脱落酸(ABA)受体RCARs在ABA信号传导过程中起关键作用,研究ABA受体的相互作用蛋白质对进一步了解植物抗逆的分子机制有重要的意义.本研究通过酵母双杂交发现CARK7与ABA受体RCAR12共转化酵母能在营养缺陷型平板SD/-Trp-Leu-Ade上生长.将CARK7与RCAR12分别克隆到原核表达载体pET-28a(+)和pGEX-6p-1中,构建融合蛋白表达载体,并进行原核表达和亲和纯化.纯化所得蛋白进行GST-pull down实验,结果表明RCAR12能够把CARK7蛋白拉下来.上述结果说明CARK7与RCAR12在体外存在相互作用.进一步通过双分子荧光互补实验发现CARK7与RCAR12在烟草叶片表皮细胞中能产生荧光信号,说明两者在体内同样具有明显的相互作用.  相似文献

植物激素ABA是植物应对非生物胁迫的响应因子,广泛参与了植物生长发育的调控.ABA结合其受体后抑制PP2Cs的磷酸酶活性,释放SnRK2s,从而启动ABA信号转导途径.本文采用酵母双杂交系统,GST-pull down技术研究蛋白质激酶CARK3与ABA受体RCAR12在体外的相互作用,结果显示CARK3和RCAR12共转入酵母后,在三缺平板上能够正常生长;经His-Tag抗体检测,CARK3与RCAR12出现明显且单一的条带.同时,采用双分子荧光互补实验研究CARK3与RCAR12在体内的相互作用,结果显示CARK3与RCAR12共转化烟草后能观察到较强的荧光.体内外实验表明,CARK3与RCAR12存在明显的相互作用,CARK3可能通过直接磷酸化ABA受体RCAR12来调控ABA信号途径的生理应答.  相似文献

为研究钙依赖蛋白激酶CPK11参与ABA信号通路的方式,本文利用体外酵母双杂实验(Y2H)以及体内双分子荧光互补实验(BiFC)分析CPK11与ABA响应元件结合因子(ABA-responsive element binding factors, ABFs)ABF4之间的关系.酵母双杂交实验表明CPK11与ABF4存在体外相互作用,双分子荧光互补实验表明CPK11与ABF4存在体内相互作用.以上实验共同证明CPK11与ABF4存在直接相互作用.作为CPK11的同源蛋白CPK4与转录因子ABF4在植物体内也存在相互作用.以上实验表明CPK11及其同源蛋白CPK4可能通过与转录因子ABF4相互作用从而参与钙离子介导的ABA信号通路.  相似文献

为进一步分析ABA尿苷二磷酸葡萄糖基转移酶UGT71B6参与植物抗逆反应的具体机制,本研究通过体外酵母双杂交实验与GST-Pull down实验、体内双分子荧光互补实验(BIFC)分析了ABA受体RCARs与UGT71B6之间的相互作用关系.实验结果显示ABA受体RCAR12、RCAR13与UGT71B6,在体内、体外均存在直接的相互作用.研究表明UGT71B6的生理功能与ABA受体之间存在重要联系, UGT71B6可能通过ABA受体的介导参与了植物逆境应答反应.实验结果也为初步探究ABA受体是否参与ABA稳态调节过程提供一定参考.  相似文献

以拟南芥脱落酸(ABA)受体RCAR1和以文为诱饵蛋白质,利用酵母双杂交(Y2H)技术筛选出转录因子AtMYB44,本文进行了pull down及重转酵母验证,确定它们之间的关系,实验结果显示RCAR1与AtMYB44之间确实存在相互作用.此外,为了确定RCAR1在AtMYB44上的作用区域,构建了不同长度的AtMYB44cDNA序列的PGADT7载体,利用酵母双杂交实验分析,表明只有AtMYB44的N端54~105氨基酸序列与RCAR1存在相互作用.  相似文献

