电泳羟基磷灰石色谱吸附BSA机理   总被引:3,自引:0,他引:3  
将电泳与羟基磷灰石色谱技术耦合 ,开发了一种新的电泳色谱技术。建立了羟基磷灰石 (HAP)吸附牛血清白蛋白 (BSA)的物理模型和数学模型 ,建立了不同的实验装置以研究电场下 HAP吸附 BSA的反应动力学特性以及电动色谱吸附 BSA过程的传质动力学特性。实验结果表明 ,施加电场不会改变吸附反应的平衡常数与速率 ,但在电泳羟基磷灰石色谱装置中 ,施加电场在羟基磷灰石颗粒表面产生的电渗流能够加强扰动 ,减少液膜扩散阻力 ,提高羟基磷灰石色谱的动态吸附容量  相似文献

在以 P25 二氧化钛为钛源, 水热反应时间为1, 2, 3, 4天条件下, 合成钛酸纳米管(TNTs)形成过程中的4种产物, 采用XRD, SAXS和N2吸附/脱附等温线等手段进行表征。以Cd(Ⅱ)为代表污染物, 研究4种产物对Cd(Ⅱ)的吸附, 并着重分析4种产物的结构对其吸附能力的影响。粉末XRD结果表明, TNTs的4种产物均为单斜晶胞结构。SAXS和N2吸附/脱附表征证实, 水热法制备TNTs经历二氧化钛源结构解体→钛酸纳米管形成→管状结构解体的过程, TNTs-3d具有最为规整的管状形貌。吸附等温线结果表明, Langmuir-Freundlich模型能够更好地拟合吸附等温结果, 且Cd(Ⅱ)的吸附量大小顺序为TNTs-3d ≈ TNTs-1d > TNTs-4d > TNTs-2d。进一步的分析表明, 样品的回转半径、孔体积对吸附量影响较小, 而比表面积和平均孔径可能会显著影响吸附容量。  相似文献

将末端带OH、COOH、NH2、CH3的巯基化合物自组装于金片表面形成膜,利用表面等离子体共振(SPR)技术实现了牛血清白蛋白(BSA)在金片表面自组装膜上的非特异性吸附研究.以铁氰化钾为探针,利用循环伏安(CV)和电化学阻抗(EI)方法探究了膜在表面的形态.通过测量膜表面对水的静态接触角表征膜的形成及质量.SPR实时监测得到BSA在自组装膜表面的结合强弱顺序为:CH3>NH2>COOH>OH.同时还研究了形成自组装膜物质的链长对其抗非特异性吸附效果的影响.  相似文献

对在不同工艺下制备的两组纳米SiC颗粒分别进行了小角X射线散射(SAXS)、X射线衍射(XRD)和扫描电镜(SEM)测试.结果显示,两组颗粒试样存在尺寸和粒度分布差异,不同测量方法所包含的信息各有侧重.利用二维探测器XRD系统进行了SAXS和XRD测量,探讨了该系统在SAXS测量中的数据采集和处理方法,对这些测试结果及其与SEM的结果进行了综合分析比较.发现SAXS更适合于样品尺寸的总体状态表征。能提供更丰富的信息,计算结果更可靠.  相似文献

以天然产物油菜秸秆纤维素粉为基质,二甲基甲酰胺为交联剂,磷酸为修饰剂,制备了新型磷酸化油菜秸秆纤维素生物吸附剂.用红外光谱、透射电子显微镜和X射线光电子能谱,对油菜秸秆纤维素粉和磷酸化油菜秸秆纤维素吸附剂进行了表征.研究了油菜秸秆纤维素粉改性前后溶液的pH、吸附时间、BSA的初始浓度、温度和离子强度等因素对牛血清白蛋白(BSA)的吸附的影响.结果表明:在25℃,pH 5.0、6.0,吸附20 h的条件下,磷酸化油菜秸秆纤维素粉微球对BSA的吸附容量为127.39 mg/g,而未修饰油菜秸秆纤维素粉微球对BSA的吸附容量只有68.98 mg/g,说明改性获得较好的性能.吸附剂的吸附等温线符合Langmuir方程,属单分子层吸附.  相似文献

