为了研究分析新疆盐湖嗜盐古菌资源,从艾丁湖中分离筛选出一批嗜盐古菌,对其进行了形态特征,生理生化特性研究和分子鉴定。发现其中1株菌(D -20)的生理生化特性和产淀粉酶特性比较突出,经不同温度,不同pH,不同温度下的研究以及16S rDNA 序列的系统发育学研究表明:该菌株为革兰氏染色阴性菌,最适生长温度为37℃,最适生长pH8.0,最适盐浓度15%,能产过氧化氢酶、尿酶,淀粉酶,硝酸盐还原为阳性,不产 H2S ,对青霉素,氯四环素,红霉素,新霉素具有抗性,能利用赖氨酸,蛋氨酸,精氨酸,以果糖,蔗糖和核糖作为碳源。经分子鉴定该菌是 Natrinema属的新成员,定名为 Natrinema aidingensis D -20,Gene Bank登录号为 AY298734。菌株保藏号为12.657。  相似文献

对百合中分离得到的一株放线菌进行传代培养,得到生长良好及性状稳定的实验菌株,然后测定其生理生化特性以及对几种不同病原菌的抑菌活性.该实验菌株最适生长温度为23℃,最适生长pH值为9,产淀粉酶和过氧化氢酶,对绿脓杆菌和肺炎双球菌等具有抑制作用.  相似文献

四川泡菜乳酸菌的分离鉴定及其特性研究   总被引:7,自引:0,他引:7  
以6种四川泡菜汁为材料,分离到11株菌,均为革兰氏阳性菌株,接触酶实验呈阴性.从中筛选出一株产酸能力强、生长速度快的菌株,菌株编号为0708262-1.采用乳酸定性分析测出该菌发酵产物为乳酸.之后经Biolog微生物鉴定仪进一步鉴定,确定该菌株为Lactococcus lactis.该菌株的最适生长pH 为5~7,最适生长温度为30 ℃,最适生长盐浓度为0~5%.  相似文献

红球菌属菌株lawq是专一性脱硫菌,其脱硫酶系由dszA、dszB和dszC组成,其中关键酶是dszC。以脱硫活力作为产脱硫酶系的基准,研究了C、N和S对产酶的影响,实验结果发现甘油是最佳的碳源,适宜的质量浓度为2g/L;氯化铵是合适氮源,适宜质量浓度为2g/L;二苯并噻吩(DBT)是最佳硫源,适宜浓度为0.2mmol/L;以DBT为唯一硫源考察了pH、温度和装液量对lawq生长的影响,结果表明该菌的最适生长pH为7.0,最适生长温度为33℃,溶氧对菌体生长有较大影响。利用该菌体对油品进行脱硫实验,发现32h可以将油品中硫脱除50.47%,说明该菌具有工业应用前景。  相似文献

【目的】筛选分离新的纤维素酶产酶菌株,并对其生长特性和产酶特性进行研究。【方法】从堆肥样品中分离得到一株产纤维素酶菌株,定名为CP1,利用16SrRNA序列对比和Biolog微生物鉴定系统进行鉴定。通过测定菌株生长速率确定其最适生长条件。采用CMC发酵培养基进行纤维素酶的研究,确定其产酶曲线。测定粗酶液最适作用温度和pH值,热稳定性和金属离子对其影响。【结果】经鉴定该菌株为梭形芽孢杆菌(Lysinibacillus fusiformis),其最适生长温度为30℃,最适生长pH值为6。在含CMC-Na的培养基培养,该菌株的生长与产酶同步进行,培养48h菌株生长量达到最大,培养液的CMC酶活力同时达到最大值,为0.46U/mL。该菌株所产的纤维素酶既有酸性CMC酶活力,又有碱性CMC酶活力,并以碱性CMC酶为主,酸性CMC酶的活力只有碱性CMC酶的71.4%。其酸性CMC酶的最适作用pH值为6.0,碱性CMC酶的最适作用pH值为8.0。另外,该菌株所产碱性CMC酶活的最适作用温度为40~50℃,而且0.5%的Cu2+使其酶活降低45%。【结论】菌株CP1为首次报道的梭形芽孢杆菌产纤维素酶新菌株,具有独特的酸碱性环境下的纤维素酶活力。  相似文献

利用双层平板培养技术,从云南腾冲地区高温温泉边酸性水中分离出1株螺旋状铁氧化细菌ML-04,对该菌的理化特性研究结果显示,该菌专性化能自养,可利用亚铁和黄铁矿为能源,不能氧化硫磺、四硫酸盐、硫代硫酸盐,最适生长温度40℃,最适生长pH2.5.对砷黄铁矿的浸矿实验表明,ML-04菌株可以有效浸出矿粉中的铁和硫元素.  相似文献

一株来自深海沉积物的低温、嗜压菌的分离鉴定   总被引:1,自引:0,他引:1  
从西太平洋5 060 m深的海底沉积物中分离到一组低温菌,对其中一株细菌WP02-2-25进行了生理生化和分子水平鉴定.结果表明该菌革兰氏染色阴性;温度生长范围在28℃以下,最适生长温度15℃,pH耐受范围为4.5~9.5;最适pH值7.5.能够利用D-葡萄糖、甘油和麦芽糖作为唯一碳源生长并产酸,氧化酶和接触酶呈阳性,可以水解明胶和还原硝酸盐,但不能继续还原亚硝酸盐,产H2S气体.依据其生理生化性质、系统发育学、16SrDNA序列同源性分析初步鉴定该菌属于深海中特有的菌属Moritella属,与嗜压菌Moritella japonica种有99.47%的同源性.生理生化指标表明,这两株菌不完全相同,Moritella japonica可以利用果糖、不能利用麦芽糖,而WP02-2-25正好相反.另外,两者的最适生长温度也不一致,前者属于嗜冷菌的范畴;后者属于耐冷菌.  相似文献

