天然药物川芎嗪对脂质过氧化内皮细胞的保护作用研究   总被引:2,自引:0,他引:2  
研究天然药物川芎嗪对内皮细胞氧化损伤时细胞存活率以及MCP-1、ICAM-1mRNA表达水平的影响,探讨其抗动脉粥样硬化的可能机制。用血管内皮细胞与不同浓度川芎嗪孵育,给予氧化低密度脂蛋白损伤,通过MTr和RT—PCR技术,检测内皮细胞存活率和MCP-1、ICAM-1mRNA表达水平的变化。结果显示内皮细胞与川芎嗪预孵育后,其细胞存活率明显增加,MCP-1和ICAM-1mRNA的表达水平明显下调。说明氧化低密度脂蛋白可诱导内皮细胞存活率下降,MCP-1和ICAM-1mRNA表达水平增加,川芎嗪可以减轻其过量表达,保护内皮细胞。  相似文献

研究红景天苷(SAL)对同型半胱氨酸(Hcy)诱导体外培养的人脐静脉内皮细胞(HUVECs)氧化应激损伤的保护作用及可能机制.选择3-8代生长状态良好的HUVECs用于实验,采用SAL预孵育HUVECs 2h后,再加入Hcy(1 mmol/L)共孵育12 h诱导内皮细胞氧化应激损伤.采用MTT和LDH法分析细胞损伤,荧光探针法DCFH-DA分析细胞内活性氧(ROS)水平,酶联免疫吸附法分析细胞内丙二醛(MDA),超氧化物歧化酶(SOD)的含量,qRT-PCR技术检测Nrf2、HO-1、NQO1的基因表达.结果显示,Hcy显著引起血管内皮细胞损伤,而10、50μmol/L的红景天苷干预给药后,能够提高Hcy诱导的细胞存活率,明显降低LDH、MDA的水平,增加胞内SOD活性.与Hcy处理组相比,SAL干预给药能够降低Hcy所致细胞内ROS的增高,上调Nrf2、抗氧化酶基因HO-1、醌氧化还原酶NQO1的mRNA表达.本研究表明,红景天苷有效抑制Hcy诱导的HUVECs氧化应激损伤,其作用与红景天苷增强细胞清除自由基能力,调控Nrf2信号有关.  相似文献

目的:观察ERK5抑制剂XMD8-92对M1型巨噬细胞标志性炎症因子单核细胞趋化因子(MCP-1)、诱导型一氧化氮合酶(iNOS)表达的影响.方法:常规培养巨噬细胞RAW264.7,氧化低密度脂蛋白(质量浓度为10μg/mL)和脂多糖(质量浓度为1 ng/mL)建立细胞模型,XMD8-92(浓度为2.5μmol/mL)抑制ERK5表达.实验分为空白、模型和模型+抑制剂组,分别应用RT-qPCR法和蛋白印迹法检测各组细胞MCP-1、iNOS mRNA和蛋白的表达;CCK-8法检测细胞生存率.结果:与正常组相比,模型组MCP-1和iNOS mRNA和蛋白的表达显著升高,有统计学差异(P0.01);与模型组比较,模型+抑制剂组MCP-1和iNOS mRNA和蛋白的表达降低,具有显著统计学差异(P0.01);与空白组相比较,模型组细胞生存率升高,加入XMD8-92后细胞生存率降低,有统计学差异(P0.01).结论:XMD8-92对M1型巨噬细胞的活性有抑制作用,并可抑制MCP-1和iNOS蛋白和mRNA的表达.可能通过抑制ERK5活化,抑制M1型巨噬细胞标志性炎症因子表达,减轻炎症反应.  相似文献

目的:观察瓜蒌薤白半夏汤(GXBD)对Apo-E-/-小鼠动脉粥样硬化模型(AS)C反应蛋白(CRP)、白介素-6(IL-6)、肿瘤坏死因子(TNF-α)、细胞间黏附分子-1(ICAM-1)和血管细胞黏附分子-1(VCAM-1)表达的影响,并探讨其可能的机制.方法:以高脂饲料饲喂雄性Apo-E-/-小鼠建立AS模型,将AS模型小鼠随机分为模型对照组(MS)、辛伐他汀组(XFTT)、GXBD高、低剂量组,选取C57BL/6J雄性小鼠作为正常对照组,每组10只,连续灌胃(ig)给药8周.末次给药24h后,检测血清总胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白(LDL-C)、高密度脂蛋白(HDL-C)、及血清CRP、IL-6、TNF-α水平,HE染色观察主动脉组织病理学变化;Western-Blot法检测主动脉VCAM-1和ICAM-1蛋白表达水平.结果:模型组小鼠TC、TG、LDL-C、HDL-C、及血清CRP、IL-6、TNF-α显著高于正常对照组(P0.05),主动脉VCAM-1、ICAM-1蛋白表达显著高于正常对照组(P0.05),模型组小鼠主动脉出现明显粥样硬化斑块;GXBD各组和辛伐他汀组小鼠血脂及血清CRP、IL-6、TNF-α水平显著低于模型组(P0.05),主动脉组织VCAM-1、ICAM-1蛋白表达水平显著低于模型组(P0.05),且主动脉组织粥样硬化病变明显减轻.结论:GXBD对Apo-E-/-小鼠AS病变有较好的治疗作用,其机制与其调节血脂代谢、抑制炎症因子及主动脉组织VCAM-1、ICAM-1蛋白的表达相关.  相似文献

