脐带血T淋巴细胞集落培养和亚群的关系

摘    要:应用细胞培养和间接免疫荧光法测定脐带血,成人外周血,成人骨髓的T淋巴细胞集落形成情况及培养前后淋巴细胞亚群的变化。结果表明:脐带血的CFU-TL显低于成人外周血(P〈0.01),略低地成人骨髓(P〈0.05);培养前脐带因的CO3、C含量均显低于成人外周血(P〈0.01),成成人骨髓相近似。脐带血的CD8细胞含量与成人外周血和成人骨髓相似(P〈0.05);培养后的CFU-TL中的CT3细胞含量

Abstract:T lymphocyte colony form ation of human cord blood, adult peripheral blood (PB) and adult bone marrow (BM) was studied by means of cell culture and change in per-centage of T lymphocyte subsets before and after culture. The results showed that the number of CFU-TL of the cord blood was much lower than that of the normal PB(P<0.001) and slightly lower than of BM (P<0.05). The percentages of CD + 3 and CD + 4 cells in the cord blood before culture were much lower than those of normal PB (P<0.01), but was approximated to those of BM (P>0.05). The percentage of CD + 8 cells in the cord bllod was similar to that of the normal PB and BM (P>0.05) both. No significant difference was found in the percentage of CD + 3 cells in CFU-TL from the cord blood before culture and those of the normal PB and MB, but the per-centage of CD + 4 cells in cord blood was much lower than that either of the normal PB or BM (P<0.05) on the contrary the percentage of CD + 8 cells in the cord blood after culture was much higher than that form both the normal PB (P<0.01) and BM (P<0.05), thus, the ratio of CD + 4/CD + 8 in the cord blood after culture was much lower than that of either the adult PB or BM. In adclitron, the total percentage of CD + 4 and CD + 8 cells in cord blood or in BM before culture was higher than that of CD + 3 cells, This might be due to double labeling of the cells.
