草鱼肝胰脏中两种胰蛋白酶的纯化及部分性质的研究   总被引:3,自引:1,他引:2  
本试验采用硫酸铵分级沉淀和鸡卵类粘蛋白偶联的琼脂糖凝胶亲和层析的方法,从草鱼(Ctenopharyngodon ldellus)肝胰脏中提取胰蛋白酶进行纯化,得到两种酶,即胰蛋白酶A(GTA)和胰蛋白酶B(GTB),并对其性质进行了分析。结果表明,两种酶的SDS—PAGE电泳均呈现出单一条带,表明已得到高度纯化,两种酶分子量约为27Kda。以BANPA为底物.测得草鱼胰蛋白酶ACGTA)的动力学常数(Km)为0.37mM。以TAME为底物测得两种胰蛋白酶最适反应温度为60℃,最适反应pH值为8.0,当温度超过60℃,它们不稳定,这两种胰蛋白酶在pH值9.0—11.0时较稳定。  相似文献

通过DEAE-Sephacel离子交换层析、Sephacryl S-200 HR凝胶过滤层析、羟基磷灰石层析和Phenyl Sepharose 6-Fast Flow疏水层析,从草鱼(Ctenopharyngodon idellus)肌肉中分离纯化得到了一种亮氨酸氨肽酶,并对其生物化学性质进行了研究.结果表明,草鱼肌肉亮氨酸氨肽酶分子量约为105 ku,是单体结构.最适温度为40 ℃,最适pH为7.5,属于金属蛋白酶.除了Leu-MCA,该酶对其它多种荧光合成底物也有一定程度的分解作用,但对内肽酶对应的荧光底物则没有分解作用.  相似文献

通过硫酸铵分级盐析、DEAE-Sepharose阴离子交换柱层析、Phenyl-Sepharose疏水柱层析、Sephacryl S-200凝胶过滤柱层析等方法,从日本鳗鲡(Anguilla japonica)肝胰腺中分离纯化得到胰蛋白酶,SDS-PAGE显示其分子质量为21.5 ku。以Boc-Phe-Ser-Arg-MCA为底物,胰蛋白酶最适温度和最适pH值分别为40 ℃和8.5。动力学实验表明,Km值和kcat值分别为3.1 μmol/L和59.9 s-1。丝氨酸蛋白酶抑制剂Pefabloc SC、PMSF、benzamidine、STI等对其有特异抑制效果。底物特异性结果显示,该酶特异分解Arg和Lys残基的C端。肽质量指纹图谱获得5个片段,共76个氨基酸残基,与基因文库中的日本鳗鲡胰蛋白酶氨基酸序列完全相同。说明本研究所纯化的蛋白质为胰蛋白酶。  相似文献

采用缓冲液提取、盐析、凝胶过滤及离子交换层析等方法,从苦荞种子中分离纯化出1种类胰蛋白酶水解酶,经十二烷基硫酸钠一聚丙烯酰胺凝胶电泳(SDS—PAGE)测得其分于量为67kD,等电聚焦(IEF)测得等电点为6.3.该酶可以水解胰蛋白酶的底物BApNA,而且其活性不受苦荞胰蛋白酶抑制剂的影响.通过比较,苦荞中的胰蛋白酶水解酶在性质上与报道的甜荞BAPAase极为相似,可能属于同一类蛋白酶.  相似文献

从杜仲嫩芽中提取和纯化磷酸酯酶,并对其性质进行了初步研究.粗酶液经盐析、DEAE-Cellulose离子交换柱,Sephadex G-25和Sephadex G-200层析柱进行纯化.该酶的最适pH值为8.0,最适温度为30 ℃,以对硝基酚磷酸酯(NPP)为底物,测得Km为3.10 μmol/L.酶活的变化受金属离子的影响,当Mg2+浓度为10 mmol/L时,对酶的激活作用最强.  相似文献

在有氧条件下,利用DEAE Sepharose Fast Flow弱阴离子离子交换层析和Blue Sepharose CL-6B亲和层析,同时分离并提纯克雷伯杆菌胞内的1,3-丙二醇氧化还原酶和甘油脱氢酶.研究表明,1,3-丙二醇氧化还原酶的纯化倍数为35.86倍,回收率为5.17%,该酶最适表观反应温度为57 ℃,最适反应pH值为9.5.在30 ℃及pH=8.0~10.0时,该酶具有良好的稳定性.在45 ℃和pH=9.5条件下,该酶以1,3-丙二醇和NAD+为底物,其米氏常数Km分别为15.8,0.2 mmol·L-1.1,3-丙二醇氧化还原酶对生理反应底物3-羟基丙醛活性最大,对其他醇类也有氧化能力.Mn2+对酶有显著激活作用,巯基保护剂能明显提高酶的活力.  相似文献

