This work proposes a new approach for the prediction of the electricity price based on forecasting aggregated purchase and sale curves. The basic idea is to model the hourly purchase and the sale curves, to predict them and to find the intersection of the predicted curves in order to obtain the predicted equilibrium market price and volume. Modeling and forecasting of purchase and sale curves is performed by means of functional data analysis methods. More specifically, parametric (FAR) and nonparametric (NPFAR) functional autoregressive models are considered and compared to some benchmarks. An appealing feature of the functional approach is that, unlike other methods, it provides insights into the sale and purchase mechanism connected with the price and demand formation process and can therefore be used for the optimization of bidding strategies. An application to the Italian electricity market (IPEX) is also provided, showing that NPFAR models lead to a statistically significant improvement in the forecasting accuracy.  相似文献

Membrane proteins are key elements in cell physiology and drug targeting, but getting a high-resolution structure by crystallographic means is still enormously challenging. Novel strategies are in big demand to facilitate the structure determination process that will ultimately hasten the day when sequence information alone can provide a three-dimensional model. Cell-free or in vitro expression enables rapid access to large quantities of high-quality membrane proteins suitable for an array of applications. Despite its impressive efficiency, to date only two membrane proteins produced by the in vitro approach have yielded crystal structures. Here, we have analysed synergies of cell-free expression and crystallisation in lipid mesophases for generating an X-ray structure of the integral membrane enzyme diacylglycerol kinase to 2.28-Å resolution. The quality of cellular and cell-free-expressed kinase samples has been evaluated systematically by comparing (1) spectroscopic properties, (2) purity and oligomer formation, (3) lipid content and (4) functionality. DgkA is the first membrane enzyme crystallised based on cell-free expression. The study provides a basic standard for the crystallisation of cell-free-expressed membrane proteins and the methods detailed here should prove generally useful and contribute to accelerating the pace at which membrane protein structures are solved.  相似文献

通过扫描电子显微镜微观观察的方法对三酸化学抛光液中各组分对铜合金表面抛光效果的影响进行了研究.结果表明,采用微观观察法能够更直观有效地研究各组分对铜合金表面抛光效果的影响,在此基础上,获得铜合金表面最佳抛光液:磷酸200~300ml/L、硫酸200~400ml/L(磷酸+硫酸500~600ml/L)、硝酸200~250ml/L(紫铜)、40~150ml/L(黄铜)、盐酸5~15ml/L.经处理的铜合金表面不仅外观光亮、微观平整,而且铜合金表面制备的转化膜中性盐雾测试时间更长、防腐性能更好.根据扫描电子显微镜观察铜合金微观形貌获得最佳抛光液组成的方法,简单方便、准确有效  相似文献

Primary cilia with a diameter of ~200 nm have been implicated in development and disease. Calcium signaling within a primary cilium has never been directly visualized and has therefore remained a speculation. Fluid-shear stress and dopamine receptor type-5 (DR5) agonist are among the few stimuli that require cilia for intracellular calcium signal transduction. However, it is not known if these stimuli initiate calcium signaling within the cilium or if the calcium signal originates in the cytoplasm. Using an integrated single-cell imaging technique, we demonstrate for the first time that calcium signaling triggered by fluid-shear stress initiates in the primary cilium and can be distinguished from the subsequent cytosolic calcium response through the ryanodine receptor. Importantly, this flow-induced calcium signaling depends on the ciliary polycystin-2 calcium channel. While DR5-specific agonist induces calcium signaling mainly in the cilioplasm via ciliary CaV1.2, thrombin specifically induces cytosolic calcium signaling through the IP3 receptor. Furthermore, a non-specific calcium ionophore triggers both ciliary and cytosolic calcium responses. We suggest that cilia not only act as sensory organelles but also function as calcium signaling compartments. Cilium-dependent signaling can spread to the cytoplasm or be contained within the cilioplasm. Our study thus provides the first model to understand signaling within the cilioplasm of a living cell.  相似文献

