自生活污水中分离得到了蛭弧菌 Bd—No.2,用超声波破碎仪处理了 Bd—No.2的寄主 E.coli K_(12)和非寄主革氏负(G~+)根瘤菌1.174、革氏正(G~+)枯草芽孢杆菌及卡氏酵母菌,用它们细胞匀浆液的稀释液培养蛭弧菌 Bd—No.2,双层平板计数确证它们都能使 Bd—No.2增殖,说明可以“腐生”培养。表明某些菌体不为蛭弧菌利用是与它们细胞壁的屏障作用有关。  相似文献

用双层平板法和液体培养法对蛭弧菌裂解泛耐药菌进行研究. 结果表明: 该株蛭弧菌能裂解泛耐药鲍曼不动杆菌、泛耐药铜绿假单胞菌、大肠杆菌这3株革兰氏阴性菌, 不能裂解凝结芽孢杆菌、金黄色葡萄球菌这2株革兰氏阳性细菌.以泛耐药鲍曼不动杆菌为宿主菌, 研究影响蛭弧菌生长和发挥噬菌特性的因素. 35℃是蛭弧菌发挥噬菌作用的最适温度. 蛭弧菌的数量在pH75时达到最大值. 浓度为1%的NaCl有利于蛭弧菌发挥噬菌特性. 在培养蛭弧菌时添加0005mol/L的Mg2+、0003mol/L的Ca2+可以提高蛭弧菌的裂解活性. 0001mol/L的谷氨酸钠可提升蛭弧菌的裂解能力. 氯仿会将蛭弧菌全部灭活.  相似文献

海洋蛭弧菌BDSG1的发酵培养条件研究   总被引:3,自引:0,他引:3  
从厦门市集美鳌园附近海域分离得到一株海洋蛭弧菌BDSG1.实验结果表明,该菌的最适生长温度为25℃,最适pH值为8.3.在人工生态环境中对该蛭弧菌与宿主弧菌相互作用的消长情况的研究表明,蛭弧菌BDSG1对弧菌有较强的裂解和净化作用.通过单因素及五因子正交试验对蛭弧菌BDSG1发酵生产条件进行优化,得到了最优培养基配方,发酵最佳时间为50 h,菌液浓度可达9.5×1010pfu/mL.  相似文献

通过富集培养后从鱼体表分离得到了有活性的蛭弧菌,双层平板培养后能出现明显的透明状噬菌斑;用银染法染色后观察到蛭弧菌呈逗点状,同时观察到蛭弧菌的连续运动状态;通过16S rRNA基因序列的同源性分析结果表明,分离到的鱼体表蛭弧菌与大多数典型蛭弧菌的同源性均较高,最高达99%,可以分类到Bdellovibrio属。该实验内容与生活贴近且涉及实验技术广泛,有助于激发学生的实验热情,促进创新型人才的培养,也为今后在微生物学实验教学中开设综合创新实验奠定了基础。  相似文献

南海珠江口区域水生病原弧菌的分离鉴定及血清学分析   总被引:1,自引:0,他引:1  
从南海珠江口之深圳、珠海海域和广州城区等五个不同区域的9个水样中,利用弧菌属选择性培养基TCBS获得9类特征菌落表型的分离株.选取高频率出现的4类典型菌落的代表菌株(G,Y1,Y2,T)鉴定,革兰氏染色为G-,磷钨酸负染色透射电镜观察菌体细胞成短棒状或弯曲弧状,可见单极生鞭毛.综合特征生化鉴定结果确定橙黄色菌株Y2为霍乱弧菌,绿色菌株G为嗜盐性副溶血弧菌.血清学类群分布研究结果表明来自于不同水域样本的菌株之间存在特征血清型的显著关联性.  相似文献

通过MTT法研究了LaCl3和GdCl3对小鼠免疫细胞的作用.结果表明,GdCl3在0.001~10μmol/L浓度内均能促进小鼠脾细胞的增殖,0.001~0.1μmol/L的LaCl3促进小鼠脾细胞的增殖,其他浓度没有影响;除0.1μmol/L浓度外,其他测试浓度下的LaCl3均促进小鼠T淋巴细胞的增殖;0.001μmol/L的GdCl3抑制小鼠T淋巴细胞的增殖,0.01~1μmol/L时对小鼠T淋巴细胞的增殖没有影响,10μmol/L时转而促进小鼠T淋巴细胞的增殖.浓度为0.1μmol/L的LaCl3和GdCl3对小鼠B淋巴细胞的增殖有抑制作用,其他测试浓度下均促进小鼠B淋巴细胞的增殖.作用时间为4 h时,0.001μmol/L的LaCl3对NK细胞的活性没有影响,其他浓度下的LaCl3均能提高NK细胞的活性,0.001~10μmol/L的GdCl3均能提高NK细胞的活性;作用时间为8 h时,0.001和0.01μmol/L的LaCl3显著降低NK细胞的活性,0.1和1μmol/L的LaCl3提高NK细胞的活性,当浓度为10μmol/L时对NK细胞的活性没有影响,1μmol/L的GdCl3降低NK细胞的活性,其他测试浓度均能提高NK细胞的活性.这提示LaCl3和GdCl3对小鼠免疫细胞的影响模式与其作用浓度、作用时间以及稀土元素的种类都是密切相关的.  相似文献

