Objective: To determine the effects of albumin administration on lung injury and apoptosis in traumatic/hemorrhagic shock (T/HS) rats. Methods: Studies were performed on an in vivo model of spontaneously breathing rats with induced T/HS; the rats were subjected to femur fracture, ischemia for 30 min, and reperfusion for 20 min with Ringer's lactate solution (RS) or 5% (w/v) albumin (ALB), and the left lower lobes of the lungs were resected. Results: Albumin administered during reperfusion markedly attenuated injury of the lung and decreased the concentration of lactic acid and the number of in situ TdT-mediated dUTP nick-end labelling (TUNEL)-positive cells. Moreover, immunohistochemistry performed 24 h after reperfusion revealed increases in the level of nuclear factor κB (NF-κB), and phosphorylated p38 mitogen-activated protein kinase (MAPK) in the albumin-untreated group was down-regulated by albumin treatment when compared with the sham rats. Conclusion: Resuscitation with albumin attenuates tissue injury and inhibits T/HS-induced apoptosis in the lung via the p38 MAPK signal transduction pathway that functions to stimulate the activation of NF-κB.  相似文献

Objective: To observe the effect of angiotensin II (Ang II) on expression of gap junction channel protein connexin 43 (Cx43) in the proliferation process of vascular smooth muscle cells (VSMCs) during the early stage of arteriosclerosis. Methods: Thirty-two adult male rabbits were randomly divided into 4 groups. Rabbits in Group A were fed common diet while others in Groups B, C, and D were fed high-cholesterol diet. Losartan (10 mg/(kg·d)) and ramipril (0.5 mg/(kg·d)) were added in the diet of Groups C and D, respectively. The animals were sacrificed after 8 weeks and abdominal aortas were removed and dissected. The expression of Cx43 was assayed using RT-PCR and Western Blotting analysis. Results: Cx43 was increased markedly in both protein and mRNA level in Groups B, C, and D fed high-cholesterol diet compared with that in control group (P<0.01). Cx43 level in losartan or ramipril treated groups was higher than that in control group (P<0.01, P<0.05), but lower than that in high-cholesterol diet groups (P<0.05, P<0.01). Conclusion: Cx43 level was upregulated in VSMCs during early atherosclerosis. Losartan and ramipril can inhibit the expression of Cx43.  相似文献

为探讨缝隙连接是否参与抑制大鼠血管平滑肌细胞的增殖及可能的分子机制,本研究以原代及传代培养大鼠胸主动脉平滑肌细胞为模型,实验分2组:对照组、缝隙连接阻断剂18α-甘草次酸(18α-—Glycyrrhetinicacid,18Q—GA)组。MTT法及流式细胞术检测细胞的增殖活性,染料示踪分子传递法(划痕标记染料传输法)检测细胞的缝隙连接功能,Westernblotting法检测细胞中缝隙连接蛋白40(Cx40)和43(Cx43)表达。结果显示:(1)与对照组相比,18α-GA组MTT法测得的氏,。值及细胞周期S期比例均降低(P〈0.01),细胞增殖活性减弱;(2)与对照组相比,18α-GA组罗氏黄荧光染料传递百分数显著降低,缝隙连接功能明显减弱(P〈0.01);(3)与对照组相比,18rGA组Cx40和总Cx43蛋白表达无显著差异(P〉0.05),磷酸化Cx43与非磷酸化Cx43的比值显著降低(P〈0.01)。由此可知,18α-GA可能主要通过下调血管平滑肌细胞磷酸化Cx43蛋白表达,引起缝隙连接通讯功能减弱,从而抑制了血管平滑肌细胞的增殖。  相似文献

以人正常胃黏膜上皮细胞系GES-1和不同恶化程度的人胃癌细胞系(低恶化AGS细胞系、中恶化SGC-7901细胞系、高恶化BGC-823细胞系)为研究对象,采用激光共聚焦显微镜的荧光漂白恢复技术及划痕标记荧光染料示踪技术研究不同恶化程度的胃癌细胞系细胞间隙连接通讯(Gap junctional intercellular communication,GJIC)功能的差异;采用荧光定量PCR技术和间接免疫荧光技术检测间隙连接蛋白(Connexin43,Cx43)基因在mRNA和蛋白水平的表达差异,探求其与胃癌恶性程度的相关性.结果表明,在正常胃细胞系和低、中及高恶化胃腺癌细胞系中,细胞间的GJIC功能随着恶性程度的升高而减弱(AGS、SGC-7901)或消失(BGC-823),且Cx43蛋白及mRNA的表达量随着恶性程度的升高而下调(AGS、SGC-7901)或缺失(BGC-823).这提示胃癌恶性程度与胃癌细胞的GJIC功能显著相关(P<0.05),GJIC在胃癌发生中起重要作用.  相似文献

