翅果油树组织培养染色体制片方法的探讨及其染色体稳定性研究

本实验从解离条件、取材时间、培养时间、预处理药剂与时间等方面探讨了翅果油树愈伤组织染色体观察的制片方法,并在细胞水平上对翅果油树愈伤组织的染色体稳定性进行了研究.得出了翅果油树愈伤组织的最佳制片方法.翅果油树的染色体在组织培养过程中存在着一定程度的变异,且其变异率随继代次数的增加呈上升趋势.但细胞学的变异只能解释少数较极端的无性系变异,更多的无性系变异则有待于从DNA水平和生化水平进一步研究.

Studies of the Method for Smearing of Elaeagnus Mollis Callus and the Stability of Chromosome in Elaeagnus. Tissue Culture

The effects of hydrolyze condition, select materials time, culture time, medicines and time of pretreatment on amearing of Elaeagnus mollis callus for observing chromosome, and the stability of chromosome in Elaeagnus moUis tissue culture in cell level were studied in this paper. The best method for smearing of Elaeagnus mollis callus was obtained. Certain extent variation of chromosome existed in Elaeagnus mollis tissue culture. With the subculture time increasing, the mutation rate of chromosome of callus in Elaeagnus mollis was rising. But the variation in cell level can only explain some extreme somaclonal variation, further reseach about the more somaclonal variation have to be done in DNA and biochemistry level.