目的:研究氨磷汀对急性放射性肠炎小鼠小肠组织中核转录因子kappaB(NF-κB)通路和血管间粘附分子-1(VCAM-1)、细胞间粘附分子-1(ICAM-1)基因表达的影响和意义.方法:将小鼠分成对照组、实验组、氨磷汀腹腔注射干预组,采用6Gy 60Coγ-射线全身辐射,于第1、7、14天分批处死小鼠观察病理改变.实时荧光定量PCR(Q-PCR)方法检测不同时间点对照组、实验组和氨磷汀干预组NF-κB、VCAM-1、ICAM-1基因mRNA表达水平的变化.结果:辐射后实验组、干预组均可见明显黏膜下层微血管损伤,干预组损伤较实验组轻.辐射后第1天实验组、干预组小鼠小肠组织中NF-κB、VCAM-1、ICAM-1基因mRNA表达水平均显著升高(P0.05),干预组升高水平显著低于实验组(P0.05);第7天实验组各基因mRNA水平均呈下降趋势但仍高于对照组(P0.05),干预组达到或接近对照组水平;第14天实验组、干预组各基因mRNA表达水平与对照组比较无统计学差异(P0.05).结论:急性放射性肠炎早期即发生NF-κB、VCAM-1、ICAM-1基因mRNA的表达上调,可能在辐射后微血管损伤中发挥作用,氨磷汀可能通过下调上述基因表达对微血管起到保护作用.  相似文献

目的探讨依折麦布对高迁移率族蛋白1(HMGB1)诱导血管内皮细胞激活影响的分子机制.方法分离、培养雄性SD大鼠胸主动脉内皮细胞,分为空白对照组、HMGB1刺激组、依折麦布组(HMGB1刺激前10μmol/L依折麦布预处理)、CLI-095组(HMGB1刺激前1μmol/L CLI-095预处理).荧光定量分析中性粒细胞与内皮细胞的黏附能力;RT-PCR和Western blot检测内皮细胞中TLR4,ICAM-1、可溶性E选择素mRNA和蛋白表达水平;EMSA法检测NF-κb-p65的DNA结合活性.结果 HMGB1活化的内皮细胞与中性粒细胞黏附活力均明显高于空白对照组,差异具有统计学意义(P0.05);依折麦布组、CLI-095组内皮细胞与中性粒细胞黏附活力均明显低于HMGB1诱导组,差异具有统计学意义(P0.05).HMGB1刺激组的内皮细胞TLR4,mRNA和蛋白表达量明显高于空白对照组,差异具有统计学意义(P0.05);依折麦布组和CLI-095组mRNA和蛋白表达量均明显低于HMGB1组,差异具有统计学意义(P0.05).HMGB1组内皮细胞NF-κb p65亚单位核位移明显强于空白对照组,差异具有统计学意义(P0.05);依折麦布组和CLI-095组较HMGB1组核位移明显减弱,差异具有统计学意义(P0.05).HMGB1组ICAM-1、可溶性E选择素表达水平明显高于空白对照组,差异具有统计学意义(P0.05).而经依折麦布、CLI-095干预后可明显降低ICAM-1、可溶性E选择素表达水平,与HMGB1组之间差异具有统计学意义(P0.05).结论依折麦布可通过调节黏附分子(ICAM-1、可溶性E选择素)的表达而有效抑制HMGB1诱导的血管内皮细胞活化效应,其机制与抑制TLR4的表达和NF-κb激活有关.  相似文献

细胞间黏附分子-1(ICAM-1)是白细胞和角质细胞之间相互作用的必需因子.角质细胞中ICAM-1表达的上调与多种皮肤炎症相关.中国马尾松树皮提取物(PMBE)是具有抗氧化活性的类黄酮化合物,本文研究了PMBE对炎症中起重要作用的诱导型ICAM-1表达的影响.用1000 U/mL的IFNγ处理人的角质细胞系HaCaT细胞24 h可以明显地诱导ICAM-1的表达.PMBE(40×10-6g/mL,24 h)预处理可以抑制IFN-γ诱导的ICAM-1 mRNA和蛋白的表达.由此可以提示PMBE在转录和翻译水平上对诱导型ICAM-1的表达均有抑制作用,并且对ICAM-蛋白表达的抑制可能是通过对ICAM-1 mRNA的抑制而实现的.这些结果提示PMBE具有治疗皮肤炎症的潜在功效.  相似文献

