柄果海桐化学成分研究

摘    要: 在细胞毒试验结果指导下,采用萃取、柱层析、重结晶等方法,同步对柄果海桐(Pittosporum podocarpum Gagnep)枝叶粗提物的有效部位进行化学成分分离纯化得9个单体,通过理化数据测定及波谱数据分析鉴定了结构,分别是kielcorin(1),2,3-二甲氧基(口山)酮(2,3-dimethoxyxanthone,2),槲皮素-3,7-二甲氧基(quercitrin-3,7 -dimethoxy,3),木犀草素(luteolin,4),羽扇豆醇(lupeol,5),蜡酸(cerotic acid,6),二十八酸(1-octacosanic acid,7),β-谷甾醇(β-sitosterol,8),胡萝卜甙(daucosterol,9).化合物1~9均为首次从该植物中分离得到.细胞毒试验发现其石油醚、氯仿及乙酸乙酯提取物有细胞毒活性,其IC50分别为6.5,3.5,5.0 μg/mL.

Abstract:The chemical constituents of Pittosporum podocarpum Gagnep were studied by methods of extraction,column chromatography,crystallization and so on.Nine compounds were isolated from this plant by column chromatogramphies of sephadex LH-20 and silica gel.Their structures were identified by analyses of their physical-chemical properties and spectral datas.All these compounds were isolated for the first time from this plant.These are:kielcorin(1);2,3-dimethoxyxanthone(2);quercitrin-3,7–dimethoxy(3);luteolin(4);lupeol(5);cerotic acid(6);1-octacosanic acid(7);β-sitosterol(8);daucosterol(9).Cytotoxicity screening of extracts from Pittosporum podocarpum Gagnep was carried out by HL60 cell.Three extracting fractions of petroleum ether,chloroforn and ethyl acetate possess cytotoxic activities and the IC50 are 6.5,3.5 and 5.0 μg/mL for HL60,respectively.