从青霉(Penicillium amagasakiense)发酵液中分离纯化葡萄糖氧化酶(简称GOD),通过DEAE-32离子交换柱层析、Sephacryl S-200分子筛凝胶过滤柱层析纯化,获得比活力为472 U/mg电泳单一纯酶制剂,采用SDS-PAGE电泳,表明该酶含有两个同型的亚基,分子量为150 ku.酶学性质研究表明:该酶催化葡萄糖氧化酶反应的最适pH为5.6,最适温度为40℃.酶的热稳定性研究表明:该酶在pH 5.5~8.5区间和温度低于50℃下稳定.金属离子对酶活力的影响表明:Na+、K+、Ca2+、Pb2+、Mn2+等对酶活力基本没有影响;Al3+、Zn2+对酶有一定的激活作用;Ag+、Hg2+、Mg2+、Cd2+、Cu2+、Co2+等对该酶活力有不同程度的抑制作用.该酶以葡萄糖为底物的米氏常数Km值为122.6 mmol/L,Vm为25.52 μmol/(L·min)(pH 7.0,37℃).  相似文献

采用了Sephadex G-25层析,DEAE-Sepharose Fast Flow离子交换层析和Sephacryl S-200凝胶过滤层析,对家蚕头部粗酶匀浆液中的乙酰胆碱酯酶进行纯化,得到的样品经聚丙烯酰胺凝胶电泳(PAGE)和SDS-PAGE检测均为一条带,用SDS-PAGE法测得其分子量为77.8 kDa.酶的最适反应温度为37 ℃,最适pH为7.5,对底物碘化硫代乙酰胆碱(ATChI)的Km值为0.392 mmol/L,最适底物浓度为1.6 mmol/L,在1.6 mmol/L底物浓度以上观察  相似文献

采用DEAE Sepharose离子交换层析、Butyl Sepharose疏水层析和Blue Sepharose亲和层析等分离方法,从高产D-(-)-扁桃酸的酵母菌株BY1.1b中纯化得到电泳纯的D-(-)-扁桃酸脱氢酶.采用Sephacryl S-200分子筛层析柱测得该酶的相对分子质量约60 kDa,用SDS-PAGE电泳测得该酶亚基分子量为32 kDa,表明该酶属于同型二聚体.该酶的最适反应温度为30℃,最适反应pH为6.0.该酶在低于30℃,pH5.5~8.0较为稳定.以NADH和苯乙酮酸为底物,其Km值分别为Km(NADH)=99.2μmol/L和Km(苯乙酮酸)=263.3μmol/L.  相似文献

The Williston Basin is a significant petroleum province, containing oil production zones that include the Middle Cambrian to Lower Ordovician, Upper Ordovician, Middle Devonian, Upper Devonian and Mississippian and within the Jurassic and Cretaceous. The oils of the Williston Basin exhibit a wide range of geochemical characteristics defined as "oil families", although the geochemical signature of the Cambrian Deadwood Formation and Lower Ordovician Winnipeg reservoired oils does not match any "oil family". Despite their close stratigraphic proximity, it is evident that the oils of the Lower Palaeozoic within the Williston Basin are distinct. This suggests the presence of a new "oil family" within the Williston Basin. Diagnostic geochemical signatures occur in the gasoline range chromatograms, within saturate fraction gas chromatograms and biomarker fingerprints. However, some of the established criteria and cross-plots that are currently used to segregate oils into distinct genetic families within the basin do not always meet with success, particularly when applied to the Lower Palaeozoic oils of the Deadwood and Winnipeg Formation.  相似文献

