从石油污染场地分离到一株可降解菲的菌株SP 3,经生理生化鉴定和16S rDNA序列对比分析,初步鉴定该菌株为分枝杆菌(Mycobacterium sp.),该菌能够以菲为唯一碳源,在菲浓度为100 μg/mL的无机盐培养基中,28 ℃摇床培养7 d降解率达到了99%以上。对该菌进行了最佳发酵培养基及培养条件的优化研究,其最佳培养基配方为淀粉1%、牛肉膏1%、Na2HPO4 0.2%、NaH2PO4 0.2%,并通过单因素实验法确定了最佳培养条件为温度30 ℃,初始pH =7.5,培养时间12 h。  相似文献

为探索中度嗜盐菌在高盐有机工业废水处理中的应用,解决高盐有机工业废水处理这一难题,从山东省威海市路道口盐场晒盐池盐水中分离一株嗜盐菌株YS - 1,通过对该菌株进行原子力显微镜观察、生理生化测定,全细胞脂肪酸分析、16S rDNA序列同源性分析发现YS - 1菌株16S rD NA序列与Halomonas sp.(AB167061)的亲缘关系最为接近,结合上述其他各项结果确定该菌株为盐单胞菌属(Halomonas sp.),属中度嗜盐菌;在SBR反应器中对该菌株进行强化高盐有机废水处理试验,结果表明,含盐12%,CODCr=1 494 mg/L的高盐模拟有机废水,经72 h C ODCr去除率为90.0%,120 h 的CODCr去除率为98.1%,该菌株具有强化高盐有机废水处理的功能,通过分离筛选嗜盐菌强化高盐有机工业废水处理具有可行性.  相似文献

以葡萄糖为碳源,逐步增大培养基中丙烯腈浓度进行重复续代培养,从土样中筛选到一株腈水合酶活力较高的菌株.经形态观察和16S rDNA序列分析,该菌株鉴定为红球菌属(Rhodococcus)菌株,命名为Rhodococcus sp.TCCC 28001.对菌株TCCC 28001培养条件进行了初步研究,确定培养基为葡萄糖15 g/L,酵母粉5 g/L,尿素7g/L,Coz+10x10-5mol/L.28℃,180r/min培养72 h,腈水合酶活力达892 U/mL.  相似文献

自上海市苏州河中游分离得到一株兼性厌氧苯酚降解菌SZH3,该菌株能够在以苯酚为唯一碳源和能源的培养基上生长,经形态、生理生化以及16S rDNA序列特性分析,初步将该菌株鉴定为泛菌属(Pantoea sp.).进一步研究发现,在初始苯酚浓度为500 mg,/L时,SzH3的苯酚降解率在60 h内就可达到95.4%.该菌株最高可耐受浓度为1300 mg/L的苯酚,对苯酚最适降解条件为pH 7.0,温度30 ℃,转速200 r/min.  相似文献

从南极深海底泥中分离到一株中度嗜盐菌NJS-2,经过16S rDNA(已提交GenBank,其登录号为DQ789389)序列分析、形态学和生理生化特征分析,该菌株初步鉴定为色盐杆菌属(Chromohalobacter).以其16S rDNA序列相似性为基础构建了包括8种相关种属在内的系统发育树.同源性分析表明,NJS-2与Chromohalobacter salexigens 的16S rDNA相似性达到99%以上,应归属于色盐杆菌属,命名该菌株为Chromohalobacter sp.NJS-2.研究了菌株NJS-2的相容性溶质四氢嘧啶(Ectoine)的累积及累积条件.研究发现,30℃条件下,在含2 mol/L NaCl的培养基中培养48 h,可最大诱导生成Ectoine 300 mg/L.反复渗透性休克试验表明:NJS-2在低渗条件下能够快速将细胞内的Ectoine分泌到细胞外,而在高渗环境中能够较快地重新合成.  相似文献

红树林沉积物中的微生物对苯并[a]芘的降解研究   总被引:4,自引:0,他引:4  
采用高效液相色谱(HPLC)结合紫外检测研究了一组筛选自红树林沉积物中的混合菌,该混合菌能以苯并[a]芘(BaP)作为唯一碳源和能源进行代谢,结果表明,当BaP浓度为20 mg*L-1时,培养63 d后有32.85%的BaP被降解.经分离,此混合菌中包含5种菌,16S rDNA序列测定结果与GenBank数据库进行同源性比较,显示M1 5-1可能为一新种,其余4株菌分别为红球菌属(Rhodococcus sp.),微小杆菌属(Exiguobacterium sp.),节杆菌属(Arthrobacter sp.),芽孢杆菌属(Bacillus sp.),绘制了系统发育树,并对其系统发育进行分析.同时还强调了红树林微生物在BaP污染环境生物修复中作用的重要性.  相似文献

一株从土壤中分离的能够降解苯酚的菌株,经形态观察和16S rDNA鉴定,确定为无色杆菌属。对不同温度、pH值、盐度以及不同苯酚质量浓度培养基中菌株的生长和苯酚降解效率进行了研究,结果表明:该菌株能在30 ℃、67 h内完全降解600mg/L的苯酚,并能对1000 mg/L的苯酚起到降解作用;在温度为30 ℃、pH=7、盐度为0.3%时,该菌株的生长及苯酚降解效率较好。  相似文献

