摘    要: 在获得禽流感病毒型及亚型特异性单克隆抗体的基础上,研究建立型及亚型特异性免疫酶染色技术,用于检测或鉴定禽流感病毒.优化反应条件,确定单克隆抗体及辣根过氧化物酶标记二抗的最佳工作浓度,进行敏感性、特异性、重复性及稳定性分析,并与病毒分离鉴定比较.结果表明该方法敏感、特异,具有良好的重复性和稳定性,可用于检测临床组织样品、鸡胚及细胞培养物中的禽流感病毒.

Abstract:Based on obtaining avian influenza type and subtype-specific monoclona l antibody, the type and subtype specific immunoperoxidase staining techniques w ere developed for detection of avian influenza virus. The reactive condition an d the working titers of monoclonal antibodies and HRP-labeled second antibody wer e optimized, and the assay sensitivity, specificity, repeatability, stability we re analyzed and compared with virus isolation and identification. The results i n dicated that immunoperoxidase staining techniques were sensitive, specific, with high repeatability and stability for detection of avian influenza virus in clinical tissue samples, embryo and cell culture tissues.
