在光照800Lux~1100Lux、温度25℃~32℃的条件下进行活性光合细菌制剂的强化培养研究,结果表明,以牛肉水提取液为主要成份,并辅以无机盐、维生素B族及药效植物提取液的培养基,在3~4天就能培养出活茵数大于50亿/毫升的活性光合细菌制剂,由此证明这种培养基是一种高效、低耗、省时的活性光合细菌制剂的培养方法。  相似文献

光合细菌PSB-B4的分离与培养条件优化研究   总被引:2,自引:0,他引:2  
利用光合细菌处理有机污水是当前污水生物处理中的一种新方法。通过筛选,从污水中分离得到一株生长较快、适应性较强的光合细菌菌株,标记为PSB-B4通过菌落形态、培养特征和菌体形态学观察,细胞吸收光谱及生理生化特性测定,初步鉴定为类球红细菌(Rhodohacter sphaeroides)。对这一光合细菌菌株的培养条件进行优化研究,最终确定菌株PSB-B4的最佳培养基配方及培养条件为:酵母膏2.0g,乙酸钠4.0g,氯化铵2.0g,蛋白胨3.0g,磷酸氢二钾0.2g,硫酸镁0.2g,氯化钠1.25g,碳酸氢钠1.0g,蒸馏水1000mL,pH=7.0,培养温度28℃,光照强度4000Lux,其中酵母膏对其生长影响最大。  相似文献

本文通过在基本培养基中添加抑制其它真菌和细菌的制剂,探索出分离木霉的培养基为基本培养基中添加适量的丙酸钠和庆大霉素。通过在不同温度和pH条件下的对比实验,确定培养最佳pH值为5.2~5.7,最佳温度为22~28℃。  相似文献

金钗石斛试管苗壮苗培养和光合速率的检测   总被引:4,自引:2,他引:2  
选用金钗石斛(Dendrobium nobileL ind l)种子播种在固体、半固体、液体、滤纸拱桥液体培养基上,作种子萌发对比试验,最终研究结果:滤纸拱桥液体培养基上萌发率最高,可达90%以上。试管苗在添加或者未添加苔藓植物提取液壮根壮苗培养基上培养,最终研究结果为:前者平均每天苗净增长0.054 g,后者平均每天苗净增长0.082 g。这一实验结果充分证实了金钗石斛在未添加苔藓植物提取液的生长超过添加苔藓植物提取液生长的51.85%。光合速率的测定得出实验结果:金钗石斛第一功能叶的光合速率最高,其它功能叶依次递减。所有功能叶光合速率总平均值为2.60μmol.m-2.s-1。  相似文献

对光合菌在天然培养基、合成培养基以及来自植物纤维素水解液的半合成培养基中的培养进行了研究 ,对适宜的培养基、营养物质的稀释度、菌种接种量、pH值等进行了选择 .对选出的 4号加有纤维素水解液的培养基 ,接入 5 %的菌液 ,pH为 7.0~ 7.5 ,温度为 2 5~ 30℃ ,厌氧条件下 ,光合菌生长良好 ,菌量达 2 .2× 10 9亿个/mL ,在加入光合菌生长促进液 (1% )后 ,菌量达 4.0 1× 10 9个 /mL .  相似文献

降解有机废水转化太阳能为氢能是一条理想制氢途径 .为使选育的菌株实用性更强 ,本文选用有机废水主要降解产物—乙酸为唯一氢供体 ,在自然生态环境条件下 ,利用紫色非硫细菌培养基 ,紫色硫细菌培养基和绿硫细菌培养基 ,从不同的水域环境中进行了产氢光合细菌的筛选 .从影响太阳能转化效率主要因素出发 ,本文对分离纯化的 1 5株光合细菌进行形态学特征研究基础上 ,着重进行了最适生长温度、光合色素成分、利用硫化物能力和耐盐能力的测定 ,并将其分为 4个类群 .在对每组细菌初筛基础上 ,本文对选育出的 8个菌株进行了生长动力学测定 ,其中Z ,SP2 ,R3,Y7菌株能利用乙酸快速生长 .对其产氢动力学研究表明 ,这些菌株均具有光合放氢活性 ,其中Z菌株产氢得率最高 .在含有 2 0mmol·L-1 乙酸钠和 5mmol·L-1 谷氨酸钠的培养基中 ,其产氢得率为 3 0 8.9ml·g-1 .这 4个菌株生理生态特性完全不同 ,且能利用乙酸光合放氢 ,在有机废水光合制氢技术中具有重要的潜在应用价值 .  相似文献