根据物料恒算原理,采用分光光度法研究了37℃下牛血清蛋白(BSA)在γ-(甲基丙烯酰氧)丙基三甲氧基硅烷(KH570)与甲基丙烯酸-2-羟基乙酯(HEMA)及N-乙烯基吡咯烷酮(NVP)共聚物水凝胶膜上的等温吸附作用.结果表明,BSA的吸附量随溶液浓度增大而增大,为匀速吸附过程,其后期吸附速度受BSA向含硅水凝胶本体扩散的速度控制,低于前期吸附速度;BSA的吸附量在pH值为4.5附近为最大,酸性或碱性过大均导致吸附量下降;溶液中离子产生的“盐效应”、凝胶中NVP或者KH 570组分的增加有利于BSA的吸附.  相似文献

应用东南大学生物医学工程系吴健雄实验室研制的RIfS生物传感器,对人工合成羟基磷灰石和H50-50型聚氨酯两种材料对小牛血清白蛋白BSA、人血浆纤维蛋白原FIB和人免疫球蛋白IgG的吸附性能进行了实时原位动态研究。根据RIfS表征原理和蛋白质结构大小,引入了吸附层数计算公式,求出了2种实验材料对不同蛋白质的吸附层数。得出羟基磷灰石和H50-50型聚氨酯材料对3种蛋白质实际吸附层数分别为:IgG1.0751,BSA0.9684,FIB0.7464和IgG0.8199,BSA0.7964,FIB0.6120。证实了用RIfS生物传感器进行生物材料生物相容性研究的可行性。  相似文献

从宏观上考察聚合物在地层的分布,微观上测定表征存在状态的各个参数,采用三管并联岩心和平板模型实验研究聚合物分布规律,通过物理模拟实验测定表征聚合物存在状态的参数,同时测定不同聚合物质量浓度下的静态吸附量和动态滞留量,采用双段塞法测定不可及孔隙体积及动态滞留量等参数,并改进实验数据的处理过程.结果表明:聚合物在纵向上主要进入高渗透层,平面上主要分布在主流线区域,两翼部分质量浓度较低;静态吸附量远大于动态滞留量,并且其比值随着聚合物质量浓度的变化而变化.  相似文献

The Williston Basin is a significant petroleum province, containing oil production zones that include the Middle Cambrian to Lower Ordovician, Upper Ordovician, Middle Devonian, Upper Devonian and Mississippian and within the Jurassic and Cretaceous. The oils of the Williston Basin exhibit a wide range of geochemical characteristics defined as "oil families", although the geochemical signature of the Cambrian Deadwood Formation and Lower Ordovician Winnipeg reservoired oils does not match any "oil family". Despite their close stratigraphic proximity, it is evident that the oils of the Lower Palaeozoic within the Williston Basin are distinct. This suggests the presence of a new "oil family" within the Williston Basin. Diagnostic geochemical signatures occur in the gasoline range chromatograms, within saturate fraction gas chromatograms and biomarker fingerprints. However, some of the established criteria and cross-plots that are currently used to segregate oils into distinct genetic families within the basin do not always meet with success, particularly when applied to the Lower Palaeozoic oils of the Deadwood and Winnipeg Formation.  相似文献

