HPLC同时测定制草乌中三种生物碱质量比   总被引:4,自引:3,他引:1  
测定制草乌不同地区炮制品中新乌头碱、次乌头碱及乌头碱的质量比.采用HPLC法,Kroma-sil-C18色谱柱,甲醇-水-氯仿-三乙胺(w/w70∶30∶2∶0.1),流速1.0 mL/min,检测波长230 nm,常温.新乌头碱在10~120μg/mL,次乌头碱在3.9~46.8μg/mL,乌头碱在10~120μg/mL范围内线性关系良好,平均回收率分别为96.1%、95.8%和95.3%.制草乌中酯型生物碱质量分数均不超过0.15%,符合中国药典规定.  相似文献

对附子不同组织中乌头总碱和3种双酯型生物碱一新乌头碱、乌头碱和次乌头碱的含量进行了测定。用酸性染料比色法测定乌头类总生物碱的含量;反相高效液相色谱法测定新乌头碱、乌头碱、次乌头碱的含量,色谱柱为:Zorbax extend—C18(150mm×4.6mm,5.0μm),流动相为乙腈-0.1%乙二胺,梯度洗脱,流速为1mL/min,检测波长为240mn,柱温30℃。结果表明:附子不同组织中生物碱的含量呈现明显的差异性。子根、茎秆、叶、须根和母根中乌头类总生物碱含量分别为:5.53,3.81,14.77,8.87和4.58mg/g;3种双酯型生物碱含量在不同组织中的含量顺序为:须根〉子根〉母根〉叶片〉茎秆。该研究可为附子资源的综合利用提供科学依据。  相似文献

根据二帖类生物碱和酸性染料溴甲酚绿在酸性缓冲液中形成的络合物能被氯仿提取,并且提取液在415 nm处具有吸收值的原理,运用酸性染料比色法测定了四川境内乌头属植物总生物碱的含量.乌头碱的标准曲线范围是8.8-52.8μg/mL(r=0.999 87,n=6),其平均加样回收率为99.548%.该方法灵敏度高、简便、准确快速.  相似文献

通过高效液相色谱法对传统中药黄连中的主要成分小檗碱的含量进行测定.使用kromasil C18色谱柱(250 mm×4.6 mm,5μm)以乙腈和水作为流动相进行梯度洗脱流速1 mL/min,检测波长为345 nm.盐酸小檗碱浓度在5μg/mL~50μg/mL范围内线性关系良好,平均加样回收率为99.56%,RSD为2.75%.本方法简便、准确、快速、重复性好,可用于黄连中小檗碱的含量测定.  相似文献

探讨了附桂骨痛复方中主要有效成分乌头类总生物碱的质量浓度测定方法。依据乌头类总生物碱与溴甲酚绿在pH=3.5的柠檬酸-磷酸氢二钠缓冲液中反应生成离子对化合物的光谱特征,利用分光光度法于412.5nm波长处测定吸收度并计算含量。结果表明,乌头碱含量在10.12~22.77μg/mL范围内与吸收度线性关系良好(r=0.9996, n=6),其平均加样回收率为99.27%。该法用于测定附桂骨痛复方提取液中乌头类总生物碱的含量准确可靠、简便易行、专属性强。  相似文献

在酸性条件下,高锰酸钾氧化防己诺林碱产生化学发光,对影响化学发光强度的因素进行了实验和探讨,建立了高锰酸钾化学发光体系测定防己诺林碱的新方法.防己诺林碱浓度在0.13~300μg/mL的范围内成良好线性关系,检出限(DL)为0.078μg/mL,相对标准偏差(RSD)为2.6%.该方法已成功用于防己中防己诺林碱的含量测定,回收率为98%.此外借助紫外、荧光和化学发光等光谱手段,对体系的化学发光机理进行了初步探讨.  相似文献

采用β-环糊精敏化荧光法测定黄柏提取液中盐酸小檗碱的含量.盐酸小檗碱是黄柏的主要提取物,与β-环糊精可以形成包合物,产生敏化荧光作用.在7.50 mg/mL β-环糊精水溶液中,选择荧光激发波长(λex)345 nm,发射波长(λem)540 nm,工作曲线线性范围0.10~1.00μg/mL,检出限1.33 ng/mL,测得黄柏中盐酸小檗碱的含量为1.56%.此方法操作简单、准确,对中药中盐酸小檗碱的含量测定具有一定的参考价值.  相似文献

建立HPLC测定鞣酸小檗碱含量的方法,以提高其质量标准。Waters C18(250mm×4.6mm,5μm)色谱柱;以乙腈-0.1%磷酸(每100mL含0.1g十二烷基磺酸钠和0.1mL三乙胺)溶液为流动相;检测波长为345nm;流速1.0mL/min;柱温35℃。鞣酸小檗碱在0.05~0.40μg范围内呈良好的线性关系(R2=0.9992);鞣酸小檗碱平均加样回收率为98.56%,RSD%为2.6%。该方法简单、快速、准确,可用于鞣酸小檗碱的含量测定。  相似文献