目的探讨精索静脉曲张(VC)对青春期大鼠睾丸生精细胞Cx43的影响.方法部分结扎大鼠左肾静脉,建立精索静脉曲张模型.取雄性SD大鼠30只,随机分为2组,每组15只,分为对照组、精索静脉曲张组(手术组),每组中再分为3个亚组,每组5只,结扎持续时间为2,4,8周;达到相应的时间段后取各组大鼠左侧的睾丸,运用RT-PCR法检测Cx43及HIF-1α基因的表达,并在8周时通过Western-blot法检测Cx43及bcl-2蛋白表达水平.结果对照组在2,4,6周时Cx43及HIF-1α基因表达无明显变化;手术组在2,4,6周时HIF-1α基因表达水平提高,Cx43基因表达水平下降,与对照组相比,差异具有统计学意义(P0.05);蛋白检测结果表明:8周时手术组Cx43及bcl-2蛋白表达水平显著下降.结论精索静脉曲张可致大鼠睾丸组织缺氧,Cx43及bcl-2表达下调,并诱导睾丸组织生精细胞凋亡增加,进而影响睾丸生精功能.  相似文献

目的观察用油酸(OA)和脂多糖(LPS)两次打击急性肺损伤大鼠肺组织核因子———kB(NF_kB)p65 mRNA表达量的变化,探讨转录调节因子NF_kB p65与急性肺损伤的关系。方法用OA(0.2mL/kg)和LPS(2mg/kg)两次打击,建立两次打击急性肺损伤(ALI)模型,进行ALI大鼠血液白细胞(WBC)分类计数和肺指数测定、肺组织病理学检查,采用原位杂交检测肺组织中NF_kB p65 mRNA的表达。结果两次打击后的ALI大鼠血液中WBC和肺指数显著升高,WBC分类呈现淋巴细胞比率降低而中性粒细胞比例增高的变化,呈典型的急性炎症表现,肺脏病理改变严重。原位杂交检测显示,ALI大鼠肺组织细胞中NF_kB p65 mRNA的表达明显增强。结论在ALI大鼠肺组织细胞中NF_kB p65 mRNA的表达量增强,参与了急性肺损伤的发病作用。  相似文献

目的观察头孢曲松(CFX)通过调控与缝隙连接蛋白43(Cx43)表达在治疗创伤性颅脑损伤(TBI)的相关性机制。方法选取100只体重350~450g的4月龄的SD雄性大鼠,采用液压打击仪建立脑损伤模型,具体分组为:①假手术(sham)组、②治疗(TBI+CFX)治疗组、③甘珀酸(CBX)阻断(TBI+CFX+CBX)组、④溶媒(TBI+生理盐水)组及⑤损伤(TBI)组,每组20只。实验时间均设定为TBI后48 h。每组取10只大鼠进行行为学检测和脑组织含水量测定,另10只则获取海马区胶质组织通过蛋白免疫印记法(WB)法测定神经元缝隙连接蛋白40(Cx40)及磷酸化Cx43蛋白(p-Cx43)的变化、采用酶联免疫荧光(ELFA)检测神经元氧化应激因子NADPH氧化酶活性的变化。结果实验结果显示,采用CFX干预组神经损伤严重度评分(NSS)、脑水肿程度、腺嘌呤二核苷酸磷酸(NADPH)氧化酶活性及海马胶质细胞Cx40、p-Cx43表达均溶媒组及损伤组明显下降(P 0.05)。而CBX可明显抑制上述CFX对Cx40、p-Cx43蛋白表达下调及降低NADPH氧化酶的活性的作用(P0.05)。结论 CFX可能通过抑制Cx43蛋白磷酸化和Cx40蛋白异常增多,从而抑制氧化应激反应以达到脑损伤缓解作用。  相似文献

