新型抗病毒药物喷昔洛韦的合成

摘    要:以2-羟甲基-1,4-丁二醇为原料,采用2-氨基-6-氯嘌呤位置有择烷基化反应合成新型抗病毒药物喷昔洛韦,总收率27%。关键中间体及最终化合物的结构经元素分析、质谱、核磁共振氢谱及碳谱得到确证。合成中避免使用高温、高压及腐蚀性试剂,适于工业化生产。

Abstract:Penciclovir, 9 4 hydroxy 3 (hydroxymethyl)but 1 yl]guanine was synthesized by means of the regioselective alkylation of 2 amino 6 chloropurine with 5 (2 iodoethyl) 1,3 dioxan as a pivotal step The total yield was 27% The structures of the intermediates and the target molecule were confirmed by MS, elementary analysis, 1H NMR and 13 C NMR spectra in our experiments The route without using highly toxic reagents and high pressure and high temperature may be applied to industrial production.