Language is a means of verbal communication. People use language to communicate with each other. In the society, no two speakers are exactly alike in the way of speaking. Some differences are due to age, gender, statue and personality. Above all, gender is one of the obvious reasons. The writer of this paper tries to describe the features of women's language from these perspectives: pronunciation, intonation, diction, subjects, grammar and discourse. From the discussion of the features of women's language, more attention should be paid to language use in social context. The discovery of the prolific Ordovician Red River reservoirs in 1995 in southeastern Saskatchewan was the catalyst for extensive exploration activity which resulted in the discovery of more than 15 new Red River pools. The best yields of Red River production to date have been from dolomite reservoirs. Understanding the processes of dolomitization is, therefore, crucial for the prediction of the connectivity, spatial distribution and heterogeneity of dolomite reservoirs.The Red River reservoirs in the Midale area consist of 3~4 thin dolomitized zones, with a total thickness of about 20 m, which occur at the top of the Yeoman Formation. Two types of replacement dolomite were recognized in the Red River reservoir: dolomitized burrow infills and dolomitized host matrix. The spatial distribution of dolomite suggests that burrowing organisms played an important role in facilitating the fluid flow in the backfilled sediments. This resulted in penecontemporaneous dolomitization of burrow infills by normal seawater. The dolomite in the host matrix is interpreted as having occurred at shallow burial by evaporitic seawater during precipitation of Lake Almar anhydrite that immediately overlies the Yeoman Formation. However, the low δ18O values of dolomited burrow infills (-5.9‰~ -7.8‰, PDB) and matrix dolomites (-6.6‰~ -8.1‰, avg. -7.4‰ PDB) compared to the estimated values for the late Ordovician marine dolomite could be attributed to modification and alteration of dolomite at higher temperatures during deeper burial, which could also be responsible for its 87Sr/86Sr ratios (0.7084~0.7088) that are higher than suggested for the late Ordovician seawaters (0.7078~0.7080). The trace amounts of saddle dolomite cement in the Red River carbonates are probably related to "cannibalization" of earlier replacement dolomite during the chemical compaction.  相似文献

以AC-13级配为基础,将橡胶颗粒代替部分集料掺入混合料中,以低温弯曲试验为评价方法对不同橡胶颗粒掺量下沥青混合料的低温抗裂性进行研究,并引入应变能密度值对混合料的低温抗裂性进行综合评价.试验结果表明:橡胶颗粒沥青混合料试件的破坏微应变均超过2 300,满足冬寒区的技术指标;无论是否掺加橡胶颗粒,随着温度的下降,沥青混合料破坏时的最大弯拉强度增大,弯拉应变降低,劲度模量增大;弯曲应变能密度在胶粒掺量为1%左右时具有较大的弯曲应变能密度值,此时橡胶颗粒沥青混合料具有较好的低温抗裂性.  相似文献

理论推导与室内实验相结合,建立了低渗透非均质砂岩油藏启动压力梯度确定方法。首先借助油藏流场与电场相似的原理,推导了非均质砂岩油藏启动压力梯度计算公式。其次基于稳定流实验方法,建立了非均质砂岩油藏启动压力梯度测试方法。结果表明:低渗透非均质砂岩油藏的启动压力梯度确定遵循两个等效原则。平面非均质油藏的启动压力梯度等于各级渗透率段的启动压力梯度关于长度的加权平均;纵向非均质油藏的启动压力梯度等于各渗透率层的启动压力梯度关于渗透率与渗流面积乘积的加权平均。研究成果可用于有效指导低渗透非均质砂岩油藏的合理井距确定,促进该类油藏的高效开发。  相似文献

Quality traits in wheat (Triticum aestirum L.) were studied by quantitative trait locus (QTL) analysis in a recombinant inbred line (RIL) population, a set of 131 lines derived from Chuan 35050 × Shannong 483 cross (ChSh). Grains from RILs were assayed for 21 quality traits related to protein and starch. A total of 35 putative QTLs for 19 traits with a single QTL explaining 7.99-40.52% of phenotypic variations were detected on 10 chromosomes, 1D, 2A, 2D, 3B, 3D, 5A, 6A, 6B, 6D, and 7B. The additive effects of 30 QTLs were positive, contributed by Chuan 35050, the remaining 5 QTLs were negative with the additive effect contributed by Shannong 483. For protein traits, 15 QTLs were obtained and most of them were located on chromosomes 1 D, 3B and 6D, while 20 QTLs for starch traits were detected and most of them were located on chromosomes 3D, 6B and 7B. Only 7 QTLs for protein and starch traits were co-located in three regions on chromosomes 1D, 2A and 2D. These protein and starch trait QTLs showed a distinct distribution pattern in certain regions and chromosomes. Twenty-two QTLs were clustered in 6 regions of 5 chromosomes. Two QTL clusters for protein traits were located on chromosomes 1D and 3B, respectively, three clusters for starch traits on chromosomes 3D, 6B and 7B, and one cluster including protein and starch traits on chromosome 1D.  相似文献