研究衰老大鼠的学习行为与海马内突触相关蛋白的表达.运用Y-迷宫方法对幼年对照组(断乳期)和老年期(22月龄)雄性Sprague—Dawley大鼠进行Y-迷宫空间学习记忆行为训练,之后利用免疫组化和Western blot方法检测2组大鼠海马内突触素(synaptophysin,Syn)、生长相关蛋白(GAP-43)的表达情况.研究结果:(1)2组相比,老年组的学习能力较强,幼年对照组记忆能力较强(P〈0.05);(2)生长相关蛋白GAP-43在老年组海马内的表达显著高于幼年对照组;而突触素的表达低于幼年对照组(P〈0.01).提示,GAP-43和突触素可能参与大鼠空间分辨学习记忆的过程,老年大鼠的中枢神经系统保持着潜在的神经元退化修复的功能.  相似文献

Astragalus mongholicus (AM) derived from the dry root of Astragalus membranaceus Bge. var. mongolicus (Bge.) Hsiao is a widely used traditional Chinese medicine. The present study investigated the potential role of AM on renal fibrosis on a rat model of unilateral ureteral obstruction (UUO). We divided 48 Sprague-Dawley rats randomly into 4 groups: sham-operated group (Sham), untreated UUO group, AM-treated (10 g/(kg·d)) UUO group, and losartan-treated (20 mg/(kg·d)) UUO group as positive control. Haematoxylin & eosin (HE) and Masson staining were used to study the dynamic histological changes of the kidneys 7 and 14 d after operation. The expressions of fibronectin (FN), type I collagen (colI), hepatocyte growth factor (HGF), transforming growth factor-β1 (TGF-β1), and α-smooth muscle actin (α-SMA) were analyzed by real-time polymerase chain reaction (PCR), immunohistochemistry staining, and Western blot. Results show that, similar to losartan, AM alleviated the renal damage and decreased the deposition of FN and colI from UUO by reducing the expressions of TGF-β1 and α-SMA (P<0.05), whereas HGF increased greatly with AM treatment (P<0.05). Our findings reveal that AM could retard the progression of renal fibrosis. The renoprotective effect of AM might be related to inhibition of myofibroblast activation, inducing of HGF and reducing of TGF-β1 expression.  相似文献

目的观察邻苯二甲酸二丁酯(DBP)对成年大鼠睾丸Leydig细胞连接蛋白43(Cx43)及其引起睾酮分泌变化的影响,探讨DBP对睾酮(T)的调节是否与Cx43有关.方法将原代培养纯化的leydig细胞分为3组:50μg/mL DBP处理组,50μg/mL DBP+10μmol/L前列腺素E2(PGE2)组及DMSO处理组,分别同时处理细胞24h;采用细胞免疫荧光法检测细胞膜Cx43表达变化,Western印记法观察Cx43总蛋白量的变化,应用睾酮试剂盒测定睾酮值.结果与对照组相比,DBP处理24h后,Cx43表达与睾酮值均降低,差异具有统计学意义(P0.05);但加入Cx43增强剂(PGE2)组与DBP组相比,CX43表达明显恢复,但T值变化不明显(P0.05).结论DBP对睾酮及Cx43均有影响,但对Leydig细胞睾酮合成抑制作用并不一定是通过调节Cx43的表达来实现.  相似文献

Evidence suggests that the deterioration of communication between the sympathetic nervous system and cardiovascular system always accompanies the aging of human and animals. Cardiac sympathetic norepinephrine (NE) transporter (NET) on presynaptic membrane is a predominant component to eliminate released NE in the synaptic cleft and maintains the sensitivity of the β-adrenergic receptor (β-AR). In the present study, we investigated NET and β1-AR mRNA levels and sympathetic nerve density in cardiac sympathetic ganglion and left ventricular myocardium in 2- and 16-month-old rats with Northern blot analysis and immunohistochemistry. The expression levels of NET mRNA, NET protein and β1-AR mRNA in the ganglia or myocardia of 16-month-old rats were markedly reduced by 67%, 26%, and 43%, respectively, in comparison with those in 2-month-old rats. Our results also show that aging induces a strong decrease of the catecholaminergic nerve fiber density.  相似文献

