四株微生物絮凝剂产生菌生长条件的研究   总被引:8,自引:0,他引:8  
采用正交实验研究了淀粉、酵母膏、pH值3种因素对菌株HHE-P7、HHE-A8、HHE-P21、HHE-A26产絮凝剂的影响,得出了各菌株培养的优化比,并进一步通过实验证实在该优化配比的条件下,各菌株所产絮凝剂的絮凝效果都很高,对高岭土悬浮液的絮凝率达到95%以上。  相似文献

从土壤中筛选出8株具有絮凝活性的菌株,经复筛后得到1株絮凝活性较高的微生物絮凝剂产生菌C1,并以此为研究对象,分别考察了培养基中碳源、氮源、无机盐以及培养基初始pH值和培养温度等因素对絮凝活性的影响.研究表明,C1菌株在以葡萄糖为碳源、牛肉膏为氮源、初始pH6.0、培养温度30℃、培养时间为72h的条件下,所产絮凝剂对4%高岭土悬浊液的絮凝效率可达90.4%,显示出良好的应用前景.  相似文献

采用原生质体融合-紫外诱变技术分离得到高效絮凝剂产生菌Enterobacter sp. W16-c,并对其产絮特性和影响因素进行分析。结果表明:Enterobacter sp. W16-c的絮凝能力比出发菌提高13.91%;突变菌株发酵产絮最适碳源为蔗糖,最适氮源为酵母膏,最佳初始发酵pH值为5~9;金属离子Fe2+、Na+对Enterobacter sp. W16-c的产絮效果有促进作用;在最佳发酵条件下所产絮凝剂的絮凝率可达96.2%;Enterobacter sp. W16-c的主要成分为多糖;Enterobacter sp. W16-c是一株产絮性能稳定的高效产絮突变菌株。  相似文献

从活性污泥中筛选出1株高效微生物絮凝剂产生菌BJ-I,其絮凝率达87.6%.通过生理生化分析及16S rDNA序列鉴定该菌株为短芽孢杆菌(Bacillus brevis).该菌产絮凝剂的最适碳源和氮源分别为20 g/L的葡萄糖和2.0 g/L的蛋白胨;适宜的初始pH和培养温度分别为7.0和30℃;在培养基中添加0.5 g/L的Mg2+有利于菌体生长,但对絮凝荆合成无影响.对该絮凝剂的特性研究结果表明:菌液投放量3%、pH值在5~9、温度为60 ℃条件下絮凝效果最好,加入浓度为8%的Ca2+可显著提高其絮凝活性.实验证明,该菌起絮凝作用的物质存在于发酵液中,主要成分为杂多糖.  相似文献

一株产絮凝剂硅酸盐细菌的筛选及其絮凝特性   总被引:3,自引:0,他引:3  
从土壤中筛选到一株产絮凝剂的硅酸盐菌株B12.对其絮凝剂产生条件进行优化,并考查其絮凝特性.以质量浓度为8 g/L的葡萄糖和0.15 g/L的(NH4)2SO4作为碳源和氮源,调节初始pH值至6.9,在温度为31 ℃、转速为150 r/min的摇床中培养72 h后,菌株的絮凝活性最高,絮凝率可达93.6%.絮凝剂特性研究表明:供氧可提高细菌的生长量,但不利于絮凝剂的生成.分段培养有利于提高絮凝率.该菌起絮凝作用的主要是其胞外代谢物,其适用pH值范围广,为0.5~12,絮凝率大于90%;B12菌生物絮凝剂稳定性好,与其他无机絮凝剂相比,其絮凝活效果好、无毒、无二次污染,在矿浆固液分离及矿物废水处理方面有着广阔的应用前景.  相似文献

通过多次采样,反复分离,从油田采出水和回注污水中分离得到16株产絮凝剂菌株,经复筛,得到1株具有较高絮凝活性的细菌。细菌生长过程均可产生具有絮凝作用的胞外分泌物,经实验对油田采出水具有高絮凝作用。分别研究了碳源、氮源、无机盐、初始pH值、培养温度等对其产生絮凝剂的影响。结果表明,葡萄糖、玉米淀粉是较好的碳源,蛋白胨为最佳氮源,最适宜培养温度为320C,初始pH为7.5~9,摇床转速为150 r/m in。  相似文献

从空气和活性污泥中筛选出絮凝剂产生菌,并对其絮凝条件进行优化研究。实验共分离纯化出纯种菌株19株,经絮凝试验筛选得到有絮凝能力的菌株9株,复筛得到3株絮凝活性较高且稳定的菌株,分别命名为M-2、M-3、G-3。以M-3为例,研究了碳源、氮源、培养基初始pH值、培养时间、培养温度及通气量等对菌株絮凝性能的影响,结果表明玉米面、黄豆面分别为最佳碳源和氮源,M-3的最佳培养条件为:温度30℃,培养基初始pH值为8.5,培养时间为36或60小时,摇床转速为160r/min。  相似文献

