聚酰胺纯化刺玫果总黄酮的工艺研究

摘    要:探索聚酰胺动态吸附-解吸纯化刺玫果总黄酮的工艺条件.以总黄酮的比吸附量、吸附率及比解吸量、解吸率为考察指标,通过单因素实验对刺玫果总黄酮的纯化工艺进行优化.结果表明聚酰胺纯化刺玫果总黄酮的最佳工艺条件:上样液总黄酮的质量浓度为0.9 mg/mL、上样量为19.5 mL/g(上样液体积/聚酰胺质量)、上样液的pH值为2~4、吸附流速为1.5~2.0 mL/min、解吸液为70%乙醇水溶液、解吸流速为1.0~1.5 mL/min、解吸液的pH值为8~9、解吸液用量为12 mL/g(解吸液体积/聚酰胺质量).在上述纯化条件下,聚酰胺对刺玫果总黄酮的比吸附量平均为11.16 mg/g、比解吸量平均为10.20 mg/g、解吸率平均为91.40%,干浸膏中总黄酮含量由26.32%提高到75%以上.研究结果为刺玫果的深入开发提供了依据.

Abstract:The purification technology of total flavonoids (TFs) in Rosa davurica Pall. with polyamide resin methods by dynamic adsorption-desorption were explored. The purification process of TFs was optimized by single factor and orthogonal test using absorbance, adsorption rate, desorption quantity and desorption rate as indexes for investigation. The results showed that the optimum purification conditions of TFs in Rosa davurica Pall. were as follows for sample solution, the mass concentration of total flavonoids, loading quantity, pH value and adsorption velocity were 0.9 mg/mL, 19.5 mL/g, 2~4 and 1.5~2.0 mL/min, respectively; 70% (volume fraction) ethanol solution was used as the eluent, of which the desorption velocity, pH value and quantity were 1.0~1.5 mL/min, 8~9 and 12 mL/g, respectively. Under the above purification conditions, the average absorbance of polyamide resin to total flavonoids was 11.159 mg/g, while the average desorption quantity to total flavonoids was 10.204 mg/g. The average desorption rate was 94.44% and the purity of TFs in Rosa davurica Pall. extracts elevated from 26.32% to above 75%. The results provided reference for further development and utilization of Rosa davurica Pall.