Objective: To evaluate the effects oftramadol on the proinflammatory responses in a rat model of incisional pain by investigating its effects on nociceptive thresholds and serum interleukin-6 (IL-6) and IL-2 levels. Methods: Forty-two male Sprague-Dawley (SD) rats scheduled for plantar incision were randomly divided into 7 groups 01=6 in each group). Rats in Group 1 receiving general anesthesia with no incision were served as control; At 30 min before skin incision, Groups 2-5 were given 5 ml normal saline or 1, 10, and 20 mg/kg tramadol, respectively, intraperitoneally (i.p.); Group 6 received 10 mg/kg tramadol after operation; Group 7 received 10 mg/kg tramadol before incision, followed by 200 μg/kg naloxone after operation. Mechanical allodynia was measured by electronic yon Frey filament to evaluate the nociceptive thresholds 1 h before incision, and 1 h and 2 h after operation. Serum IL-6 and IL-2 levels were measured by enzyme-linked immunosorbent assay (ELISA) 2 h after operation. Results: Mechanical thresholds decreased significantly and serum IL-6 level increased significantly after operation in Group 2 compared with control (P〈0.01), and these changes were reversed respectively by tramadol in a dose-dependent manner (P〈0.05 and P〈0.01, respectively). IL-2 level remained unchanged after operation in Group 2, but decreased in Group 3 (P〈0.05), then gradually returned to the normal level in Groups 4 and 5. The intraperitoneally injected tramadol (10 and 20 mg/kg) produced a potent and dose-dependent antinocicptive effect on the lesioned paw. The antinocicptive effects of tramadol were partially an- tagonized by naloxone (200 μg/kg), suggesting an additional non-opioid mechanism. Conclusion: The results suggest that tramadol could be a good choice for the treatment of pain under the conditions that immunosuppression may be particularly contraindicated.  相似文献

Objective: This study is to determine the effect of the natural product parthenolide, a sesquiterpene lactone isolated from extracts of the herb Tanacetum parthenium, on the proliferation of vascular smooth muscle cells (VSMCs). Methods: Rat aortic VSMCs were isolated and cultured in vitro, and treated with different concentrations of parthenolide (10, 20 and 30 μmol/L). [3H]thymidine incorporation was used as an index of cell proliferation. Cell cycle progression and distribution were determined by flow cytometric analysis. Furthermore, the expression of several regulatory proteins relevant to VSMC proliferation including IκBα, cyclooxygenase-2 (Cox-2), p21, and p27 was examined to investigate the potential molecular mechanism. Results: Treatment with parthenolide significantly decreased the [3H]thymidine incorporation into DNA by 30%~56% relative to control values in a dose-dependent manner (P<0.05). Addition of parthenolide also increased cell population at G0/G1 phase by 19.2%~65.7% (P<0.05) and decreased cell population at S phase by 50.7%~84.8% (P<0.05), which is consistent with its stimulatory effects on p21 and p27. In addition, parthenolide also increased IκBα expression and reduced Cox-2 expression in a time-dependent manner. Conclusion: Our results show that parthenolide significantly inhibits the VSMC proliferation by inducing G0/G1 cell cycle arrest. IκBα and Cox-2 are likely involved in such inhibitory effect of parthenolide on VSMC proliferation. These findings warrant further investigation on potential therapeutic implications of parthenolide on VSMC proliferation in vivo.  相似文献

Objective: To investigate the factors favoring a positive prognosis for advanced primary peritoneal carcinoma (PPC). Methods: Twenty-four cases meeting the criteria for PPC were analyzed retrospectively for the clinicopathologic profiles. Immunohistochemistry was used to determine the expressions of p53, Top2α, Ki-67 and Her-2/neu. Then all these clinicopathological factors and molecular markers were correlated with the prognosis. Results: There were 15 cases of primary peritoneal serous papillary carcinoma (PPSPC), 6 cases of mixed epithelial carcinoma (MEC) and 3 cases of malignant mixed Mullerian tumor (MMMT). All patients underwent cytoreductive surgery with optimal debulking achieved in 3 cases. Among those receiving first-line chemotherapy, 13 patients received the TP regimen (paclitaxel-cisplatin or carboplatin) and 7 patients received the PAC regimen (cisplatin-doxorubicin-cyclophosphamide). The median overall survival of all patients was 42 months, while the breakdown for survival time for patients with PPSPC, MMT and MEC was 44, 13 and 19 months, respectively. The expressions of p53, Top2α and Ki-67 were all demonstrated in 11 cases respectively. None showed the expression of Her-2/neu. There were significant differences in the median survival between patients with PPSPC and those with MMMT (44 months vs 13 months, P〈0.05), also between patients receiving TP combination and those receiving the PAC regimen (75 months vs 28 months, P〈0.05). Another significant difference in the median progression-free survival (PFS) was identified between patients with positive p53 immunostaining and those with negative p53 immunostaining (15 months vs 47 months, P〈0.05), whereas age, menopausal status, residual tumor size and the other molecular factors did not significantly impact survival. Conclusion: Patients with PPC should be treated with a comprehensive management plan including appropriate cytoreductive surgery and responsive chemotherapy. Overestimating an o  相似文献

