氨基化的介孔二氧化硅纳米材料与pDNA作用

摘    要:采用共沉淀法合成高度有序、分散性良好的氨基化介孔二氧化硅纳米材料(Am-MSNs),并研究其吸附和保护质粒DNA(pDNA,PEGFP-N3)的性质,测定样品材料对A549和HeLa细胞的生物相容性.实验结果表明:Am-MSNs能有效吸附PEGFP-N3,并保护PEGFP-N3免受限制性内切酶的消化;Am-MSNs对A549和HeLa细胞表现出较高的生物相容性,当材料的质量浓度为125μg/mL时,两种细胞的存活率均约为100%.

Abstract:This work evaluated the adsorption and protection capacities of amino functionalized mesoporous silica nanoparticles (Am-MSNs) for pDNA (PEGFP-N3). The in vitro biocompatibilities of Am MSNs to A549 and Hela cells were also tested, respectively. The results reveal that the Am-MSNs
exhibit good adsorption capacity for PEGFP N3. Moreover, the material can also protect PEGFP N3 from enzymatic degradation completely. In addition, the
sample presents high in vitro biocompatibilities to HeLa and A549 cells.
