菜豆种质盐溶蛋白遗传多态性分析

摘    要:以39个菜豆品种和1个杂交后代为材料,通过SDS-PAGE技术获得了种子盐溶蛋白的亚基分布电泳图谱,比较了分布差异,并分析了其遗传表现.结果表明:40个供试种质的盐溶蛋白带型差异显著,电泳共分离出27条迁移率不同的条带,其中有4条保守带(均为100%表达),分子质量分别为99.039、58.267、25.234、15....

Abstract:The seeds of 39 common bean cultivars and one F hybrid were used to analyse their seed salt-soluble proteins profiles by SDS-PAGE technique in order to illuminate genetic polymorphism and the inheritance patterns among them based on varied subunits distribution.The experimental results showed that the salt-soluble proteins profiles among 40 specimens varied significantly,27 salt-soluble proteins bands were separated with different mobility,including 4 bands shared by all specimens with the molecular weight 99.039,58.267,25.234,15.776 ku,respectively,and 23 bands were polymorphism.The ratio of polymorphic site(%) was 85.18%.The sort time of the 23 bands averagely was 23.625(ranged from 1 to 39),the coefficient of variation was 40.85%.The cultivar WS4 had a unique subunit 38.685 ku.In the polymorphic bands,averagely showed polymorphic amplified bands 18.925(amplitude of variation was 9~19),the coefficient of variation was 17.5%.The results suggest that these cultivars have a high level of genetic homogeneity,meanwhile variations have been formed in their evolutionary history.Their genetic distance ranged from 0.027 to 0.420,with a mean value of 0.147.The specimens could be divided into 10 major groups by cluster analysis,in the Nei's genetic distance of 0.210,which had a strong consistency with cultivars morphological features.Therefore the seed salt-soluble proteins marker could be used to demonstrate the polymorphisms of common bean resources and to identify cultivar or hybrid as well as to make sure if the cross hybridization was successful.
