摘要:目的 研究 iRhom2Uncv 小鼠乳腺表型相关的差异表达基因,为深入研究 iRhom2Uncv 小鼠乳腺发育障碍的相关机制提供靶点信息。 方法 对 12 周龄的野生型和 iRhom2Uncv 小鼠的乳腺组织进行 whole-mounts 染色,分析乳腺发育形态,收集乳腺组织进行转录组测序,筛选差异表达基因,ELISA 检测血清激素水平。 结果 与野生型小鼠相比,iRhom2Uncv 小鼠乳腺侧枝导管较稀疏,差异最显著的 18 个基因中有 6 个在 iRhom2Uncv 小鼠中表达上调,12 个表达下调。 其中 3 个催乳素家族基因在 iRhom2Uncv 小鼠中不表达,且在血清中几乎检测不到催乳素表达,而血清中的雌激素水平相比野生型小鼠较高。 结论 iRhom2Uncv 突变的小鼠存在乳腺发育障碍,可能与 iRhom2Uncv 小鼠中催乳素和 Zfp281 的缺失表达有关,生物信息学分析发现,iRhom2Uncv 小鼠神经发育可能存在不同于野生型小鼠的表型。  相似文献

Milk contains a large amount of protein, most of which consists of a few major species synthesized in the mammary gland. The genes encoding these proteins are single-copy, and expressed during pregnancy and lactation. Although beta-lactoglobulin (BLG) is the major protein in the whey of ruminants, it is not present in rodent milk. We have generated transgenic mice carrying the sheep BLG gene, and show that in such mice, BLG is specifically and abundantly expressed in the mammary gland during lactation. This results in a remarkable alteration of milk composition. These findings suggest that the manipulation of milk composition by gene transfer has considerable potential for the improvement of dairy animals.  相似文献

Lactogenic receptor regulation in hormone-stimulated steroidogenic cells   总被引:1,自引:0,他引:1  
Receptor sites for lactogenic hormones such as prolactin (PRL), human growth hormone (HGH), and placental lactogens, are widely distributed in mammalian tissues, including mammary glands, steroid-secreting cells of the adrenal, testis, and ovary, and target cells of steroid hormone action such as liver, prostrate, and kidney. Although the biological functions of lactogen binding sites remain uncertain, a relationship between prolactin and lipoprotein metabolism is implied by the occurrence of prolactin receptors in steroidogenic cells of the gonads and adrenal, and by the ability of prolactin to increase esterified cholesterol in the testis. Recently, loss of testicular prolactin receptors has been observed following elevation of circulating luteinising hormone (LH) concentrations by the gonadotropin releasing hormone (GnRH) and its agonist analogues. The hormone dependence of lactogen receptor sites in steroid-secreting cells was further analysed in rat testis, ovary, and adrenal glands after treatment with the respective trophic hormones, gonadotropin and ACTH. In each of these tissues, rapid and transient loss of lactogen receptors was observed after trophic hormone stimulation. These findings indicate that increased turnover of lactogen receptors is an important component of the target-cell response, and suggest that prolactin receptors might be involved in the transport of lipoprotein precursors for steroid biosynthesis.  相似文献

Relaxin affects the central control of oxytocin release   总被引:1,自引:0,他引:1  
In several species the myometrium is quiescent shortly before parturition. At this time high titres of relaxin are present in the plasma and there is evidence that the hormone has a direct inhibitory action on the uterine muscle. Relaxin could also contribute to uterine quiescence by inhibiting oxytocin release. To determine whether relaxin has a central action on the release of oxytocin, we have studied the effect of intravenous injections of porcine relaxin on milk ejection in the anaesthetized lactating rat. We report that reflex milk ejection was suppressed by relaxin in a dose-dependent manner, the onset of inhibition being rapid and lasting from 10 to 60 min. After the period of inhibition the normal temporal pattern of reflex milk ejection was resumed. Mammary sensitivity to exogenous or endogenous oxytocin was reduced by relaxin but not sufficiently to explain the effects observed. Furthermore, relaxin (1 microgram per rat) injected into the cerebral ventricles profoundly disturbed the pattern of reflex milk ejection without affecting the response of the mammary gland to oxytocin. These results suggest a novel role for relaxin within the central nervous system. The site in the brain at which the effects of relaxin are exerted remains unknown.  相似文献

目的 是分析大熊猫初乳和常乳中差异表达的乳清蛋白.利用双向电泳-质谱技术首次对一只大熊猫产后第1d和第22 d的乳清蛋白进行初步的分离与鉴定,结果成功得到11种差异表达的蛋白质,值得注意的是,其中有3种蛋白质与免疫、抗菌有关,包括抗白细胞蛋白酶、溶菌酶C,C反应蛋白.推测这3种蛋白质可能参与大熊猫乳腺或幼仔的防御功能.研究结果表明,用双向电泳-质谱技术可以分析大熊猫初乳和常乳中的差异表达蛋白质,有助于认识大熊猫乳的组成和功能特性.  相似文献

