A phosphatase complex that dephosphorylates gammaH2AX regulates DNA damage checkpoint recovery

One of the earliest marks of a double-strand break (DSB) in eukaryotes is serine phosphorylation of the histone variant H2AX at the carboxy-terminal SQE motif to create gammaH2AX-containing nucleosomes. Budding-yeast histone H2A is phosphorylated in a similar manner by the checkpoint kinases Tel1 and Mec1 (ref. 2; orthologous to mammalian ATM and ATR, respectively) over a 50-kilobase region surrounding the DSB. This modification is important for recruiting numerous DSB-recognition and repair factors to the break site, including DNA damage checkpoint proteins, chromatin remodellers and cohesins. Multiple mechanisms for eliminating gammaH2AX as DNA repair completes are possible, including removal by histone exchange followed potentially by degradation, or, alternatively, dephosphorylation. Here we describe a three-protein complex (HTP-C, for histone H2A phosphatase complex) containing the phosphatase Pph3 that regulates the phosphorylation status of gammaH2AX in vivo and efficiently dephosphorylates gammaH2AX in vitro. gammaH2AX is lost from chromatin surrounding a DSB independently of the HTP-C, indicating that the phosphatase targets gammaH2AX after its displacement from DNA. The dephosphorylation of gammaH2AX by the HTP-C is necessary for efficient recovery from the DNA damage checkpoint.     

A photometric redshift of z = 6.39 +/- 0.12 for GRB 050904

Gamma-ray bursts (GRBs) and their afterglows are the most brilliant transient events in the Universe. Both the bursts themselves and their afterglows have been predicted to be visible out to redshifts of z approximately 20, and therefore to be powerful probes of the early Universe. The burst GRB 000131, at z = 4.50, was hitherto the most distant such event identified. Here we report the discovery of the bright near-infrared afterglow of GRB 050904 (ref. 4). From our measurements of the near-infrared afterglow, and our failure to detect the optical afterglow, we determine the photometric redshift of the burst to be z = 6.39 - 0.12 + 0.11 (refs 5-7). Subsequently, it was measured spectroscopically to be z = 6.29 +/- 0.01, in agreement with our photometric estimate. These results demonstrate that GRBs can be used to trace the star formation, metallicity, and reionization histories of the early Universe.     

Spinoza on the resistance of bodies

Commentators often claim that the bodies of Spinoza’s physics resist the changes they undergo. But it’s not always clear what they mean when they say this, or whether they are entitled to say it. This article clarifies what it might mean to for Spinoza’s bodies to resist change, and examines the evidence for such a view. In the first half, the author argues that there is some limited evidence for such a view, but not nearly as much as people think. In the second half, the author proposes looking for a mental analogue to collision in the realm of ideas and argues that adequacy amounts to a meaningful concept of resistance in Spinoza, albeit one that is incomplete.     

A Bayesian Approach to Modelling the Observed Bovine Spongiform Encephalopathy Epidemic

The threat of Bovine Spongiform Encephalopathy (BSE) to humans was realized in March 1996, when the Spongiform Encephalopathy Advisory Committee (SEAC) reported to the government that the most likely explanation of the new variant Creutsfeldt–Jakob Disease (nvCJD) in humans was exposure to the BSE agent. To assess the potential risk of nvCJD, it is necessary to understand the development of the BSE epidemic within the cattle population. This paper reports on work undertaken to model the observed BSE epidemic in the UK using established Bayesian methodology. © 1997 John Wiley & Sons, Ltd.     

Control of the carrier-envelope phases of a synchronously pumped femtosecond optical parametric oscillator and its applications

The intrinsic synchronization, multi-color outputs and related carrier-envelope phases （CEP） among pulses bring advantages to synchronously pumped femtosecond optical parametric oscillators and the pumping sources for broadband frequency comb generation and ultrashort waveform coherent syn-thesis. In this paper, we discuss our latest research results in this field, which cover the following as-pects： the phase relationship and energy conservation law in an OPO and related experimental verifi-cation; control of the pumping Ti-sapphire femtosecond laser＇s CEP by self-referencing technology, and its repetition-rate locking by piezoelectric transducer （PZT）; CEP locking of the pulses from the OPO by beating the non-phase-matched visible outputs against pump supercontinuum to obtain a driving signal for a fast PZT on the OPO end mirror; the generation of a broadband frequency comb spanning from 400 nm to 2.4 μm with 1.2 kHz bandwidth; and the realization of coherent interference between phase controlled pump pulses and signal second harmonic pulses.     

Natural history of a South African insect pollinator assemblage (Insecta: Coleoptera,Diptera, Hymenoptera,Lepidoptera): diagnostic notes,food web analysis and conservation recommendations

We describe activity patterns, relative abundances and pollen transport by insect floral visitors in the Skukuza Ranger District, Kruger National Park, Republic of South Africa, based on field surveys conducted during the early rainy seasons of years 2006 to 2012 (inclusive). Diagnostic notes, illustrations and natural history observations are provided for species in the families Buprestidae, Cerambycidae, Cleridae, Lycidae and Scarabaeidae (Coleoptera), Muscidae, Platystomatidae and Tabanidae (Diptera), Apidae, Halictidae, Megachilidae, Sphecidae and Vespidae (Hymenoptera), and Pieridae and Thyrididae (Lepidoptera) that transported pollen of tree or shrub species. Observations on floral phenology and pollination syndrome are presented for 27 flowering tree or shrub species. To provide a foundation for studies of the effects of drought and climate change on pollination services in the Kruger National Park, we applied methods of food web analysis to characterise this plant-pollinator assemblage. The food web analysis shows moderate levels of redundancy in plant-pollinator interactions (with connectance values averaging 0.19 for plant species and 0.20 for pollinator species), suggesting that pollination services in this system may exhibit some resilience to environmental perturbations. Possible conservation strategies for maintaining pollination services in this ecological system are discussed, including habitat management to conserve larval or nesting areas for social and solitary bees and scarab beetles, as well as further inventories and long-term monitoring of pollinator species.