The discovery of the prolific Ordovician Red River reservoirs in 1995 in southeastern Saskatchewan was the catalyst for extensive exploration activity which resulted in the discovery of more than 15 new Red River pools. The best yields of Red River production to date have been from dolomite reservoirs. Understanding the processes of dolomitization is, therefore, crucial for the prediction of the connectivity, spatial distribution and heterogeneity of dolomite reservoirs.The Red River reservoirs in the Midale area consist of 3~4 thin dolomitized zones, with a total thickness of about 20 m, which occur at the top of the Yeoman Formation. Two types of replacement dolomite were recognized in the Red River reservoir: dolomitized burrow infills and dolomitized host matrix. The spatial distribution of dolomite suggests that burrowing organisms played an important role in facilitating the fluid flow in the backfilled sediments. This resulted in penecontemporaneous dolomitization of burrow infills by normal seawater. The dolomite in the host matrix is interpreted as having occurred at shallow burial by evaporitic seawater during precipitation of Lake Almar anhydrite that immediately overlies the Yeoman Formation. However, the low δ18O values of dolomited burrow infills (-5.9‰~ -7.8‰, PDB) and matrix dolomites (-6.6‰~ -8.1‰, avg. -7.4‰ PDB) compared to the estimated values for the late Ordovician marine dolomite could be attributed to modification and alteration of dolomite at higher temperatures during deeper burial, which could also be responsible for its 87Sr/86Sr ratios (0.7084~0.7088) that are higher than suggested for the late Ordovician seawaters (0.7078~0.7080). The trace amounts of saddle dolomite cement in the Red River carbonates are probably related to "cannibalization" of earlier replacement dolomite during the chemical compaction.  相似文献

There are numerous geometric objects stored in the spatial databases. An importance function in a spatial database is that users can browse the geometric objects as a map efficiently. Thus the spatial database should display the geometric objects users concern about swiftly onto the display window. This process includes two operations:retrieve data from database and then draw them onto screen. Accordingly, to improve the efficiency, we should try to reduce time of both retrieving object and displaying them. The former can be achieved with the aid of spatial index such as R-tree, the latter require to simplify the objects. Simplification means that objects are shown with sufficient but not with unnecessary detail which depend on the scale of browse. So the major problem is how to retrieve data at different detail level efficiently. This paper introduces the implementation of a multi-scale index in the spatial database SISP (Spatial Information Shared Platform) which is generalized from R-tree. The difference between the generalization and the R-tree lies on two facets: One is that every node and geometric object in the generalization is assigned with a importance value which denote the importance of them, and every vertex in the objects are assigned with a importance value,too. The importance value can be use to decide which data should be retrieve from disk in a query. The other difference is that geometric objects in the generalization are divided into one or more sub-blocks, and vertexes are total ordered by their importance value. With the help of the generalized R-tree, one can easily retrieve data at different detail levels.Some experiments are performed on real-life data to evaluate the performance of solutions that separately use normal spatial index and multi-scale spatial index. The results show that the solution using multi-scale index in SISP is satisfying.  相似文献

本文叙述了对海南岛及其毗邻大陆边缘白垩纪到第四纪地层岩石进行古地磁研究的全部工作过程。通过分析岩石中剩余磁矢量的磁偏角及磁倾角的变化,提出海南岛白垩纪以来经历的构造演化模式如下:早期伴随顺时针旋转而向南迁移,后期伴随逆时针转动并向北运移。联系该地区及邻区的地质、地球物理资料,对海南岛上述的构造地体运动提出以下认识:北部湾内早期有一拉张作用,主要是该作用使湾内地壳显著伸长减薄,形成北部湾盆地。从而导致了海南岛的早期构造运动,而海南岛后期的构造运动则主要是受南海海底扩张的影响。海南地体运动规律的阐明对于了解北部湾油气盆地的形成演化有重要的理论和实际意义。  相似文献

The explosive growth of the Internet and database applications has driven database to be more scalable and available, and able to support on-line scaling without interrupting service. To support more client's queries without downtime and degrading the response time, more nodes have to be scaled up while the database is running. This paper presents the overview of scalable and available database that satisfies the above characteristics. And we propose a novel on-line scaling method. Our method improves the existing on-line scaling method for fast response time and higher throughputs. Our proposed method reduces unnecessary network use, i.e. , we decrease the number of data copy by reusing the backup data. Also, our on-line scaling operation can be processed parallel by selecting adequate nodes as new node. Our performance study shows that our method results in significant reduction in data copy time.  相似文献

R-Tree is a good structure for spatial searching. But in this indexing structure,either the sequence of nodes in the same level or sequence of traveling these nodes when queries are made is random. Since the possibility that the object appears in different MBR which have the same parents node is different, if we make the subnode who has the most possibility be traveled first, the time cost will be decreased in most of the cases. In some case, the possibility of a point belong to a rectangle will shows direct proportion with the size of the rectangle. But this conclusion is based on an assumption that the objects are symmetrically distributing in the area and this assumption is not always coming into existence. Now we found a more direct parameter to scale the possibility and made a little change on the structure of R-tree, to increase the possibility of founding the satisfying answer in the front sub trees. The geographic information service is enabled by the advancements in general Web service technology and the focused efforts of the OGC in defining XML-based Web GIS service. Based on these models, this paper addresses the issue of services chaining,the process of combining or pipelining results from several interoperable GIS Web Services to create a customized solution. This paper presents a mediated chaining architecture in which a specific service takes responsibility for performing the process that describes a service chain. We designed the Spatial Information Process Language (SIPL) for dynamic modeling and describing the service chain, also a prototype of the Spatial Information Process Execution Engine (SIPEE) is implemented for executing processes written in SIPL. Discussion of measures to improve the functionality and performance of such system will be included.  相似文献