Lactation is almost exclusively associated with pregnancy and giving birth. Although lactation can be induced without a preceding pregnancy in some species, this requires exogenous hormones, artificially intense or extended suckling or both. Spontaneous lactation, lactation by females that have neither been pregnant nor experimentally manipulated, is extremely unusual among eutherians. Among nondomesticated animals, spontaneous lactation has been observed repeatedly only in the dwarf mongoose Helogale parvula. We report here spontaneous lactation by free-living dwarf mongooses using data on urinary oestrogen conjugate concentrations (n = 560, 65 females) and body weight (n = 3,096, 25 females) from a population in Serengeti National Park, Tanzania. We use demographic data from this population to demonstrate that spontaneous lactation, and thus the endocrine phenomena that induce it, increase the evolutionary fitness of lactating females.  相似文献

We have constructed a mammary gland expression vector that contained the goat β-casein gene pro-moter, 5'upstream regulatory region, exons 1, 2, intron 1 as well as the human serum albumin (hALB) mini-gene (including the full-long sequences of hALB cDNA and its intron 1). Injection of the vector into mouse tail veins showed that the recombinant construct was expressed only in mammary glands. The vector was microinjected into the mouse fertilized eggs, followed by transferring the eggs into the foster mice. 33 F0 mice were obtained. Of the 33, 8 mice (5 , 3 ) were transgenic with hALB gene integration identified by PCR as well as Southern blot hybridization. The integration rate was 24.2% (8/33). Western blot analysis showed that 3 female transgenic mice had hALB expression in their milk. The hALB contents in milk reached 3.54, 0.21 and 3.03 g/L, respectively.  相似文献

The anterior pituitary gland, which is derived from a common primordium originating in Rathke's pouch, contains phenotypically distinct cell types, each of which express discrete trophic hormones: adrenocorticotropic hormone (ACTH), thyroid-stimulating hormone (TSH), prolactin, growth hormone, and follicle stimulating hormone (FSH)/luteinizing hormone (LH). The structurally related prolactin and growth hormone genes, which are evolutionarily derived from a single primordial gene, are expressed in discrete cell types--lactotrophs and somatotrophs, respectively--with their expression virtually limited to the pituitary gland. The pituitary hormones exhibit a temporal pattern of developmental expression with rat growth hormone and prolactin characteristically being the last hormones expressed. The reported co-expression of these two structurally related neuroendocrine genes within single cells prior to the appearance of mature lactotrophs, in a subpopulation of mature anterior pituitary cells, and in many pituitary adenomas raises the possibility that the prolactin and growth hormone genes are developmentally controlled by a common factor(s). We now report the identification and characterization of nucleotide sequences in the 5'-flanking regions of the rat prolactin and growth hormone genes, respectively, which act in a position- and orientation-independent fashion to transfer cell-specific expression to heterologous genes. At least one putative trans-acting factor required for the growth hormone genomic sequence to exert its effects is apparently different from those modulating the corresponding enhancer element(s) of the prolactin gene because a pituitary 'lactotroph' cell line producing prolactin but not growth hormone selectively fails to express fusion genes containing the growth hormone enhancer sequence.  相似文献

目的 了解不同妊娠期、泌乳期绿色荧光(Green Fluorescence Protein,简称GFP)裸鼠雌鼠血浆中雌激素、孕激素、泌乳素及各组织中泌乳素及泌乳素受体mRNA水平变化。方法 通过电化学发光法测定血浆中激素的水平,通过荧光定量PCR法测定泌乳素及泌乳素受体mRNA相对表达量。结果 GFP裸鼠雌鼠妊娠期内雌激素和泌乳素水平不断下降、孕激素的水平先升后缓慢下降、组织中泌乳素mRNA水平较低、泌乳素受体水平较高;而在泌乳期内雌激素、孕激素水平较稳定、组织中泌乳素mRNA水平较高,泌乳素受体水平较低。结论 不同时期GFP裸鼠雌鼠体内性激素的水平不同,参与裸鼠妊娠调控过程,为解决雌鼠不孕、易流产等问题提供了基础参考值和内分泌水平的解决思路。  相似文献

采用CMT法对河北省数家奶牛场泌乳牛群进行了隐性乳房炎检测.共检测297头泌乳牛1 134个乳区.结果表明:奶牛隐性乳房炎阳性率为56.9%,乳区阳性率为31.4%.数据分析表明,奶牛隐性乳房炎的阳性率与营养状况、胎次、泌乳期有很大关系.不同营养状况、胎次和泌乳期奶牛的隐性乳房炎阳性率差异显著(P<0.05),而不同乳区隐性乳房炎的阳性率则差异不显著(P>0.05).  相似文献