The writer of this paper tries to describe the features of women's language from these perspectives: pronunciation, intonation, diction, subjects, grammar and discourse. From the discussion of the features of women's language, more attention should be paid to language use in social context. The discovery of the prolific Ordovician Red River reservoirs in 1995 in southeastern Saskatchewan was the catalyst for extensive exploration activity which resulted in the discovery of more than 15 new Red River pools. The best yields of Red River production to date have been from dolomite reservoirs. Understanding the processes of dolomitization is, therefore, crucial for the prediction of the connectivity, spatial distribution and heterogeneity of dolomite reservoirs.The Red River reservoirs in the Midale area consist of 3~4 thin dolomitized zones, with a total thickness of about 20 m, which occur at the top of the Yeoman Formation. Two types of replacement dolomite were recognized in the Red River reservoir: dolomitized burrow infills and dolomitized host matrix. The spatial distribution of dolomite suggests that burrowing organisms played an important role in facilitating the fluid flow in the backfilled sediments. This resulted in penecontemporaneous dolomitization of burrow infills by normal seawater. The dolomite in the host matrix is interpreted as having occurred at shallow burial by evaporitic seawater during precipitation of Lake Almar anhydrite that immediately overlies the Yeoman Formation. However, the low δ18O values of dolomited burrow infills (-5.9‰~ -7.8‰, PDB) and matrix dolomites (-6.6‰~ -8.1‰, avg. -7.4‰ PDB) compared to the estimated values for the late Ordovician marine dolomite could be attributed to modification and alteration of dolomite at higher temperatures during deeper burial, which could also be responsible for its 87Sr/86Sr ratios (0.7084~0.7088) that are higher than suggested for the late Ordovician seawaters (0.7078~0.7080). The trace amounts of saddle dolomite cement in the Red River carbonates are probably related to "cannibalization" of earlier replacement dolomite during the chemical compaction.  相似文献

以AC-13级配为基础,将橡胶颗粒代替部分集料掺入混合料中,以低温弯曲试验为评价方法对不同橡胶颗粒掺量下沥青混合料的低温抗裂性进行研究,并引入应变能密度值对混合料的低温抗裂性进行综合评价.试验结果表明:橡胶颗粒沥青混合料试件的破坏微应变均超过2 300,满足冬寒区的技术指标;无论是否掺加橡胶颗粒,随着温度的下降,沥青混合料破坏时的最大弯拉强度增大,弯拉应变降低,劲度模量增大;弯曲应变能密度在胶粒掺量为1%左右时具有较大的弯曲应变能密度值,此时橡胶颗粒沥青混合料具有较好的低温抗裂性.  相似文献

理论推导与室内实验相结合,建立了低渗透非均质砂岩油藏启动压力梯度确定方法。首先借助油藏流场与电场相似的原理,推导了非均质砂岩油藏启动压力梯度计算公式。其次基于稳定流实验方法,建立了非均质砂岩油藏启动压力梯度测试方法。结果表明:低渗透非均质砂岩油藏的启动压力梯度确定遵循两个等效原则。平面非均质油藏的启动压力梯度等于各级渗透率段的启动压力梯度关于长度的加权平均;纵向非均质油藏的启动压力梯度等于各渗透率层的启动压力梯度关于渗透率与渗流面积乘积的加权平均。研究成果可用于有效指导低渗透非均质砂岩油藏的合理井距确定,促进该类油藏的高效开发。  相似文献

Quality traits in wheat (Triticum aestirum L.) were studied by quantitative trait locus (QTL) analysis in a recombinant inbred line (RIL) population, a set of 131 lines derived from Chuan 35050 × Shannong 483 cross (ChSh). Grains from RILs were assayed for 21 quality traits related to protein and starch. A total of 35 putative QTLs for 19 traits with a single QTL explaining 7.99-40.52% of phenotypic variations were detected on 10 chromosomes, 1D, 2A, 2D, 3B, 3D, 5A, 6A, 6B, 6D, and 7B. The additive effects of 30 QTLs were positive, contributed by Chuan 35050, the remaining 5 QTLs were negative with the additive effect contributed by Shannong 483. For protein traits, 15 QTLs were obtained and most of them were located on chromosomes 1 D, 3B and 6D, while 20 QTLs for starch traits were detected and most of them were located on chromosomes 3D, 6B and 7B. Only 7 QTLs for protein and starch traits were co-located in three regions on chromosomes 1D, 2A and 2D. These protein and starch trait QTLs showed a distinct distribution pattern in certain regions and chromosomes. Twenty-two QTLs were clustered in 6 regions of 5 chromosomes. Two QTL clusters for protein traits were located on chromosomes 1D and 3B, respectively, three clusters for starch traits on chromosomes 3D, 6B and 7B, and one cluster including protein and starch traits on chromosome 1D.  相似文献