从活性污泥中初筛分离出21株产絮凝剂的菌株,复筛得到一株具有较高,较快絮凝活性的微生物产生菌,将其命名为WZU-1。通过培养条件优化,考察了各种因素如碳源、氮源、pH、温度、培养时间、摇床转速对絮凝效果的影响。实验结果表明,WZU-1对高岭土悬浊液的最佳絮凝条件为:碳源为葡萄糖、氮源为尿素、pH为8.0、温度为38℃、摇床转速为200r/min、培养时间为36h,在最佳条件下对4‰高岭土悬浊液的絮凝率达到95.16%。同时对该菌株絮凝性能进行了研究,结果表明该菌絮凝活性物质分布在发酵液离心后的上清液中,为该菌代谢产物;并具有较好的热稳定性。  相似文献

The Williston Basin is a significant petroleum province, containing oil production zones that include the Middle Cambrian to Lower Ordovician, Upper Ordovician, Middle Devonian, Upper Devonian and Mississippian and within the Jurassic and Cretaceous. The oils of the Williston Basin exhibit a wide range of geochemical characteristics defined as "oil families", although the geochemical signature of the Cambrian Deadwood Formation and Lower Ordovician Winnipeg reservoired oils does not match any "oil family". Despite their close stratigraphic proximity, it is evident that the oils of the Lower Palaeozoic within the Williston Basin are distinct. This suggests the presence of a new "oil family" within the Williston Basin. Diagnostic geochemical signatures occur in the gasoline range chromatograms, within saturate fraction gas chromatograms and biomarker fingerprints. However, some of the established criteria and cross-plots that are currently used to segregate oils into distinct genetic families within the basin do not always meet with success, particularly when applied to the Lower Palaeozoic oils of the Deadwood and Winnipeg Formation.  相似文献

The discovery of the prolific Ordovician Red River reservoirs in 1995 in southeastern Saskatchewan was the catalyst for extensive exploration activity which resulted in the discovery of more than 15 new Red River pools. The best yields of Red River production to date have been from dolomite reservoirs. Understanding the processes of dolomitization is, therefore, crucial for the prediction of the connectivity, spatial distribution and heterogeneity of dolomite reservoirs.The Red River reservoirs in the Midale area consist of 3~4 thin dolomitized zones, with a total thickness of about 20 m, which occur at the top of the Yeoman Formation. Two types of replacement dolomite were recognized in the Red River reservoir: dolomitized burrow infills and dolomitized host matrix. The spatial distribution of dolomite suggests that burrowing organisms played an important role in facilitating the fluid flow in the backfilled sediments. This resulted in penecontemporaneous dolomitization of burrow infills by normal seawater. The dolomite in the host matrix is interpreted as having occurred at shallow burial by evaporitic seawater during precipitation of Lake Almar anhydrite that immediately overlies the Yeoman Formation. However, the low δ18O values of dolomited burrow infills (-5.9‰~ -7.8‰, PDB) and matrix dolomites (-6.6‰~ -8.1‰, avg. -7.4‰ PDB) compared to the estimated values for the late Ordovician marine dolomite could be attributed to modification and alteration of dolomite at higher temperatures during deeper burial, which could also be responsible for its 87Sr/86Sr ratios (0.7084~0.7088) that are higher than suggested for the late Ordovician seawaters (0.7078~0.7080). The trace amounts of saddle dolomite cement in the Red River carbonates are probably related to "cannibalization" of earlier replacement dolomite during the chemical compaction.  相似文献

From the discussion of the features of women's language, more attention should be paid to language use in social context. 以AC-13级配为基础,将橡胶颗粒代替部分集料掺入混合料中,以低温弯曲试验为评价方法对不同橡胶颗粒掺量下沥青混合料的低温抗裂性进行研究,并引入应变能密度值对混合料的低温抗裂性进行综合评价.试验结果表明:橡胶颗粒沥青混合料试件的破坏微应变均超过2 300,满足冬寒区的技术指标;无论是否掺加橡胶颗粒,随着温度的下降,沥青混合料破坏时的最大弯拉强度增大,弯拉应变降低,劲度模量增大;弯曲应变能密度在胶粒掺量为1%左右时具有较大的弯曲应变能密度值,此时橡胶颗粒沥青混合料具有较好的低温抗裂性.  相似文献

理论推导与室内实验相结合,建立了低渗透非均质砂岩油藏启动压力梯度确定方法。首先借助油藏流场与电场相似的原理,推导了非均质砂岩油藏启动压力梯度计算公式。其次基于稳定流实验方法,建立了非均质砂岩油藏启动压力梯度测试方法。结果表明:低渗透非均质砂岩油藏的启动压力梯度确定遵循两个等效原则。平面非均质油藏的启动压力梯度等于各级渗透率段的启动压力梯度关于长度的加权平均;纵向非均质油藏的启动压力梯度等于各渗透率层的启动压力梯度关于渗透率与渗流面积乘积的加权平均。研究成果可用于有效指导低渗透非均质砂岩油藏的合理井距确定,促进该类油藏的高效开发。  相似文献