华南地区海水养殖鱼类主要弧菌病原的分离与鉴定   总被引:1,自引:0,他引:1  
对海南、广东、广西等华南地区的主要海水养殖系统的斜带石斑鱼、红鳍笛鲷、褐点石斑鱼和珍珠龙胆(棕点石斑鱼♀×鞍带石斑鱼♂)等多种海水鱼类的弧菌病开展了流行病学调查,从病鱼体内共分离获得弧菌63株,人工感染试验结果表明,其中4株为强毒株,17株为弱毒株,42株为无毒株.经鉴定,哈维氏弧菌有43株(19株毒力株)、溶藻弧菌7株(1株毒力株)、副溶血弧菌5株、轮虫弧菌2株(1株毒力株)、需钠弧菌1株、无法确定的弧菌属其他细菌5株.  相似文献

为确定菲牛蛭(Hirudinaria manillensis)体质量、年龄和应激刺激与菲牛蛭繁殖性能的关系,采用单因素方差分析在不同体质量、年龄、应激次数条件下,菲牛蛭的产卵和孵化情况.结果显示:不同体质量菲牛蛭亲本其繁殖能力有显著差异(P〈0.05),体质量为10~〈15g亲本产茧数最多,卵茧孵化率和孵化数最高;不同年龄菲牛蛭亲本,其繁殖能力差异显著(P〈0.01),实验中2~3龄亲本产茧数最多,卵茧孵化率和孵化数最高;适当的环境刺激对菲牛蛭的繁殖有促进作用,实验中1或2次操作刺激对菲牛蛭亲本产茧数、孵化率和孵化数均有明显的增加(P〈0.01).建议在菲牛蛭人工繁殖过程中,选取10~〈15g体质量、2~3年龄亲本进行繁殖,同时在繁殖过程中可适当对亲本进行1或2次刺激.  相似文献

淤泥容纳、滋生和释放病原微生物威胁城市景观水环境安全。利用陶化后的淤泥颗粒固定蛭弧菌净化城市景观水体中病原微生物,可避免物化消毒对景观水体中其他动植物造成伤害。陶化淤泥颗粒固定蛭弧菌及固定化蛭弧菌对大肠杆菌、实际景观水体中总大肠菌群和粪大肠菌群裂解的试验结果表明:淤泥陶化温度为800℃、吸附时间为6 h、培养温度为30℃、摇床转速为140 r/min时,蛭弧菌固定化的效果最好;在此条件下固定的蛭弧菌,在72 h的试验周期内,对大肠杆菌的去除率达到97.4%,远超对照组的38.4%;采用陶化淤泥颗粒固定蛭弧菌去除城市景观水体中总大肠菌群和粪大肠菌群两种指示性病原微生物,去除率分别达到97.5%和78%。  相似文献

为了优化蛭弧菌的生长条件,在前期研究确定适于蛭弧菌BD1生长的最佳pH、盐度、二价金属离子(Ca2+、Mg2+)、谷氨酸钠的浓度范围的基础上,综合考虑4个因子对蛭弧菌BD1生长的影响,利用Design Expert 6.0软件进行响应面分析,优化菌体生长条件,得到该菌株生长模型,并在模型最优值时得到各因子的水平。结果表明:BD1的最优培养条件为盐度(NaCl浓度)为2.9%,培养液pH值为7.4,Ca2+、Mg2+浓度为6.4 mmol,谷氨酸钠浓度为4.2 mmol。菌体生长过程中,盐度和二价金属离子,谷氨酸钠和二价金属离子,谷氨酸钠与盐度交互作用不显著,pH与盐度,pH与谷氨酸钠,pH与二价金属离子交互作用显著。菌体生长模型达到显著水平,可以对BD1在不同条件下的生长情况进行分析和预测。  相似文献

The Williston Basin is a significant petroleum province, containing oil production zones that include the Middle Cambrian to Lower Ordovician, Upper Ordovician, Middle Devonian, Upper Devonian and Mississippian and within the Jurassic and Cretaceous. The oils of the Williston Basin exhibit a wide range of geochemical characteristics defined as "oil families", although the geochemical signature of the Cambrian Deadwood Formation and Lower Ordovician Winnipeg reservoired oils does not match any "oil family". Despite their close stratigraphic proximity, it is evident that the oils of the Lower Palaeozoic within the Williston Basin are distinct. This suggests the presence of a new "oil family" within the Williston Basin. Diagnostic geochemical signatures occur in the gasoline range chromatograms, within saturate fraction gas chromatograms and biomarker fingerprints. However, some of the established criteria and cross-plots that are currently used to segregate oils into distinct genetic families within the basin do not always meet with success, particularly when applied to the Lower Palaeozoic oils of the Deadwood and Winnipeg Formation.  相似文献