为探讨黄芪总皂苷(ASTs)对三氧化二砷(ATO)诱导H9c2细胞损伤的保护作用及其机制,采用6μg·mL-1ATO孵育12 h建立H9c2细胞损伤模型.实验设空白组、模型组、ASTs低/中/高浓度组,ASTs组预孵育12 h,弃去含药培养基,继续用6μg·mL-1 ATO孵育12 h,终止实验.通过CCK-8法检测细胞存活率,Hoechst 33342染色法观察细胞凋亡程度,流式细胞术定量测定细胞凋亡率,RT-qPCR测定Bcl-2、Bax、Caspase-3、Caspase-9表达水平,Western Blot测其蛋白.结果表明:ATO能够显著降低心肌细胞存活率、增加凋亡率(P<0.05);与ATO模型组比较,ASTs各组能显著提高细胞存活率,降低细胞凋亡率(P<0.05),降低Caspase-3、Caspase-9、Bax mRNA表达水平,增加Bcl-2 mRNA表达水平(P<0.05),显著降低细胞中Caspase-3、Caspase-9蛋白表达水平(P<0.05),升高Bcl-2/Bax比值(P<0....  相似文献

 通过利用复合因素制备兔动脉粥样硬化(AS)易损斑块模型,研究四妙勇安汤对AS易损斑块的影响及作用机制。随机将40只日本大耳白兔分为正常组10只,实验组30只。正常组予普通饲料,实验组利用复合因素制备兔主AS易损斑块模型,8周时实验组随机分为模型组、辛伐组、四妙组,继续予以高脂饮食;各给药组开始给药至24周取材,观察主动脉的病理形态,检测血清MCP-1、ICAM-1、CRP含量和斑块内HSP60、TNF-α、NF-κB表达。结果显示,模型组内膜明显增厚、纤维帽较薄,斑块内有大量脂质沉积和炎症细胞浸润,血清MCP-1、ICAM-1、CRP随时间延长呈上升趋势,主动脉斑块TNF-α、HSP60、NF-κB表达增加。在给药后各时间点四妙组血清MCP-1、ICAM-1水平均低于模型组(P<0.05或P<0.01),对CRP水平影响不明显。斑块内TNF-α、HSP60、NF-κB阳性面积百分比均低于模型组(P<0.01),ICAM-1 MRNA表达低于模型组(P<0.01)。由此得出,四妙勇安汤能够稳定AS易损斑块,其作用可能是通过抑制炎症反应实现的。  相似文献

为探讨血府逐瘀汤对动脉粥样硬化(atherosclerosis,AS)大鼠主动脉壁细胞间黏附分子-1(ICAM-1)表达水平和脂质过氧化物的影响,将40只大鼠随机分为正常对照组、模型组、血脂康组、血府逐瘀汤高剂量组、血府逐瘀汤低剂量组,对照组饲喂正常饲料,模型组给予高脂饲料,治疗组在给予高脂饲料的同时给予治疗药物,12周后免疫组化法观察主动脉壁ICAM-1的表达水平、检测血清超氧化物歧化酶(SOD)、丙二醛(MDA)的含量.结果显示:模型组ICAM-1的表达水平呈强阳性表达,治疗组各组均呈弱阳性表达,各组间均有统计学意义(P<0.05).治疗组SOD水平较模型组显著升高,MDA含量显著降低,各组间均有统计学意义(P<0.05).这说明血府逐瘀汤可以通过下调ICAM-1的表达,清除脂质过氧化物、保护内皮细胞起到抗AS的作用.  相似文献

The Williston Basin is a significant petroleum province, containing oil production zones that include the Middle Cambrian to Lower Ordovician, Upper Ordovician, Middle Devonian, Upper Devonian and Mississippian and within the Jurassic and Cretaceous. The oils of the Williston Basin exhibit a wide range of geochemical characteristics defined as "oil families", although the geochemical signature of the Cambrian Deadwood Formation and Lower Ordovician Winnipeg reservoired oils does not match any "oil family". Despite their close stratigraphic proximity, it is evident that the oils of the Lower Palaeozoic within the Williston Basin are distinct. This suggests the presence of a new "oil family" within the Williston Basin. Diagnostic geochemical signatures occur in the gasoline range chromatograms, within saturate fraction gas chromatograms and biomarker fingerprints. However, some of the established criteria and cross-plots that are currently used to segregate oils into distinct genetic families within the basin do not always meet with success, particularly when applied to the Lower Palaeozoic oils of the Deadwood and Winnipeg Formation.  相似文献