从神经、血管、代谢、离子通道等多个角度出发,研究糖尿病性勃起功能障碍(DMED)的发病机制,为开展DMED的综合治疗临床研究提供前期研究基础.成年雄性SD大鼠50只,随机取35只大鼠用于制作糖尿病模型,其余大鼠作为正常对照.饲养8周后,用阿朴吗啡法筛选DMED大鼠,用电刺激勃起神经测定海绵体内压(ICP)方法评价勃起功能.取正常组和DMED组大鼠的阴茎海绵体组织,分成若干份,用免疫组化法测定海绵体组织一氧化氮合酶(NOS)的3种亚型(nNOS,eNOS,iNOS)的变化;用Western Blot法测定神经生长因子(NGF)在海绵体组织的表达差异;用放射免疫法测定海绵体组织血管紧张素II(AngII)含量差异;用Western Blot法测定血管内皮生长因子(VEGF)和缝隙连接蛋白Connexin-43含量差异.与正常对照组相比,DMED大鼠ICP测定值明显降低;海绵体组织中nNOS、eNOS表达明显降低,而iNOS表达明显增加;NGF蛋白表达明显增加;血管紧张素II水平显著升高;VEGF蛋白表达明显减少;Connexin-43蛋白表达减少.以上试验结果说明DMED发病机制复杂,与神经、血管、代谢、离子通道等均有关.提示在今后临床治疗研究和实践中,应该从各个角度出发,针对多因素发病机制,采用综合治疗的方法,以提高疗效.  相似文献

To explore cell malignant phenotype correlated changes of cell surface adhesion molecules and cell-cell communication in carcinogenesis, human stomach transformed and cancer cell lines were investigated. Expressions of E-cadherin, N-cadherin,α-catenin, β-catenin as well as gap junction (GJ) protein Cx32 were studied by utilization of immunoblotting, immunocytochemical and fluorescent dye transfer methods. Mammalian normal stomach mucosal cells expressed E-cadherin but not N-cadherin. E-cadherin im-munofluorescence was detected at cell membranous adher-ens junctions (AJ) where colocalization with immunofluo-rescent staining of inner surface adhesion plaque proteins αnd β-catenins was observed. The existence of E-cadherin/ catenin (α-, β-) protein complexes as AJ was suggested. In transformed and stomach cancer cells E-cadherin was inhibited, instead, N-cadherin was expressed and localized at membranous AJ where co-staining with α- and β-catenin fluorescence was observed. Formation of N-cadherin/catenin (α-, β-) protein complex at AJs of transformed and cancer cells was suggested. The above observations were further supported by immunoblotting results. Normal stomach muscosal and transformed cells expressed Cx32 at membranous GJ and were competent of gap junction communication (GJIC). In stomach cancer cells, Cx32 was inhibited and GJIC was defective. The results suggested that changes of signal pathways mediated by both cell adhesion and cell communication systems are associated intracellular events of stomach carcinogenesis. The alteration of cadherin isoform from E- to N-cadherin in transformed and stomach cancer cells is the first report.  相似文献

