细菌F质粒转移实验的有效方法的研究

摘    要:本文报道了一种简易有效的F′质拉转移的方法。这种方法仅用了包含链霉素(str)和氯霉素(cm)的选择性培养基,就能选出转移子。其目的是希望把F′从供体转移给受体,供体不抗str,受体不抗cm,把杂交中的混合菌涂布在LB cm str平皿上,供体和受体均被杀死,只有转移子才能生长。选得的转移子是:F′lac~ ::Tn9 pro~ /Δ(lac pro)gal E str~r重组体。

Abstract:This paper reports a simple efficient method of F' plasmid transfer experiments. In this method, only by using two selectivity medium cnotaining streptomycin and chloromycetin can the transfer factors be selected. In the cross of strain XAC(F' lac Tnq pro )/A(lac pro)rif nal (cm') with strain F-(lac pro) galE strr, to transfer an F' lac Tn9 pro from the XAC to the F- strain is the goal to be attained. The mating mixture can be plated on plates with streptomycin and chloromycetin. All of XAC cells are killed by the streptomycin. All of F- cells are killed by the chloromycetin. Since the donor and the recipient eannot grow, only those transfer factor cells which have received the F' lac Tn 9 pro from XAG will form clones on this selection plate. Thus, the selection is for F' lac Tn9 pro /?(lac pro) gal E strr Figure 4. It is a better method in the transfer experiments of plasmid.
