血府逐瘀汤对动脉粥样硬化大鼠细胞间黏附分子和脂质过氧化物的影响

摘    要:为探讨血府逐瘀汤对动脉粥样硬化(atherosclerosis,AS)大鼠主动脉壁细胞间黏附分子-1(ICAM-1)表达水平和脂质过氧化物的影响,将40只大鼠随机分为正常对照组、模型组、血脂康组、血府逐瘀汤高剂量组、血府逐瘀汤低剂量组,对照组饲喂正常饲料,模型组给予高脂饲料,治疗组在给予高脂饲料的同时给予治疗药物,12周后免疫组化法观察主动脉壁ICAM-1的表达水平、检测血清超氧化物歧化酶(SOD)、丙二醛(MDA)的含量.结果显示:模型组ICAM-1的表达水平呈强阳性表达,治疗组各组均呈弱阳性表达,各组间均有统计学意义(P<0.05).治疗组SOD水平较模型组显著升高,MDA含量显著降低,各组间均有统计学意义(P<0.05).这说明血府逐瘀汤可以通过下调ICAM-1的表达,清除脂质过氧化物、保护内皮细胞起到抗AS的作用.

Abstract:To investigate the effects of Xuefu Zhuyu decoction on intercellular adhesion molecule and lipid peroxidation of atherosclerosis rats.40 SD rats were randomly divided into control group,model group and Xuefu Zhuyu high-dose group,Xuefu Zhuyu low dose group and Xuezhikang group.The control group was fed with normal diet,model group was fed with high fat diet,the treatment group was fed with high fat diet and given treatment drugs.12 weeks later,the expression of ICAM-1 proteinum in the endarterium of AS rats was detected by immunohistochemical technique,SOD and MDA of serum were tested.ICAM-1 of model group expression levels showed a strong positive expression,the treatment group showed a weak positive expression(P<0.05),compared with model group,the serum contents of SOD in test group were increased,while MDA were decreased significantly(P<0.05).Experiment showed Xuefu Zhuyu decoction could decrease the regulation of ICAM-1 expression,remove lipid peroxides,protect endothelial cells,prevent and treat atherosclerosis.
