本文将具有高度特异性的生物分子核酸适配体引入分子印迹技术,并结合磁性纳米粒子材料比表面积大、操作简便性和易于修饰性于一体,制备出了新型蛋白分子印迹材料.首先获得性能更好的硅烷化的磁性微球,之后加入功能单体、交联剂和衍生化适配体,以溶菌酶为模板分子,制备出磁性溶菌酶蛋白分子印迹适配体-纳米粒子.实验结果显示,不同材料的蛋白吸附能力顺序由高到低分别为结合适配体的蛋白印迹材料,结合适配体的非蛋白印记材料,蛋白印迹材料和无适配体的非蛋白分子印记材料.初步研究表明,适配体与磁性粒子的结合可以有效提高蛋白质分子印迹材料的印迹效果.  相似文献

流行饰品是运用价格低廉的材料,采用大批量工业化生产,用于与特定服装搭配,在特定场合佩戴,表达设计者与佩戴者的思想与情绪,个性和态度的身体装饰品.相对于传统珠宝而言,流行饰品的搭配功能大于材料固有的保值功能;相对于工业产品而言,流行饰品的装饰功能大于实际功能,此外,它还需具备表达个性与态度的艺术品特质.流行饰品设计创新很大程度来源于造型的创新,而造型创新的根基在于灵活运用相关材料与工艺,该文从流行饰品材料与工艺课程入手,结合本人设计教学实践进行阶段性梳理小结,论述材料、工艺、造型三者不可分割的关系,探讨立体构成原理在该课程中应用的必要性与特异性.  相似文献

目的:分析铜绿假单胞菌(pseudomonas aeruginoso,,PA)的LesA与其靶位点的特异性结合的特性.方法:以PCR法扩增PA的LexA基因,将其克隆于原核表达载体pET-22b( ),转化大肠埃希菌BL21(DE3),IPTG诱导表达,采用镍离子亲合柱层析和Superdex-75凝胶过滤层析分离和纯化目的蛋白,以含推定LexA结合位点的片段CTGTN27ACAG为探针,用凝胶阻滞试验检测LexA蛋白与探针序列结合的能力及特异性.结果:构建了pET22b-LexA重组质粒,IPTG诱导目的蛋白表达率约为25%,获得纯度大于95%的目的蛋白;CTGTN27ACAG能与LexA蛋白发生特异性结合.结论:推定Lex4结合位点CTGTN27ACAG能与PA的LexA蛋白特异性结合,可能是LexA蛋白的结合靶位点.  相似文献

Pelota在进化上是非常保守的RNA结合蛋白,人源Pelota mRNA分布于几乎所有的组织并作为一个多功能的蛋白参与多种生物途径.为解析人源Pelota C端结构域(C-terminal domain,CTD)的晶体结构,首先在大肠杆菌(Escherichia coli)中表达,并采用亲和层析、凝胶过滤柱层析的方法,获得了纯度大于97%的蛋白.动态光散射实验表明纯化的蛋白有较高的均一性.在筛选了1 852个结晶条件后,优化的人源Pelota CTD蛋白晶体能衍射X射线至0.26nm分辨率.蛋白晶体的空间群为P6522,晶胞常数a=7.882nm,b=7.882nm,c=19.746nm.上述结果为进一步研究Pelota的功能及其与下游蛋白的相互作用奠定了结构基础.  相似文献

该工作采用分子动力学模拟和主成分分析等方法研究了抑制剂SF2535、RVX08和RVXOH与BRD4蛋白第一结构域(BRD4-1)的结合对BRD4-1蛋白构象的影响.此外,为进一步研究三个抑制剂与BRD4-1蛋白的结合能力,采用分子力学泊松-玻尔兹曼表面积(MM-PBSA)方法计算了抑制剂与BRD4-1蛋白的结合自由能.结果表明SF2535与BRD4-1蛋白的结合能力最强.本研究不仅有助于更好的了解BRD4-1的功能和内在动力学,而且还可为以BRD4-1为靶标的有效抗癌药物的研发提供理论基础.  相似文献