光合细菌对有机磷农药毒害大麦的缓解作用   总被引:1,自引:0,他引:1  
研究了不同浓度的光合细菌与3种有机磷农药(马拉硫磷、对硫磷、久效磷)复合作用对大麦毒性的影响.结果表明:102~106个细菌·mL-1的光合细菌对三种有机磷农药的毒害均有缓解作用,种子萌发率、根长、叶绿素含量和过氧化物酶(POD)活性都有不同程度的提高.但不同浓度的光合细菌作用效果不同,102个细菌·mL-1光合细菌可显著缓解有机磷农药对种子萌发率的抑制作用;104个细菌·mL-1光合细菌对有机磷农药毒害的根POD活性缓解作用强;而106个细菌·mL-1光合细菌则对有机磷农药毒害大麦的根长、叶绿素含量和叶POD活性的缓解效果最显著.同一浓度光合细菌对不同有机磷农药毒性的缓解作用存在差异.  相似文献

畜禽养殖污染是造成我国农村面源污染的主要原因之一.传统的畜禽粪便处理方式是通过一系列处理方法使其达标排放,浪费了其中大量的有机物、氨氮等物质.以牛粪水为处理对象,首先筛选破胶剂对其进行预处理,再将其作为微生物培养基,进行光合细菌培养.实验结果表明:在牛粪水中加入破胶剂JP-1处理后得到的上清液作为培养基原料,在温度为35.5℃、pH值为7.67~7.98、最佳光照时间为48 h的条件下培养光合细菌,其菌液吸光度达到2.185.;因此,以牛粪水为原料的光合细菌培养方法具有可行性.  相似文献

采用光合细菌中的紫色非硫细菌为材料,通过单因素试验和正交试验分析了其对碳源、氮源、酵母膏和复合无机盐的利用能力.结果表明,乙酸钠和酵母膏对紫色非硫细菌的生长影响达到显著水平;其优化培养基为每升中含乙酸钠3 g、氯化铵3 g、酵母膏0.1 g,复合无机盐溶液1 mL,这为快速培养高活性光合细菌奠定了基础.  相似文献

The Williston Basin is a significant petroleum province, containing oil production zones that include the Middle Cambrian to Lower Ordovician, Upper Ordovician, Middle Devonian, Upper Devonian and Mississippian and within the Jurassic and Cretaceous. The oils of the Williston Basin exhibit a wide range of geochemical characteristics defined as "oil families", although the geochemical signature of the Cambrian Deadwood Formation and Lower Ordovician Winnipeg reservoired oils does not match any "oil family". Despite their close stratigraphic proximity, it is evident that the oils of the Lower Palaeozoic within the Williston Basin are distinct. This suggests the presence of a new "oil family" within the Williston Basin. Diagnostic geochemical signatures occur in the gasoline range chromatograms, within saturate fraction gas chromatograms and biomarker fingerprints. However, some of the established criteria and cross-plots that are currently used to segregate oils into distinct genetic families within the basin do not always meet with success, particularly when applied to the Lower Palaeozoic oils of the Deadwood and Winnipeg Formation.  相似文献