Language is a means of verbal communication. People use language to communicate with each other. In the society, no two speakers are exactly alike in the way of speaking. Some differences are due to age, gender, statue and personality. Above all, gender is one of the obvious reasons. The writer of this paper tries to describe the features of women's language from these perspectives: pronunciation, intonation, diction, subjects, grammar and discourse. From the discussion of the features of women's language, more attention should be paid to language use in social context. The discovery of the prolific Ordovician Red River reservoirs in 1995 in southeastern Saskatchewan was the catalyst for extensive exploration activity which resulted in the discovery of more than 15 new Red River pools. The best yields of Red River production to date have been from dolomite reservoirs. Understanding the processes of dolomitization is, therefore, crucial for the prediction of the connectivity, spatial distribution and heterogeneity of dolomite reservoirs.The Red River reservoirs in the Midale area consist of 3~4 thin dolomitized zones, with a total thickness of about 20 m, which occur at the top of the Yeoman Formation. Two types of replacement dolomite were recognized in the Red River reservoir: dolomitized burrow infills and dolomitized host matrix. The spatial distribution of dolomite suggests that burrowing organisms played an important role in facilitating the fluid flow in the backfilled sediments. This resulted in penecontemporaneous dolomitization of burrow infills by normal seawater. The dolomite in the host matrix is interpreted as having occurred at shallow burial by evaporitic seawater during precipitation of Lake Almar anhydrite that immediately overlies the Yeoman Formation. However, the low δ18O values of dolomited burrow infills (-5.9‰~ -7.8‰, PDB) and matrix dolomites (-6.6‰~ -8.1‰, avg. -7.4‰ PDB) compared to the estimated values for the late Ordovician marine dolomite could be attributed to modification and alteration of dolomite at higher temperatures during deeper burial, which could also be responsible for its 87Sr/86Sr ratios (0.7084~0.7088) that are higher than suggested for the late Ordovician seawaters (0.7078~0.7080). The trace amounts of saddle dolomite cement in the Red River carbonates are probably related to "cannibalization" of earlier replacement dolomite during the chemical compaction.  相似文献

以AC-13级配为基础,将橡胶颗粒代替部分集料掺入混合料中,以低温弯曲试验为评价方法对不同橡胶颗粒掺量下沥青混合料的低温抗裂性进行研究,并引入应变能密度值对混合料的低温抗裂性进行综合评价.试验结果表明:橡胶颗粒沥青混合料试件的破坏微应变均超过2 300,满足冬寒区的技术指标;无论是否掺加橡胶颗粒,随着温度的下降,沥青混合料破坏时的最大弯拉强度增大,弯拉应变降低,劲度模量增大;弯曲应变能密度在胶粒掺量为1%左右时具有较大的弯曲应变能密度值,此时橡胶颗粒沥青混合料具有较好的低温抗裂性.  相似文献

理论推导与室内实验相结合,建立了低渗透非均质砂岩油藏启动压力梯度确定方法。首先借助油藏流场与电场相似的原理,推导了非均质砂岩油藏启动压力梯度计算公式。其次基于稳定流实验方法,建立了非均质砂岩油藏启动压力梯度测试方法。结果表明:低渗透非均质砂岩油藏的启动压力梯度确定遵循两个等效原则。平面非均质油藏的启动压力梯度等于各级渗透率段的启动压力梯度关于长度的加权平均;纵向非均质油藏的启动压力梯度等于各渗透率层的启动压力梯度关于渗透率与渗流面积乘积的加权平均。研究成果可用于有效指导低渗透非均质砂岩油藏的合理井距确定,促进该类油藏的高效开发。  相似文献

Quality traits in wheat (Triticum aestirum L.) were studied by quantitative trait locus (QTL) analysis in a recombinant inbred line (RIL) population, a set of 131 lines derived from Chuan 35050 × Shannong 483 cross (ChSh). Grains from RILs were assayed for 21 quality traits related to protein and starch. A total of 35 putative QTLs for 19 traits with a single QTL explaining 7.99-40.52% of phenotypic variations were detected on 10 chromosomes, 1D, 2A, 2D, 3B, 3D, 5A, 6A, 6B, 6D, and 7B. The additive effects of 30 QTLs were positive, contributed by Chuan 35050, the remaining 5 QTLs were negative with the additive effect contributed by Shannong 483. For protein traits, 15 QTLs were obtained and most of them were located on chromosomes 1 D, 3B and 6D, while 20 QTLs for starch traits were detected and most of them were located on chromosomes 3D, 6B and 7B. Only 7 QTLs for protein and starch traits were co-located in three regions on chromosomes 1D, 2A and 2D. These protein and starch trait QTLs showed a distinct distribution pattern in certain regions and chromosomes. Twenty-two QTLs were clustered in 6 regions of 5 chromosomes. Two QTL clusters for protein traits were located on chromosomes 1D and 3B, respectively, three clusters for starch traits on chromosomes 3D, 6B and 7B, and one cluster including protein and starch traits on chromosome 1D.  相似文献