成人T细胞白血病(Adult T-cell leukemia,ATL)是与人类T淋巴细胞白血病1型病毒(Human T-cell leukemia virus type 1,HTLV-1)感染密切相关的恶性淋巴细胞白血病.HTLV-1反义编码的HBZ(HTLV-1 b ZIP factor)蛋白在ATL发生过程中扮演极为重要的角色.为了寻找治疗成人T细胞白血病的方法,设计了能与HBZ蛋白形成二聚体,从而封闭HBZ蛋白功能的靶向多肽R8-HBAP.通过免疫共沉淀实验,发现R8-HBAP可以有效抑制HBZ蛋白与下游靶蛋白c-Jun的结合,报告基因实验显示,R8-HBAP能阻遏HBZ蛋白对AP-1信号通路的调控作用.MTT和流式细胞术实验发现,R8-HBAP能抑制ATL细胞的恶性增殖并促进细胞凋亡.因此,靶向多肽R8-HBAP通过阻遏HBZ蛋白的功能从而抑制白血病细胞的恶性增殖,为R8-HBAP的开发利用及成人T细胞白血病的治疗提供一定的实验依据.  相似文献

采用超高压力下通电烧结技术制备了钨体积分数为0%、25%、50%、75%和100%的钨铁功能梯度材料.研究了材料组分和工艺参数对W/Fe功能梯度材料的显微形貌及力学性能的影响.结果表明:当施加压力为9 GPa,通电功率为11 kW,通电时间60 s时可以获得相对密度大于98%的钨铁功能梯度材料,其组分分布与设计成分保持一致.  相似文献

乳腺癌易感基因2(Breast Cancer Susceptibility Gene 2,BRCA2)是能抑制细胞发生恶变的关键基因.p53蛋白是参与细胞过程的主调控蛋白,p53蛋白要想行使其功能,则需要BRCA2及其产物对p53进行调控.选择BRCA2中的BRC2重复基元,用Fmoc固相合成法合成了BRC2及其3个片段.通过反相高效液相色谱(RPHPLC)和质谱对其进行表征,得到纯度大于90%的目标肽链.利用圆二色谱法初步探究了BRC2及其3个片段与靶肽p53(171-192)的相互作用.结果表明,BRC2的3个肽片段中,片段NEVGFRGFYSAHG在BRC2肽行使其功能中起着主要的作用.  相似文献

目的 对转ScPIP1基因烟草植株进行研究以检测ScPIP1的功能及转基因植株的抗逆能力.方法 使用PEG4000对材料进行人工模拟干旱胁迫;使用NaCl对材料进行盐胁迫;使用显微镜对材料叶片气孔形态及指标进行测定.材料的萌发及生长指标使用标尺测量.对所获数据进行统计学分析.结果 Pt2的气孔密度是WT和Pt5的2倍,气孔开度则是WT大于Pt5,Pt5大于Pt2.在不同浓度PEG-4000和NaCl胁迫条件下,Pt2转基因植株的萌发率均大于WT和Pt5.且在胁迫下生长过程中,Pt2的根长与株高均大于WT和Pt5.结论 ScPIP1参与了植物对非生物逆境的应答,并且Pt2是有较高抗逆能力的转基因系.  相似文献

The discovery of the prolific Ordovician Red River reservoirs in 1995 in southeastern Saskatchewan was the catalyst for extensive exploration activity which resulted in the discovery of more than 15 new Red River pools. The best yields of Red River production to date have been from dolomite reservoirs. Understanding the processes of dolomitization is, therefore, crucial for the prediction of the connectivity, spatial distribution and heterogeneity of dolomite reservoirs.The Red River reservoirs in the Midale area consist of 3~4 thin dolomitized zones, with a total thickness of about 20 m, which occur at the top of the Yeoman Formation. Two types of replacement dolomite were recognized in the Red River reservoir: dolomitized burrow infills and dolomitized host matrix. The spatial distribution of dolomite suggests that burrowing organisms played an important role in facilitating the fluid flow in the backfilled sediments. This resulted in penecontemporaneous dolomitization of burrow infills by normal seawater. The dolomite in the host matrix is interpreted as having occurred at shallow burial by evaporitic seawater during precipitation of Lake Almar anhydrite that immediately overlies the Yeoman Formation. However, the low δ18O values of dolomited burrow infills (-5.9‰~ -7.8‰, PDB) and matrix dolomites (-6.6‰~ -8.1‰, avg. -7.4‰ PDB) compared to the estimated values for the late Ordovician marine dolomite could be attributed to modification and alteration of dolomite at higher temperatures during deeper burial, which could also be responsible for its 87Sr/86Sr ratios (0.7084~0.7088) that are higher than suggested for the late Ordovician seawaters (0.7078~0.7080). The trace amounts of saddle dolomite cement in the Red River carbonates are probably related to "cannibalization" of earlier replacement dolomite during the chemical compaction.  相似文献

