香菇多糖联合依地酸钙钠对铅中毒小鼠学习记忆功能及海马NO,nNOS的影响

摘    要:将50只小鼠随机分成空白对照组、模型对照组、阳性对照组、香菇多糖低剂量组、香菇多糖高剂量组5个组,用Morris水迷宫检测小鼠学习记忆功能,微分电位溶出法测定血铅含量,硝酸还原酶法检测NO含量,免疫组化方法观察nNOS阳性细胞表达.结果表明,香菇多糖高剂量组能显著改善铅中毒小鼠的学习记忆功能.与模型对照组相比,香菇多糖高剂量组能显著降低血铅浓度(P<0.01),提高海马NO含量(P<0.01),且增强nNOS阳性神经元表达CA1(P< 0.05),CA3(P<0.01).这提示香菇多糖联合依地酸钙钠能改善铅中毒小鼠的学习记忆功能,其作用机制可能与海马NO含量增高及nNOS神经元表达增强有关.

Abstract:The 50 mice were randomly divided into five groups,blank group,negative control group,positive control group,lentinan low dose group and high dose group.Morris water maz was used to monitor the learning and memory ability.The lead concentrations of blood were measured by electric potential stripping method.In the mean time,The content of NO was tested by nitrate reductase method.The expression of nNOS positive cells were observed with immunohistochemistry.The results indicated that compared with that of negative control group,the learning and memory ability of lead poisoning mice could be markedly improved by lentinan high dose group lentinan high dose group,and the lead concentrations of blood could be significantly decreased(P<0.01).The hippocampal intracellular NO content was obviously increased(P<0.01).The nNOS positive cells of hippecarnpus were increased CA1(P<0.05),CA3(P<0.01).Lentinan and calcium disodium edetate could improve the learning and memory on lead poisoning mice.The mechanism may be related with increasing NO content and the expression of nNOS positive cells in hippoeampus.