建立一种同时测定十味龙胆花颗粒中的有效成分龙胆苦苷和盐酸小檗碱的高效液相色谱法.样品用50%甲醇超声提取,在Zorbax SB-C18柱上分离,以含1%乙酸的甲醇-水为流动相进行梯度洗脱,270 nm紫外检测,外标法定量.龙胆苦苷的线性范围为0.4~40μg/mL(r=0.9993),检出限0.14μg/mL;盐酸小檗碱的线性范围为0.15~30μg/mL(r=0.9999),检出限为0.03μg/mL;盐酸小檗碱的加标回收率为96.0%,相对标准偏差为1.8%;龙胆苦苷的加标回收率为103%,相对标准偏差为1.6%.方法简便快速,重现性好,结果准确,可用于药品十味龙胆花颗粒中胆苦苷和盐酸小檗碱的分析检测.  相似文献

The Williston Basin is a significant petroleum province, containing oil production zones that include the Middle Cambrian to Lower Ordovician, Upper Ordovician, Middle Devonian, Upper Devonian and Mississippian and within the Jurassic and Cretaceous. The oils of the Williston Basin exhibit a wide range of geochemical characteristics defined as "oil families", although the geochemical signature of the Cambrian Deadwood Formation and Lower Ordovician Winnipeg reservoired oils does not match any "oil family". Despite their close stratigraphic proximity, it is evident that the oils of the Lower Palaeozoic within the Williston Basin are distinct. This suggests the presence of a new "oil family" within the Williston Basin. Diagnostic geochemical signatures occur in the gasoline range chromatograms, within saturate fraction gas chromatograms and biomarker fingerprints. However, some of the established criteria and cross-plots that are currently used to segregate oils into distinct genetic families within the basin do not always meet with success, particularly when applied to the Lower Palaeozoic oils of the Deadwood and Winnipeg Formation.  相似文献

The discovery of the prolific Ordovician Red River reservoirs in 1995 in southeastern Saskatchewan was the catalyst for extensive exploration activity which resulted in the discovery of more than 15 new Red River pools. The best yields of Red River production to date have been from dolomite reservoirs. Understanding the processes of dolomitization is, therefore, crucial for the prediction of the connectivity, spatial distribution and heterogeneity of dolomite reservoirs.The Red River reservoirs in the Midale area consist of 3~4 thin dolomitized zones, with a total thickness of about 20 m, which occur at the top of the Yeoman Formation. Two types of replacement dolomite were recognized in the Red River reservoir: dolomitized burrow infills and dolomitized host matrix. The spatial distribution of dolomite suggests that burrowing organisms played an important role in facilitating the fluid flow in the backfilled sediments. This resulted in penecontemporaneous dolomitization of burrow infills by normal seawater. The dolomite in the host matrix is interpreted as having occurred at shallow burial by evaporitic seawater during precipitation of Lake Almar anhydrite that immediately overlies the Yeoman Formation. However, the low δ18O values of dolomited burrow infills (-5.9‰~ -7.8‰, PDB) and matrix dolomites (-6.6‰~ -8.1‰, avg. -7.4‰ PDB) compared to the estimated values for the late Ordovician marine dolomite could be attributed to modification and alteration of dolomite at higher temperatures during deeper burial, which could also be responsible for its 87Sr/86Sr ratios (0.7084~0.7088) that are higher than suggested for the late Ordovician seawaters (0.7078~0.7080). The trace amounts of saddle dolomite cement in the Red River carbonates are probably related to "cannibalization" of earlier replacement dolomite during the chemical compaction.  相似文献

The writer of this paper tries to describe the features of women's language from these perspectives: pronunciation, intonation, diction, subjects, grammar and discourse. From the discussion of the features of women's language, more attention should be paid to language use in social context. 以AC-13级配为基础,将橡胶颗粒代替部分集料掺入混合料中,以低温弯曲试验为评价方法对不同橡胶颗粒掺量下沥青混合料的低温抗裂性进行研究,并引入应变能密度值对混合料的低温抗裂性进行综合评价.试验结果表明:橡胶颗粒沥青混合料试件的破坏微应变均超过2 300,满足冬寒区的技术指标;无论是否掺加橡胶颗粒,随着温度的下降,沥青混合料破坏时的最大弯拉强度增大,弯拉应变降低,劲度模量增大;弯曲应变能密度在胶粒掺量为1%左右时具有较大的弯曲应变能密度值,此时橡胶颗粒沥青混合料具有较好的低温抗裂性.  相似文献

理论推导与室内实验相结合,建立了低渗透非均质砂岩油藏启动压力梯度确定方法。首先借助油藏流场与电场相似的原理,推导了非均质砂岩油藏启动压力梯度计算公式。其次基于稳定流实验方法,建立了非均质砂岩油藏启动压力梯度测试方法。结果表明:低渗透非均质砂岩油藏的启动压力梯度确定遵循两个等效原则。平面非均质油藏的启动压力梯度等于各级渗透率段的启动压力梯度关于长度的加权平均;纵向非均质油藏的启动压力梯度等于各渗透率层的启动压力梯度关于渗透率与渗流面积乘积的加权平均。研究成果可用于有效指导低渗透非均质砂岩油藏的合理井距确定,促进该类油藏的高效开发。  相似文献