目的研究阿托伐他汀对自发性高血压大鼠肥厚心肌过氧化物酶体增殖物激活受体γ(PPARγ)及p21表达的影响,了解阿托伐他汀改善左室肥厚的可能机制.方法筛选8周龄同批次自发性高血压大鼠20只作为研究对象,将其随机分为空白组和观察组,每组10只,另选8周龄同批次健康大鼠作为对照组,3组大鼠的体质量、周龄、雌雄比例比较无统计学意义.空白组和健康对照组大鼠采用蒸馏水灌胃,观察组大鼠采用剂量50 mg/(kg·d-1)的阿托伐他汀灌胃,所有实验大鼠每5周测1次血压,10周后测量大鼠的血脂水平、心肌肥厚,采用免疫组化及RT-PCR观察PPARγ和p21的表达,并进行3组比较.结果 10周后,观察组大鼠的心肌肥厚指标明显低于空白组(P0.01);PPARγ和p21的表达明显高于空白组(P0.01);心肌组织PPARγ和p21 mRNA的表达呈负相关.结论阿托伐他汀通过上调自发性高血压大鼠肥厚心肌组织中PPARγ,p21的表达来调节细胞周期,可有效改善高血压左室肥厚.  相似文献

The bglS gene encoding endo-1,3-1,4-β-glucanase from Bacillus subtil& was cloned and sequenced in this study. The bglS expression cassette, including PGK1 promoter, bglS gene fused to the signal sequence of the yeast mating pheromone a-factor (MFals), and ADH1 terminator with G418-resistance as the selected marker, was constructed. Then one of the PEP4 allele of Saccharomyces cerevisiae WZ65 strain was replaced by bglS expression cassette using chromosomal integration of polymerase chain reaction (PCR)-mediated homologous recombination, and the bglS gene was expressed simultaneously. The recombinant strain S. cerevisiae (SC-βG) was preliminarily screened by the clearing hydrolysis zone formed after the barley β-glucan was hydrolyzed in the plate and no proteinase A (PrA) activity was measured in fermenting liquor. The results of PCR analysis of genome DNA showed that one of the PEP4 allele had been replaced and bglS gene had been inserted into the locus of PEP4 gene in recombinant strains. Different endo-1,3-1,4-β-glucanase assay methods showed that the recombinant strain SC-βG had high endo-1,3-1,4-β-glucanase expression level with the maximum of 69.3 U/(h·ml) after 60 h of incubation. Meanwhile, the Congo Red method was suitable for the determination of endo-1,3-1,4-β-glucanase activity during the actual brewing process. The current research implies that the constructed yeast strain could be utilized to improve the industrial brewing property of beer.  相似文献

运用免疫组化SP法及计算机图像分析技术,对SHR大鼠幼年(6周,n=15)、成年(12月,n=15),以及正常血压对照组大鼠京都Wistar(Wistar-Kyoto,WKY)幼年(6周,n=10)、成年(12月,n=10),4组大鼠心肌、肾脏、大脑切片VEF蛋白的表达水平进行了定量分析;探讨了自发性高血压大鼠(spontaneously hypertensiverat,SHR)血压升高前后心肌、肾脏、脑内血管内皮生长因子(vascular endothelial growth factor,VEGF)表达水平的变化及其意义;发现在4组大鼠心脏各部、肾小球内、大脑皮质等处均观察到散在分布的VEGF免疫反应阳性细胞,阳性反应物呈棕色颗粒状,位于胞浆中。12月龄SHR组表达水平较其它三组均高(P<0.05),阳性细胞数多且灰度值低(P<0.05);而6周龄、12月龄WKY及6周龄SHR三组间表达水平差异无显著性(P>0.05)。结果表明随着成年SHR大鼠血压的升高,心肌、肾脏、脑内VEGF蛋白的表达水平上调,可能是对高血压所致的靶器官缺血缺氧的一种代偿反应,对微血管的新生和存活有积极意义。  相似文献

We investigated the possibility of producing chicken alpha interferon (ChlFN-α) in transgenic plants. The cDNA encoding ChlFN-α was introduced into lettuce (Lactuca sativa L.) plants by using an agro-infiltration transient expression system. The ChlFN-a gene was correctly transcribed and translated in the lettuce plants according to RT-PCR and ELISA assays. Recombinant protein exhibited antiviral activity in vitro by inhibition of vesicular stomatitis virus (VSV) replication on chicken embryonic fibroblast (CEF). The results demonstrate that biologically active avian cytokine with potential pharmaceutical applications could be expressed in transgenic lettuce plants and that it is possible to generate interferon protein in forage plants for preventing infectious diseases of poultry.  相似文献