The discovery of the prolific Ordovician Red River reservoirs in 1995 in southeastern Saskatchewan was the catalyst for extensive exploration activity which resulted in the discovery of more than 15 new Red River pools. The best yields of Red River production to date have been from dolomite reservoirs. Understanding the processes of dolomitization is, therefore, crucial for the prediction of the connectivity, spatial distribution and heterogeneity of dolomite reservoirs.The Red River reservoirs in the Midale area consist of 3~4 thin dolomitized zones, with a total thickness of about 20 m, which occur at the top of the Yeoman Formation. Two types of replacement dolomite were recognized in the Red River reservoir: dolomitized burrow infills and dolomitized host matrix. The spatial distribution of dolomite suggests that burrowing organisms played an important role in facilitating the fluid flow in the backfilled sediments. This resulted in penecontemporaneous dolomitization of burrow infills by normal seawater. The dolomite in the host matrix is interpreted as having occurred at shallow burial by evaporitic seawater during precipitation of Lake Almar anhydrite that immediately overlies the Yeoman Formation. However, the low δ18O values of dolomited burrow infills (-5.9‰~ -7.8‰, PDB) and matrix dolomites (-6.6‰~ -8.1‰, avg. -7.4‰ PDB) compared to the estimated values for the late Ordovician marine dolomite could be attributed to modification and alteration of dolomite at higher temperatures during deeper burial, which could also be responsible for its 87Sr/86Sr ratios (0.7084~0.7088) that are higher than suggested for the late Ordovician seawaters (0.7078~0.7080). The trace amounts of saddle dolomite cement in the Red River carbonates are probably related to "cannibalization" of earlier replacement dolomite during the chemical compaction.  相似文献

以AC-13级配为基础,将橡胶颗粒代替部分集料掺入混合料中,以低温弯曲试验为评价方法对不同橡胶颗粒掺量下沥青混合料的低温抗裂性进行研究,并引入应变能密度值对混合料的低温抗裂性进行综合评价.试验结果表明:橡胶颗粒沥青混合料试件的破坏微应变均超过2 300,满足冬寒区的技术指标;无论是否掺加橡胶颗粒,随着温度的下降,沥青混合料破坏时的最大弯拉强度增大,弯拉应变降低,劲度模量增大;弯曲应变能密度在胶粒掺量为1%左右时具有较大的弯曲应变能密度值,此时橡胶颗粒沥青混合料具有较好的低温抗裂性.  相似文献

理论推导与室内实验相结合,建立了低渗透非均质砂岩油藏启动压力梯度确定方法。首先借助油藏流场与电场相似的原理,推导了非均质砂岩油藏启动压力梯度计算公式。其次基于稳定流实验方法,建立了非均质砂岩油藏启动压力梯度测试方法。结果表明:低渗透非均质砂岩油藏的启动压力梯度确定遵循两个等效原则。平面非均质油藏的启动压力梯度等于各级渗透率段的启动压力梯度关于长度的加权平均;纵向非均质油藏的启动压力梯度等于各渗透率层的启动压力梯度关于渗透率与渗流面积乘积的加权平均。研究成果可用于有效指导低渗透非均质砂岩油藏的合理井距确定,促进该类油藏的高效开发。  相似文献

Quality traits in wheat (Triticum aestirum L.) were studied by quantitative trait locus (QTL) analysis in a recombinant inbred line (RIL) population, a set of 131 lines derived from Chuan 35050 × Shannong 483 cross (ChSh). Grains from RILs were assayed for 21 quality traits related to protein and starch. A total of 35 putative QTLs for 19 traits with a single QTL explaining 7.99-40.52% of phenotypic variations were detected on 10 chromosomes, 1D, 2A, 2D, 3B, 3D, 5A, 6A, 6B, 6D, and 7B. The additive effects of 30 QTLs were positive, contributed by Chuan 35050, the remaining 5 QTLs were negative with the additive effect contributed by Shannong 483. For protein traits, 15 QTLs were obtained and most of them were located on chromosomes 1 D, 3B and 6D, while 20 QTLs for starch traits were detected and most of them were located on chromosomes 3D, 6B and 7B. Only 7 QTLs for protein and starch traits were co-located in three regions on chromosomes 1D, 2A and 2D. These protein and starch trait QTLs showed a distinct distribution pattern in certain regions and chromosomes. Twenty-two QTLs were clustered in 6 regions of 5 chromosomes. Two QTL clusters for protein traits were located on chromosomes 1D and 3B, respectively, three clusters for starch traits on chromosomes 3D, 6B and 7B, and one cluster including protein and starch traits on chromosome 1D.  相似文献