为分析拟南芥钙依赖蛋白激酶CPK12在不同物种中序列的相似性,并研究CPK12与ABA信号通路的关系,本研究对不同物种CPK12氨基酸序列进行比较,以及通过体内和体外实验验证CPK12与ABF4之间是否存在直接相互作用关系.通过对拟南芥(Arabidopsis thaliana),油菜(Brassica napus)与荠菜(Capsella rubella)等物种的同源氨基酸序列进行比较,获得了它们的亲缘关系.利用酵母双杂交系统,将CPK12与ABF4共转酵母后,在四缺型培养基上能够正常生长,这说明CPK12与ABF4在体外有相互作用.同时,利用双分子荧光互补实验,CPK12与ABF4共转烟草后能够在激发光下观察到烟草细胞核中出现明显荧光信号,这说明CPK12与ABF4在植物体内也存在相互作用,为CPK12参与ABA信号途径提供了直接证据.  相似文献

本文以RCAR1为诱饵蛋白通过酵母双杂交(Y2H)技术筛选出了DWD基因编码的蛋白,同时在用ABI2参与筛选的过程中,同样得到了DWD这个基因. 进行了pull down 实验验证它们之间的相互作用,实验结果显示:在体外,RCAR1、RCAR3与DWD之间确实存在相互作用,同时ABI2与DWD也存在相互作用. 其后对DWD结构的进行分析和实验,发现ABI2和DWD的相互作用区域集中在AtDWDC端,大概是145~350位氨基酸之间的WD40结构域里.  相似文献

本文通过酵母双杂交实验(Y2H)和双荧光互补实验(BIFC)分别从体外和体内条件下检测了AtDWD与CUL4型E3连接酶的底物识别蛋白结合蛋白DDB1a和DDB1b的相互作用,发现AtDWD与DDB1b有较强的相互作用,而与DDB1a的相互作用较弱,相互作用区域位于AtDWD的C末端部位.说明了AtDWD是潜在的CUL4型E3连接酶的底物识别亚基.此外,本文中还发现AtDWD过表达植株对冷冻胁迫更加敏感.AtDWD可能参与冷胁迫信号通路.  相似文献

脱落酸(abscisic acid,ABA)是一种重要的植物激素,它能使植物适应逆境胁迫,从而调节植物的生长发育状况.现今ABA受体已被公认为RCARs/PYR/PYLs家族蛋白.本实验通过拟南芥为模式植物,利用酵母双杂交系统共转化方法从拟南芥cDNA文库中筛选与RCAR3相互作用的蛋白质,获得多个阳性克隆,从中选取一个与生长素调节相关的基因NIT1的全长cDNA重新转化酵母验证,发现与RCAR3仍然有相互作用,排除了假阳性的可能.从而为下一步研究该基因与RCAR3的相互关系做好了准备.  相似文献

The discovery of the prolific Ordovician Red River reservoirs in 1995 in southeastern Saskatchewan was the catalyst for extensive exploration activity which resulted in the discovery of more than 15 new Red River pools. The best yields of Red River production to date have been from dolomite reservoirs. Understanding the processes of dolomitization is, therefore, crucial for the prediction of the connectivity, spatial distribution and heterogeneity of dolomite reservoirs.The Red River reservoirs in the Midale area consist of 3~4 thin dolomitized zones, with a total thickness of about 20 m, which occur at the top of the Yeoman Formation. Two types of replacement dolomite were recognized in the Red River reservoir: dolomitized burrow infills and dolomitized host matrix. The spatial distribution of dolomite suggests that burrowing organisms played an important role in facilitating the fluid flow in the backfilled sediments. This resulted in penecontemporaneous dolomitization of burrow infills by normal seawater. The dolomite in the host matrix is interpreted as having occurred at shallow burial by evaporitic seawater during precipitation of Lake Almar anhydrite that immediately overlies the Yeoman Formation. However, the low δ18O values of dolomited burrow infills (-5.9‰~ -7.8‰, PDB) and matrix dolomites (-6.6‰~ -8.1‰, avg. -7.4‰ PDB) compared to the estimated values for the late Ordovician marine dolomite could be attributed to modification and alteration of dolomite at higher temperatures during deeper burial, which could also be responsible for its 87Sr/86Sr ratios (0.7084~0.7088) that are higher than suggested for the late Ordovician seawaters (0.7078~0.7080). The trace amounts of saddle dolomite cement in the Red River carbonates are probably related to "cannibalization" of earlier replacement dolomite during the chemical compaction.  相似文献