采用形态学和系统发育分析对一株嗜盐古菌DSFD111进行了分类学鉴定.分离到的DSFD111菌株为革兰氏阴性球菌,不产芽胞,细胞内含类脂粒.最适生长温度为37℃,最适pH为7,最适生长NaCl浓度为4.3mol/L,表明该菌株属于极端嗜盐菌.根据16S rRNA基因序列的系统发育分析,DSFD111菌株归于盐盒菌属(Haloarcula),与最近缘的Haloarcula argentinensis的相似度为99%.  相似文献

从不同生物环境中分离紫色非硫菌,测定其类胡萝卜素含量,筛选出富含类胡萝卜素的菌株.对该菌株的形态学特征、碳氮源利用情况、最适生长pH值及盐度等生物学特征进行研究;利用PCR技术扩增其16S rRNA基因,测序后进行16S rRNA基因序列分析,研究其分类地位.结果:分离自暨南大学明湖底泥的菌株R2,在所分离的8株紫色非硫菌中,类胡萝卜素质量分数最高,为4.83 mg/g(干菌体),该菌株符合红游动菌属(Rhodoplanes)的生物学特性,其最适生长pH为7.0,盐度为0.2%;16S rRNA基因的分子系统学分析表明,该菌株与红游动菌属的Rhodoplanessp.HA17相似性为99.4%.可确定筛选出一株高产类胡萝卜素的菌株,该菌株属于红游动菌属(Rhodoplanes).  相似文献

Language is a means of verbal communication. People use language to communicate with each other. In the society, no two speakers are exactly alike in the way of speaking. Some differences are due to age, gender, statue and personality. Above all, gender is one of the obvious reasons. The writer of this paper tries to describe the features of women's language from these perspectives: pronunciation, intonation, diction, subjects, grammar and discourse. From the discussion of the features of women's language, more attention should be paid to language use in social context. The discovery of the prolific Ordovician Red River reservoirs in 1995 in southeastern Saskatchewan was the catalyst for extensive exploration activity which resulted in the discovery of more than 15 new Red River pools. The best yields of Red River production to date have been from dolomite reservoirs. Understanding the processes of dolomitization is, therefore, crucial for the prediction of the connectivity, spatial distribution and heterogeneity of dolomite reservoirs.The Red River reservoirs in the Midale area consist of 3~4 thin dolomitized zones, with a total thickness of about 20 m, which occur at the top of the Yeoman Formation. Two types of replacement dolomite were recognized in the Red River reservoir: dolomitized burrow infills and dolomitized host matrix. The spatial distribution of dolomite suggests that burrowing organisms played an important role in facilitating the fluid flow in the backfilled sediments. This resulted in penecontemporaneous dolomitization of burrow infills by normal seawater. The dolomite in the host matrix is interpreted as having occurred at shallow burial by evaporitic seawater during precipitation of Lake Almar anhydrite that immediately overlies the Yeoman Formation. However, the low δ18O values of dolomited burrow infills (-5.9‰~ -7.8‰, PDB) and matrix dolomites (-6.6‰~ -8.1‰, avg. -7.4‰ PDB) compared to the estimated values for the late Ordovician marine dolomite could be attributed to modification and alteration of dolomite at higher temperatures during deeper burial, which could also be responsible for its 87Sr/86Sr ratios (0.7084~0.7088) that are higher than suggested for the late Ordovician seawaters (0.7078~0.7080). The trace amounts of saddle dolomite cement in the Red River carbonates are probably related to "cannibalization" of earlier replacement dolomite during the chemical compaction.  相似文献

理论推导与室内实验相结合,建立了低渗透非均质砂岩油藏启动压力梯度确定方法。首先借助油藏流场与电场相似的原理,推导了非均质砂岩油藏启动压力梯度计算公式。其次基于稳定流实验方法,建立了非均质砂岩油藏启动压力梯度测试方法。结果表明:低渗透非均质砂岩油藏的启动压力梯度确定遵循两个等效原则。平面非均质油藏的启动压力梯度等于各级渗透率段的启动压力梯度关于长度的加权平均;纵向非均质油藏的启动压力梯度等于各渗透率层的启动压力梯度关于渗透率与渗流面积乘积的加权平均。研究成果可用于有效指导低渗透非均质砂岩油藏的合理井距确定,促进该类油藏的高效开发。  相似文献

There are numerous geometric objects stored in the spatial databases. An importance function in a spatial database is that users can browse the geometric objects as a map efficiently. Thus the spatial database should display the geometric objects users concern about swiftly onto the display window. This process includes two operations:retrieve data from database and then draw them onto screen. Accordingly, to improve the efficiency, we should try to reduce time of both retrieving object and displaying them. The former can be achieved with the aid of spatial index such as R-tree, the latter require to simplify the objects. Simplification means that objects are shown with sufficient but not with unnecessary detail which depend on the scale of browse. So the major problem is how to retrieve data at different detail level efficiently. This paper introduces the implementation of a multi-scale index in the spatial database SISP (Spatial Information Shared Platform) which is generalized from R-tree. The difference between the generalization and the R-tree lies on two facets: One is that every node and geometric object in the generalization is assigned with a importance value which denote the importance of them, and every vertex in the objects are assigned with a importance value,too. The importance value can be use to decide which data should be retrieve from disk in a query. The other difference is that geometric objects in the generalization are divided into one or more sub-blocks, and vertexes are total ordered by their importance value. With the help of the generalized R-tree, one can easily retrieve data at different detail levels.Some experiments are performed on real-life data to evaluate the performance of solutions that separately use normal spatial index and multi-scale spatial index. The results show that the solution using multi-scale index in SISP is satisfying.  相似文献
