The NOX toolbox: validating the role of NADPH oxidases in physiology and disease

Reactive oxygen species (ROS) are cellular signals but also disease triggers; their relative excess (oxidative stress) or shortage (reductive stress) compared to reducing equivalents are potentially deleterious. This may explain why antioxidants fail to combat diseases that correlate with oxidative stress. Instead, targeting of disease-relevant enzymatic ROS sources that leaves physiological ROS signaling unaffected may be more beneficial. NADPH oxidases are the only known enzyme family with the sole function to produce ROS. Of the catalytic NADPH oxidase subunits (NOX), NOX4 is the most widely distributed isoform. We provide here a critical review of the currently available experimental tools to assess the role of NOX and especially NOX4, i.e. knock-out mice, siRNAs, antibodies, and pharmacological inhibitors. We then focus on the characterization of the small molecule NADPH oxidase inhibitor, VAS2870, in vitro and in vivo, its specificity, selectivity, and possible mechanism of action. Finally, we discuss the validation of NOX4 as a potential therapeutic target for indications including stroke, heart failure, and fibrosis.     

基于18S rDNA序列鉴定北部湾养殖区引发藻华的一株原甲藻

高度保守的核糖体18SrDNA序列可用于经济快捷且精确地鉴定浮游植物的种类。2018年夏季,广西北部湾近海的对虾养殖区内发生了原甲藻引起的藻华,藻华的优势种密度为5.5×105 cells·L~(-1),本文采用形态学观察法结合序列分析鉴定的方法来鉴定该藻的种类。结果表明,在形态上,这株分离培养获得的原甲藻(编号BBGA-9003),其大部分细胞类似心形或卵形,少部分细胞近似三角形,鞭毛2条,细胞长和宽分别为(16.55±1.42)μm和(13.84±1.86)μm。该原甲藻经18SrDNA基因序列测定并进行BLAST分析,结果表明该原甲藻序列和GenBank中56株不同种原甲藻具有很高的同源性,在系统进化树上与不同来源的微小原甲藻聚在一大分支上,且与各微小原甲藻间的遗传距离均小于与其他种间的遗传距离。因此,从分子生物学的角度鉴定该株原甲藻属于微小原甲藻,此种藻华的发生意味着北部湾养殖区有害藻华种类多样性的增加。本研究为18SrDNA序列测定用于海洋浮游植物种类鉴定提供了实验依据,也为广西北部湾养殖区有害藻华的防治提供基础资料。     

Effects of thyroidectomy on monoamine oxidase activities toward tyramine and serotonin in the circumventricular nuclei of the rat.

Following thyroidectomy, monoamine oxidase (MAO) activities toward tyramine decreased significantly by 20% in the nucleus periventricularis and the nucleus arcuatus among the 3 hypothalamic nuclei of the rat, while MAO activity toward serotonin decreased significantly by 10% only in the nucleus periventricularis. It is suggested that thyroidectomy induced selective changes on the multiple forms of MAO in the discrete circumventricular nuclei.     

Character and OTU stability in five taxonomic groups

The character and OTU stability of classifications based on UPGMA clustering and maximum parsimony (MP) trees were compared for 5 datasets (families of angiosperms, families of orthopteroid insects, species of the fish genusIctalurus, genera of the salamander family Salamandridae, and genera of the frog family Myobatrachidae). Stability was investigated by taking different sized random subsamples of OTUs or characters, computing UPGMA clusters and an MP tree, and then comparing the resulting trees with those based on the entire dataset. Agreement was measured by two consensus indices, that of Colless, computed from strict consensus trees, and Stinebrickner's 0.5-consensus index. Tests of character stability generally showed a monotone decrease in agreement with the standard as smaller sets of characters are considered. The relative success of the two methods depended upon the dataset. Tests of OTU stability showed a monotone decrease in agreement for UPGMA as smaller sets of OTUs are considered. But for MP, agreement decreased and then increased again on the same scale. The apparent superiority of UPGMA relative to MP with respect to OTU stability depended upon the dataset. Considerations other than stability, such as computer efficiency or accuracy, will also determine the method of choice for classifications.     

A study on the Web intelligence

This paper surveys important aspects of Web Intelligence (WI). WI explores the fundamental roles as well as practical impacts of Artificial Intelligence (AI) and advanced Information Technology (IT) on the next generation of Web - related products, systens, and activities. As a direction for scientific research and devlopment,WI can be extremely beneficial for the field of Artificial Intelligence in Education (AIED). This paper covers these issues only very briefly. It focuses more on other issues in WI, such as intelligent Web services, and semantic web, and proposes how to use them as basis for tackling new and challenging research problems in AIED.     

韩国的全球化和行政改革

本文回顾了1997年经济危机后韩国政府采取的改革方案,并做出结论:与普遍的看法相反,韩国政府的大部分改革方案并不是基于新公共管理系统(NPM)的.NPM倡导的打破常规的概念是要把官僚主义者从政府内部的束缚中解放出来,然而韩国政府的改革目标却指向加强官僚政治的内部控制.另一个结论是:以NPM为导向的改革方案不适合韩国政府,因为韩国缺少成功实施NPM方案所应具备的先决条件,如基于规则的政府和充满活力的市场.本文强调了加强官僚政治外部控制机制的重要性.体制僵化、腐败、生产力低下这样的问题长期困扰韩国政府,这些问题不能通过采用NPM所建议的市场原则来解决.因此,本文的结论是:新统治理论是根除这些问题的更为行之有效的方法.     

Serotonin and 5-hydroxyindoleacetic acid concentrations in individual hypothalamic nuclei and other brain areas of rat

Summary Serotonin and 5-hydroxyindoleacetic acid (5-HIAA) were measured in individual nuclei of rat hypothalamus and other brain areas using HPLC with electrochemical detection. 5-HIAA levels were first demonstrated in hypothalamic and some discrete brain areas. The 5-HIAA/5-HT ratio was highest in the n. caudatus putamen, high in the n. ventromedialis and lowest in the n. suprachiasmaticus.
