摘    要:本研究以出芽短梗霉发酵液为原料提取黑色素,在单因素实验基础上选取实验因素与水平,用二水平实验确定影响黑色素色价的主要因素,然后根据Box-Behnken中心组合实验设计原理采用三因素三水平的响应面分析法,得到最佳提取工艺为:酸沉时间90 min、乙醇添加量93.17%、乙醇浓度90.42%。在此条件下提取黑色素色价可达44.8μ/mL。

Abstract:The extraction of melanin from Aureobasidium pullulans fermented liquid was studied in this paper. On the basis of single factor experiments, the main factors that influence the valeur of melanin were determined by two-level experiment, and then the optimum conditions for the extraction of the melanin from A. pullulans fermented liquid were obtained through a three-variable, three-level Box--Benhnken center--united experiment design and response surface methodology. The results showed that the optimum conditions were as follows: acid sinking time 90 rain, ethanol add volume 93.17 % and ethanol concentration90.42 %. Under such conditions, the maximal valeur of melanin was up to 44.8 μ/mL.