本文叙述了对海南岛及其毗邻大陆边缘白垩纪到第四纪地层岩石进行古地磁研究的全部工作过程。通过分析岩石中剩余磁矢量的磁偏角及磁倾角的变化,提出海南岛白垩纪以来经历的构造演化模式如下:早期伴随顺时针旋转而向南迁移,后期伴随逆时针转动并向北运移。联系该地区及邻区的地质、地球物理资料,对海南岛上述的构造地体运动提出以下认识:北部湾内早期有一拉张作用,主要是该作用使湾内地壳显著伸长减薄,形成北部湾盆地。从而导致了海南岛的早期构造运动,而海南岛后期的构造运动则主要是受南海海底扩张的影响。海南地体运动规律的阐明对于了解北部湾油气盆地的形成演化有重要的理论和实际意义。  相似文献

The explosive growth of the Internet and database applications has driven database to be more scalable and available, and able to support on-line scaling without interrupting service. To support more client's queries without downtime and degrading the response time, more nodes have to be scaled up while the database is running. This paper presents the overview of scalable and available database that satisfies the above characteristics. And we propose a novel on-line scaling method. Our method improves the existing on-line scaling method for fast response time and higher throughputs. Our proposed method reduces unnecessary network use, i.e. , we decrease the number of data copy by reusing the backup data. Also, our on-line scaling operation can be processed parallel by selecting adequate nodes as new node. Our performance study shows that our method results in significant reduction in data copy time.  相似文献

R-Tree is a good structure for spatial searching. But in this indexing structure,either the sequence of nodes in the same level or sequence of traveling these nodes when queries are made is random. Since the possibility that the object appears in different MBR which have the same parents node is different, if we make the subnode who has the most possibility be traveled first, the time cost will be decreased in most of the cases. In some case, the possibility of a point belong to a rectangle will shows direct proportion with the size of the rectangle. But this conclusion is based on an assumption that the objects are symmetrically distributing in the area and this assumption is not always coming into existence. Now we found a more direct parameter to scale the possibility and made a little change on the structure of R-tree, to increase the possibility of founding the satisfying answer in the front sub trees. There are numerous geometric objects stored in the spatial databases. An importance function in a spatial database is that users can browse the geometric objects as a map efficiently. Thus the spatial database should display the geometric objects users concern about swiftly onto the display window. This process includes two operations:retrieve data from database and then draw them onto screen. Accordingly, to improve the efficiency, we should try to reduce time of both retrieving object and displaying them. The former can be achieved with the aid of spatial index such as R-tree, the latter require to simplify the objects. Simplification means that objects are shown with sufficient but not with unnecessary detail which depend on the scale of browse. So the major problem is how to retrieve data at different detail level efficiently. This paper introduces the implementation of a multi-scale index in the spatial database SISP (Spatial Information Shared Platform) which is generalized from R-tree. The difference between the generalization and the R-tree lies on two facets: One is that every node and geometric object in the generalization is assigned with a importance value which denote the importance of them, and every vertex in the objects are assigned with a importance value,too. The importance value can be use to decide which data should be retrieve from disk in a query. The other difference is that geometric objects in the generalization are divided into one or more sub-blocks, and vertexes are total ordered by their importance value. With the help of the generalized R-tree, one can easily retrieve data at different detail levels.Some experiments are performed on real-life data to evaluate the performance of solutions that separately use normal spatial index and multi-scale spatial index. The results show that the solution using multi-scale index in SISP is satisfying.  相似文献

The geographic information service is enabled by the advancements in general Web service technology and the focused efforts of the OGC in defining XML-based Web GIS service. Based on these models, this paper addresses the issue of services chaining,the process of combining or pipelining results from several interoperable GIS Web Services to create a customized solution. This paper presents a mediated chaining architecture in which a specific service takes responsibility for performing the process that describes a service chain. We designed the Spatial Information Process Language (SIPL) for dynamic modeling and describing the service chain, also a prototype of the Spatial Information Process Execution Engine (SIPEE) is implemented for executing processes written in SIPL. Discussion of measures to improve the functionality and performance of such system will be included.  相似文献