Leber congenital amaurosis (LCA) is a blinding retinal disease that presents within the first year after birth. Using exome sequencing, we identified mutations in the nicotinamide adenine dinucleotide (NAD) synthase gene NMNAT1 encoding nicotinamide mononucleotide adenylyltransferase 1 in eight families with LCA, including the family in which LCA was originally linked to the LCA9 locus. Notably, all individuals with NMNAT1 mutations also have macular colobomas, which are severe degenerative entities of the central retina (fovea) devoid of tissue and photoreceptors. Functional assays of the proteins encoded by the mutant alleles identified in our study showed that the mutations reduce the enzymatic activity of NMNAT1 in NAD biosynthesis and affect protein folding. Of note, recent characterization of the slow Wallerian degeneration (Wld(s)) mouse model, in which prolonged axonal survival after injury is observed, identified NMNAT1 as a neuroprotective protein when ectopically expressed. Our findings identify a new disease mechanism underlying LCA and provide the first link between endogenous NMNAT1 dysfunction and a human nervous system disorder.  相似文献

In this article, I will view realist and non-realist accounts of scientific models within the larger context of the cultural significance of scientific knowledge. I begin by looking at the historical context and origins of the problem of scientific realism, and claim that it is originally of cultural and not only philosophical, significance. The cultural significance of debates on the epistemological status of scientific models is then related to the question of ‘intelligibility’ and how science, through models, can give us knowledge of the world by presenting us with an ‘intelligible account/picture of the world’, thus fulfilling its cultural-epistemic role. Realists typically assert that science can perform this role, while non-realists deny this. The various strategies adopted by realists and non-realists in making good their respective claims, is then traced to their cultural motivations. Finally I discuss the cultural implications of adopting realist or non-realist views of models through a discussion of the views of Rorty, Gellner, Van Fraassen and Clifford Hooker on the cultural significance of scientific knowledge.  相似文献

The multiferroic properties of Bi Fe O3-based ceramics were improved through optimizing their sintering method and doping with certain rare earth elements in pure Bi Fe O3. Some methods, especially liquid-phase sintering method has largely decreased the densities of oxygen vacancies and Fe2in Bi Fe O3-based ceramics, and thus their resistivity became high enough to measure the saturated polarization and the large piezoelectric d33 coefficient under the high electric field of [150 k V/cm. Besides,multiferroic properties were improved through the rare earth elements’ doping in pure Bi Fe O3. Magnetization commonly increases with the proportional increase of Nd,La, Sm and Dy contents up to *30 %, while ferroelectric phase can transform to paraelectric phase at a certain proportion. An improved magnetoelectric coupling was often observed at ferroelectric phase with a relatively large proportion. Besides, an enhanced piezoelectric coefficient is expected in Bi Fe O3-based ceramics with morphotropic phase boundaries as they are already observed in thin epitaxial Bi Fe O3 films.  相似文献

Rare earth Dy~(3+)and divalent Mn~(2+)elements substituting W-type hexagonal ferrites Ba_(1-x)Dy_xZn_2Fe_(16-y)Mn_yO_(27)(x=0,0.02,0.06,0.1 and y=0,0.1,0.3,0.5)were prepared by sol-gel method.The thermo-gravimetric analysis(TGA)and differential scanning calorimetry(DSC)was carried out to find the temperature at which single phase can be obtained.XRD patterns indicate the presence of the single phase for all the synthesized samples with the absence of any extra peak due to unreacted material and secondary phases.The occurrence of absorption bands at low wave numbers(563 and 446 cm~(-1)),can be assigned to the stretching vibration of metal and oxygen ions in FTIR spectra,which also confirms the single hexagonal phase for prepared material.The grains are of platelet like structure,which enhances the microwave absorption properties of hexagonal ferrites.The synthesized material exhibits the minimum reflection loss of-20.9 dB at 11.8 GHz frequency,which reflects the applications of this material in super high frequency devices.The microwave conductivity of the material increases with frequency.  相似文献

The main objective of the study was to control the degradation rate of material at a higher degradation rate improving the chemical stability of the material. Ta is known to have good chemical resistance, biocompatibility and show no adverse biological response. In the present study, SiO2–Na2O–CaO–P2O5 bioceramics with different Ta2O5 contents was prepared by solid state sintering method at 1000 °C. The as-sintered ceramics were subjected to immersion studies in stimulated body fluid (SBF) for 21 days under static condition and characterized by XRD, FTIR, SEM, and AAS. The findings of the research indicate that the addition of Ta2O5 controlled degradability, and all samples showed sufficient bioactivity.  相似文献
