正丙醇、铜离子、烟酰胺对红霉素合成的作用

摘    要:在摇瓶培养过程中,发酵初期加入正丙醇可使红霉素的效价提高49.31%,正丙醇提高了丙酸激酶的比活力,表现出对该酶诱导作用,而对丙酰CoA合成酶比活力无影响。加入铜离子和烟酰胺可使红霉素效价分别提高24%和179/5。用HPLC测定发酵液中有机酸含量,分析测定结果认为发酵过程可分为3个阶段:积累期、转折期和生产期。

Abstract:Addition of propanol during erythromycin fermentation with in shake flask improved the titer by 49.3%.Propanol was found to increase the specific activity of propionate kinase,but it had no(influence) on the specific activity of propionyl-CoA synthetase.Addition of Cu~(2+) and niacinamide,(increased erythromycin) titer by 24% and 17%,respectively.Using HPLC,organic acid in the broth were measured;it was suggested that the fermentation process can be divided into three phases: accumulative phase,transitional phase and production phase.In production phase,the productivity of erythromycin was the highest.