Language is a means of verbal communication. People use language to communicate with each other. In the society, no two speakers are exactly alike in the way of speaking. Some differences are due to age, gender, statue and personality. Above all, gender is one of the obvious reasons. The writer of this paper tries to describe the features of women's language from these perspectives: pronunciation, intonation, diction, subjects, grammar and discourse. From the discussion of the features of women's language, more attention should be paid to language use in social context. 理论推导与室内实验相结合,建立了低渗透非均质砂岩油藏启动压力梯度确定方法。首先借助油藏流场与电场相似的原理,推导了非均质砂岩油藏启动压力梯度计算公式。其次基于稳定流实验方法,建立了非均质砂岩油藏启动压力梯度测试方法。结果表明:低渗透非均质砂岩油藏的启动压力梯度确定遵循两个等效原则。平面非均质油藏的启动压力梯度等于各级渗透率段的启动压力梯度关于长度的加权平均;纵向非均质油藏的启动压力梯度等于各渗透率层的启动压力梯度关于渗透率与渗流面积乘积的加权平均。研究成果可用于有效指导低渗透非均质砂岩油藏的合理井距确定,促进该类油藏的高效开发。  相似文献

There are numerous geometric objects stored in the spatial databases. An importance function in a spatial database is that users can browse the geometric objects as a map efficiently. Thus the spatial database should display the geometric objects users concern about swiftly onto the display window. This process includes two operations:retrieve data from database and then draw them onto screen. Accordingly, to improve the efficiency, we should try to reduce time of both retrieving object and displaying them. The former can be achieved with the aid of spatial index such as R-tree, the latter require to simplify the objects. Simplification means that objects are shown with sufficient but not with unnecessary detail which depend on the scale of browse. So the major problem is how to retrieve data at different detail level efficiently. This paper introduces the implementation of a multi-scale index in the spatial database SISP (Spatial Information Shared Platform) which is generalized from R-tree. The difference between the generalization and the R-tree lies on two facets: One is that every node and geometric object in the generalization is assigned with a importance value which denote the importance of them, and every vertex in the objects are assigned with a importance value,too. The importance value can be use to decide which data should be retrieve from disk in a query. The other difference is that geometric objects in the generalization are divided into one or more sub-blocks, and vertexes are total ordered by their importance value. With the help of the generalized R-tree, one can easily retrieve data at different detail levels.Some experiments are performed on real-life data to evaluate the performance of solutions that separately use normal spatial index and multi-scale spatial index. The results show that the solution using multi-scale index in SISP is satisfying.  相似文献

本文叙述了对海南岛及其毗邻大陆边缘白垩纪到第四纪地层岩石进行古地磁研究的全部工作过程。通过分析岩石中剩余磁矢量的磁偏角及磁倾角的变化,提出海南岛白垩纪以来经历的构造演化模式如下:早期伴随顺时针旋转而向南迁移,后期伴随逆时针转动并向北运移。联系该地区及邻区的地质、地球物理资料,对海南岛上述的构造地体运动提出以下认识:北部湾内早期有一拉张作用,主要是该作用使湾内地壳显著伸长减薄,形成北部湾盆地。从而导致了海南岛的早期构造运动,而海南岛后期的构造运动则主要是受南海海底扩张的影响。海南地体运动规律的阐明对于了解北部湾油气盆地的形成演化有重要的理论和实际意义。  相似文献
