胞仪检测法。结果:2μg/m的4HPR作用24、48、72h后,Hela细胞的凋亡百分比分别为4.1%、6.6%、26%;4μg/ml的4HPR分别为5.1%、8.3%、28%、8μg/ml的4HPR分别为5.9%、6.2%、34%。细胞周期分布图中,G1细胞明显减少。结论:上述结果提示Hela细胞凋亡的百分比与4HPR的作用时间、作用浓度呈正相关。  相似文献

为了研究甲醛致昆虫细胞DNA-蛋白质的交联作用(DNA—protein crosslinks,DPC)和DNA的断裂作用,以斜纹夜蛾SL-1细胞为材料,采用KCI-SDS沉淀法和彗星实验来检测液态甲醛染毒后SL-1细胞中DPC的含量及DNA的断裂效应.KCISDS沉淀法的结果表明,低浓度的液态甲醛(25μmol/L,125μmol/L)不能引起DPC,较高浓度(625μmol/L)可以引起明显的DPC(P〈0.01);而彗星实验的结果则显示甲醛在低浓度(5μmol/L,25μmol/L)时可以引起DNA链的断裂(P〈0.01),在较高浓度(625μmol/L)时尾部DNA%和尾矩比空白对照显著降低(P〈0.01),表明此时甲醛所致的DPC掩盖了DNA断裂的作用.结论是甲醛在较高浓度时可以导致明显的DPC作用,而在低浓度时以DNA断裂为主.  相似文献

目的:探讨白血病细胞对三氧化二砷(As2O3)敏感性的差异。方法:用台盼蓝色染色区分死活细胞,检测细胞活性;利用碘化丙啶(PI)染色和流式细胞技术测定凋亡细胞和细胞DNA的含量。结果:0.50μmol/L以上浓度的As2O3与细胞共同孵育48h以后对NB4细胞有明显的杀伤作用;而对K562细胞来说,As2O3浓度要达到2.0μmol/L以上,72h以后才有明显的杀伤作用。结论:白血病细胞对As2O3有不同的敏感性。  相似文献

There are numerous geometric objects stored in the spatial databases. An importance function in a spatial database is that users can browse the geometric objects as a map efficiently. Thus the spatial database should display the geometric objects users concern about swiftly onto the display window. This process includes two operations:retrieve data from database and then draw them onto screen. Accordingly, to improve the efficiency, we should try to reduce time of both retrieving object and displaying them. The former can be achieved with the aid of spatial index such as R-tree, the latter require to simplify the objects. Simplification means that objects are shown with sufficient but not with unnecessary detail which depend on the scale of browse. So the major problem is how to retrieve data at different detail level efficiently. This paper introduces the implementation of a multi-scale index in the spatial database SISP (Spatial Information Shared Platform) which is generalized from R-tree. The difference between the generalization and the R-tree lies on two facets: One is that every node and geometric object in the generalization is assigned with a importance value which denote the importance of them, and every vertex in the objects are assigned with a importance value,too. The importance value can be use to decide which data should be retrieve from disk in a query. The other difference is that geometric objects in the generalization are divided into one or more sub-blocks, and vertexes are total ordered by their importance value. With the help of the generalized R-tree, one can easily retrieve data at different detail levels.Some experiments are performed on real-life data to evaluate the performance of solutions that separately use normal spatial index and multi-scale spatial index. The results show that the solution using multi-scale index in SISP is satisfying.  相似文献

The discovery of the prolific Ordovician Red River reservoirs in 1995 in southeastern Saskatchewan was the catalyst for extensive exploration activity which resulted in the discovery of more than 15 new Red River pools. The best yields of Red River production to date have been from dolomite reservoirs. Understanding the processes of dolomitization is, therefore, crucial for the prediction of the connectivity, spatial distribution and heterogeneity of dolomite reservoirs.The Red River reservoirs in the Midale area consist of 3~4 thin dolomitized zones, with a total thickness of about 20 m, which occur at the top of the Yeoman Formation. Two types of replacement dolomite were recognized in the Red River reservoir: dolomitized burrow infills and dolomitized host matrix. The spatial distribution of dolomite suggests that burrowing organisms played an important role in facilitating the fluid flow in the backfilled sediments. This resulted in penecontemporaneous dolomitization of burrow infills by normal seawater. The dolomite in the host matrix is interpreted as having occurred at shallow burial by evaporitic seawater during precipitation of Lake Almar anhydrite that immediately overlies the Yeoman Formation. However, the low δ18O values of dolomited burrow infills (-5.9‰~ -7.8‰, PDB) and matrix dolomites (-6.6‰~ -8.1‰, avg. -7.4‰ PDB) compared to the estimated values for the late Ordovician marine dolomite could be attributed to modification and alteration of dolomite at higher temperatures during deeper burial, which could also be responsible for its 87Sr/86Sr ratios (0.7084~0.7088) that are higher than suggested for the late Ordovician seawaters (0.7078~0.7080). The trace amounts of saddle dolomite cement in the Red River carbonates are probably related to "cannibalization" of earlier replacement dolomite during the chemical compaction.  相似文献

Future mobile communication systems aim at providing very high data transmission rates, even in high-mobility scenarios such as high-speed wheel-track trains, maglev trains, highway vehicles, airplanes, guided missiles or spacecraft. A particularly important commercial application is the strong and increasing worldwide demand for high- speed broadband wireless communications (up to 574.8 km/ h test speeds or 380 km/h commercial speeds) in railways, providing data, voice and video services for applications such as onboard entertainment services to passengers, train control, train dispatch, train sensor status handling and sur- veillance. In such high-mobility scenarios, there are a number of communication challenges, including fast hand- over, location updating, high-speed channel modeling, estimation and equalization, anti-Doppler spreading tech- niques, fast power control, and dedicated network architec- ture. 本文叙述了对海南岛及其毗邻大陆边缘白垩纪到第四纪地层岩石进行古地磁研究的全部工作过程。通过分析岩石中剩余磁矢量的磁偏角及磁倾角的变化,提出海南岛白垩纪以来经历的构造演化模式如下:早期伴随顺时针旋转而向南迁移,后期伴随逆时针转动并向北运移。联系该地区及邻区的地质、地球物理资料,对海南岛上述的构造地体运动提出以下认识:北部湾内早期有一拉张作用,主要是该作用使湾内地壳显著伸长减薄,形成北部湾盆地。从而导致了海南岛的早期构造运动,而海南岛后期的构造运动则主要是受南海海底扩张的影响。海南地体运动规律的阐明对于了解北部湾油气盆地的形成演化有重要的理论和实际意义。  相似文献

Being the primary media of geographical information and the elementary objects manipulated, almost all of maps adopt the layer-based model to represent geographic information in the existent GIS. However, it is difficult to extend the map represented in layer-based model. Furthermore, in Web-Based GIS, It is slow to transmit the spatial data for map viewing. In this paper, for solving the questions above, we have proposed a new method for representing the spatial data. That is scale-based model. In this model we represent maps in three levels: scale-view, block, and spatial object, and organize the maps in a set of map layers, named Scale-View, which associates some given scales.Lastly, a prototype Web-Based GIS using the proposed spatial data representation is described briefly.  相似文献