The hormone relaxin has recently been shown to inhibit not only uterine muscle contraction, but also the release of oxytocin into the plasma. Intravenous injection of porcine relaxin in anaesthetized lactating rats inhibits milk ejection and injection of relaxin into the cerebral ventricles disturbs the pattern of the milk ejection reflex. Recent experiments performed in vivo indicate that relaxin might act not only in the uterus, but also in the hypothalamus and possibly in the neurohypophysis. We tested this hypothesis in vitro by studying the effect of relaxin on hormone release from isolated neural lobes of the pituitary and isolated neurosecretory nerve endings of the neurohypophysis from the rat. We report here that relaxin has a dual effect on neurohypophysial hormone secretion. Under basal conditions, vasopressin and oxytocin release was inhibited by relaxin but, when the nerve endings were depolarized, vasopressin and oxytocin secretion was potentiated. We also found that relaxin acts at a stage before the increase in cytoplasmic free Ca2+ that is necessary for inducing hormone release, possibly by gating the calcium channel.  相似文献

目的利用从临床乳腺炎奶牛乳样中分离纯化的金黄色葡萄球菌(S.aureus)感染小鼠乳腺,建立奶牛金黄色葡萄球菌乳腺炎小鼠模型。方法将产后8~10 d的ICR远交系母鼠随机分为3组,包括无菌生理盐水对照组,低剂量攻菌组(含金黄色葡萄球菌2×102CFU)和高剂量攻菌组(含金黄色葡萄球菌5×106CFU)。以小鼠第4对乳腺为攻菌部位,用非损伤方法经乳头管注入生理盐水或金黄色葡萄球菌。攻菌24h后采集乳汁进行白细胞计数;从眼球采血,进行血常规检测和流式细胞计数;之后处死母鼠,取乳腺组织进行匀浆细菌计数,并做乳腺组织切片。结果高剂量攻菌组不论是体温、组织学观察,还是乳汁白细胞计数、乳腺匀浆细菌计数、血常规指标和流式细胞计数,都和对照组有显著或极显著差异(P〈0.05);而低剂量攻菌组与对照组相比,除血液CD4+/CD8+比值差异显著外(P〈0.05),其余指标差异均不显著。结论金黄色葡萄球菌浓度为5×106CFU、攻菌时间24 h时,成功建立了远交系小鼠金黄色葡萄球菌乳腺炎模型,而2×102CFU细菌浓度则不适于远交系小鼠乳腺炎模型的建立。  相似文献

Bm-152是前期研究中发现的一个在家蚕丝腺中高表达的非编码RNA.采用实时荧光定量PCR技术对Bm-152在家蚕5龄到预蛹期不同发育天数丝腺中的表达谱进行检测.同时,在个体中过表达Bm-152,检测Bm-152可能参与的信号通路.结果显示,Bm-152在家蚕5龄幼虫丝腺发育过程中无明显的变化规律,但在吐丝第1天表达量较高,随后表达量又下降.通过在血淋巴中注射2μg piggyBac[A3-EGFP-A3-Bm-152]过表达载体,48h后发现Bm-152在丝腺组织中无明显过表达,但在非丝腺组织中可实现明显过表达,且表达量上调1.66倍;蜕皮激素信号通路基因Usp在非丝腺组织中上调1.95倍,E75A在非丝腺组织中上调2.26倍,保幼激素信号通路基因Met在非丝腺组织中上调1.79倍.表明非编码RNA Bm-152可能通过蜕皮激素和保幼激素信号通路参与家蚕发育,该结果为进一步探索Bm-152的功能提供了方向.  相似文献

应用组织化学方法及免疫组化技术对正常组和急性攻毒组大鼠乳腺组织血管内皮生长因子(Vascular Endothelial Growth Factor,VEGF)的表达水平、肥大细胞的活力和炎性细胞的动态变化进行了研究.结果表明,急性攻毒后6 h乳腺组织VEGF在腺泡间、小叶间质、血管周围及部分肥大细胞内的表达出现一个峰值,而后表达开始减弱,攻毒48 h后表达又迅速增强;攻毒组大鼠的肥大细胞及其脱颗粒和炎性细胞随着攻毒时间的延长急剧增多,均极显著高于正常组(P〈0.01),且其变化趋势均与VEGF表达趋势相吻合;同时,小叶间质内的小血管内皮细胞开始出现裂隙、脱落损伤.由此表明,VEGF和肥大细胞参与大鼠乳腺炎的发病过程.  相似文献
