Zn(Ⅱ)、Mn(Ⅱ)竞争结合人血清蛋白研究

摘    要:用平衡透析法在生理pH值(7.43)条件下,研究了等摩尔比的Zn(Ⅱ)与Mn(Ⅱ)离子竞争结合人血清白蛋白(HSA)。Scatchard图分析表明,Zn(Ⅱ)与Mn(Ⅱ)在HSA中没有共同的强结合位点,在竞争和非竞争结合时,Zn(Ⅱ)的强结合位点数目均为1,Mn(Ⅱ)的强结合位点数目均为2。Hill图分析说明Zn(Ⅱ)与Mn(Ⅱ)之间具有一定的正协同效应。

Abstract:Equilibrium dialysis was used to investigate the competition binding for human serum albumin (HSA) between equimolar Zn(Ⅱ) and Mn (Ⅱ) ions at physiological pH (7. 43). The results of Scatchard plots indicated that HSA possesses two strong binding sites for Mn (Ⅱ),and one strong binding site for Zn(Ⅱ). The competitive bindings have no insignificant effect on the number of strong binding sites for Zn (Ⅱ) and Mn (Ⅱ),which suggested that there are not any common strong binding sites for Zn (Ⅱ) and Mn (Ⅱ) in HSA. Whereas,the competitive binding increased the number of weak binding sites of Zn(Ⅱ) and Mn(Ⅱ) in serum albumins. The analyses of Hill plots also showed that the cooperative effect existed between Zn (Ⅱ) and Mn (Ⅱ) ions during the process of competitive binding with HSA.