The discovery of the prolific Ordovician Red River reservoirs in 1995 in southeastern Saskatchewan was the catalyst for extensive exploration activity which resulted in the discovery of more than 15 new Red River pools. The best yields of Red River production to date have been from dolomite reservoirs. Understanding the processes of dolomitization is, therefore, crucial for the prediction of the connectivity, spatial distribution and heterogeneity of dolomite reservoirs.The Red River reservoirs in the Midale area consist of 3~4 thin dolomitized zones, with a total thickness of about 20 m, which occur at the top of the Yeoman Formation. Two types of replacement dolomite were recognized in the Red River reservoir: dolomitized burrow infills and dolomitized host matrix. The spatial distribution of dolomite suggests that burrowing organisms played an important role in facilitating the fluid flow in the backfilled sediments. This resulted in penecontemporaneous dolomitization of burrow infills by normal seawater. The dolomite in the host matrix is interpreted as having occurred at shallow burial by evaporitic seawater during precipitation of Lake Almar anhydrite that immediately overlies the Yeoman Formation. However, the low δ18O values of dolomited burrow infills (-5.9‰~ -7.8‰, PDB) and matrix dolomites (-6.6‰~ -8.1‰, avg. -7.4‰ PDB) compared to the estimated values for the late Ordovician marine dolomite could be attributed to modification and alteration of dolomite at higher temperatures during deeper burial, which could also be responsible for its 87Sr/86Sr ratios (0.7084~0.7088) that are higher than suggested for the late Ordovician seawaters (0.7078~0.7080). The trace amounts of saddle dolomite cement in the Red River carbonates are probably related to "cannibalization" of earlier replacement dolomite during the chemical compaction.  相似文献

The writer of this paper tries to describe the features of women's language from these perspectives: pronunciation, intonation, diction, subjects, grammar and discourse. From the discussion of the features of women's language, more attention should be paid to language use in social context. 以AC-13级配为基础,将橡胶颗粒代替部分集料掺入混合料中,以低温弯曲试验为评价方法对不同橡胶颗粒掺量下沥青混合料的低温抗裂性进行研究,并引入应变能密度值对混合料的低温抗裂性进行综合评价.试验结果表明:橡胶颗粒沥青混合料试件的破坏微应变均超过2 300,满足冬寒区的技术指标;无论是否掺加橡胶颗粒,随着温度的下降,沥青混合料破坏时的最大弯拉强度增大,弯拉应变降低,劲度模量增大;弯曲应变能密度在胶粒掺量为1%左右时具有较大的弯曲应变能密度值,此时橡胶颗粒沥青混合料具有较好的低温抗裂性.  相似文献

理论推导与室内实验相结合,建立了低渗透非均质砂岩油藏启动压力梯度确定方法。首先借助油藏流场与电场相似的原理,推导了非均质砂岩油藏启动压力梯度计算公式。其次基于稳定流实验方法,建立了非均质砂岩油藏启动压力梯度测试方法。结果表明:低渗透非均质砂岩油藏的启动压力梯度确定遵循两个等效原则。平面非均质油藏的启动压力梯度等于各级渗透率段的启动压力梯度关于长度的加权平均;纵向非均质油藏的启动压力梯度等于各渗透率层的启动压力梯度关于渗透率与渗流面积乘积的加权平均。研究成果可用于有效指导低渗透非均质砂岩油藏的合理井距确定,促进该类油藏的高效开发。  相似文献

Quality traits in wheat (Triticum aestirum L.) were studied by quantitative trait locus (QTL) analysis in a recombinant inbred line (RIL) population, a set of 131 lines derived from Chuan 35050 × Shannong 483 cross (ChSh). Grains from RILs were assayed for 21 quality traits related to protein and starch. A total of 35 putative QTLs for 19 traits with a single QTL explaining 7.99-40.52% of phenotypic variations were detected on 10 chromosomes, 1D, 2A, 2D, 3B, 3D, 5A, 6A, 6B, 6D, and 7B. The additive effects of 30 QTLs were positive, contributed by Chuan 35050, the remaining 5 QTLs were negative with the additive effect contributed by Shannong 483. For protein traits, 15 QTLs were obtained and most of them were located on chromosomes 1 D, 3B and 6D, while 20 QTLs for starch traits were detected and most of them were located on chromosomes 3D, 6B and 7B. Only 7 QTLs for protein and starch traits were co-located in three regions on chromosomes 1D, 2A and 2D. These protein and starch trait QTLs showed a distinct distribution pattern in certain regions and chromosomes. Twenty-two QTLs were clustered in 6 regions of 5 chromosomes. Two QTL clusters for protein traits were located on chromosomes 1D and 3B, respectively, three clusters for starch traits on chromosomes 3D, 6B and 7B, and one cluster including protein and starch traits on chromosome 1D.  相似文献
