噬菌体展示禽流感病毒抗原变异性基因片段文库的构建

构建T7噬菌体展示禽流感病毒抗原变异性基因片段文库. 首先， 从Gene Bank中查找筛选禽流感病毒抗原变异性基因, 将其截短、 修饰、 简并后得到禽流感病毒抗原变异性基因微阵列. 其次， 将合成的禽流感病毒抗原变异性基因片段文库扩增、 酶切, 链接到双酶切后的T7噬菌体载体基因上, 构成重组噬菌体DNA. 最后， 重组噬菌体DNA经体外包装和扩增, 得到T7噬菌体展示文库, 并进行T7噬菌体展示文库滴度、 重组率和免疫活性测定. 实验结果表明, 从Gene Bank中查找、 筛选、 剪切和修饰共获得96 258条序列构建T7噬菌体展示文库, 原始文库滴度为3.6×107个菌落/mL, 重组率大于90%. 用禽流感病毒H5N1抗体进行捕获, 经聚合酶链式反应(PCR)鉴定, 得到理想目的条带, 证明噬菌体表面展示蛋白具有抗原活性, 可用于禽流感病毒感染患者的快速检测及抗原表位筛选.     

三球式和转台式角动量守恒演示器的研制与应用

本文详细介绍了笔者研制的三球式和转台式两种新型角动量守恒演示器的结构、特点和使用方法。     

微波辐射合成7-羟基-4-甲基香豆素

以浓硫酸为催化剂，采用微波辐射技术，由间苯二酚与乙酰乙酸乙酯缩合制备7－羟基－4－4甲基香豆素。结果表明，微波辐射合成7－羟基－4－4甲基香豆素的最以应条件为：间苯二酚5.5g，乙酰乙酸乙酯6.8ml，浓硫酸1.5ml，微波功率240W，微波辐射时间6min，产率84．1％，不采用微波辐射，反应时间大于10h，产率78％，红外光谱分析表明，7－羟基－4－4甲基香豆素的特征峰与7－羟基－4－甲基香豆素的各官能团的吸收峰吻合，测定7－羟基－4－甲基香豆素的熔点为189－192℃。     

MPEG-7标准及其应用

MPEG-7是国际标准化组织即将推出的新一代多媒体国际标准,该标准主要用于对各种不同类型的多媒体信息进行标准化描述,并将该描述与所描述的内容相联系,以实现对多媒体信息快速有效的搜索.在未来的多媒体信息检索服务中,MPEG-7将发挥主导作用.本文主要从标准的目标、体系结构及应用等方面对MPEG-7进行了描述.     

关于C_(11)A_7·CaF_2稳定性的研究

本文采用X射线衍射分析方法,研究了影响C_(11)A_7·CaF_2稳定性的因素,结果表明C_(11)A_7·CaF_2在1450℃以内,能够稳定存在;Mgo,K_2O对C_(11)A_7·CaF_2稳定性没有影响;Na_2O、P_2O_5和CaO能与C_(11)A_7·CaF_2发生反应、分别生成少量NC_6A_3、Ca_5F(PO_4)_3和C_3A,但在硅酸盐水泥熟料的正常煅烧制度下,反应程度较低,对C_(11)A_7·CaF_2稳定性不大.熟料中铝酸盐相存在的形式,主要取决于氟在熟料中的分布,CaF_2掺量较低,温度超过1350℃,C_(11)A_7CaF_2消失,生成C_3A;CaG_2掺量较高时,温度高达1450℃,熟料中的铝酸盐相仍为C_(11)A_7·CaF_2,而不生成C_2A.     

试析当前美国实力地位的变化

当前,美国深陷伊拉克战争泥潭,有人提出美实力地位正发生由盛转衰的变化.通过分析可知美国硬实力仍举世无双,软实力在世界各地仍具有重要影响,而美国创新机制和自我修复能力为美提供巨大的力量来源.在未来较长时间内美国仍将保持一超独霸地位.另一方面,世界各国对美国更加缺乏信任,伊拉克战争大大销蚀美实力等,都构成了美国实力的局限.鉴此,美国不得不加强国际合作,承担相应国际责任,外交政策最终向现实主义回归.     

The concentration quenching characteristics of ~5D_3-~7F_J and ~5D_4-~7F_J (J=0―6) transitions of Tb~(3 ) in YBO_3:Tb~(3 ) phosphor

The photoluminescence quenching behaviors of 5D3-7FJ and 5D4-7FJ (J = 0―6) transitions of Tb3 in YBO3:Tb under 130―290 nm excitation were systematically investigated. The results revealed that the quenching concentrations of both 5D3-7FJ and 5D4-7FJ transitions of Tb3 in YBO3:Tb were mainly de-pendent on excitation wavelength. Particularly,the quenching concentrations of 5D4-7FJ transitions of Tb3 under 130―290 nm excitation were correlated with excitation bands of YBO3:Tb. The quenching concentrations of 5D3-7FJ transitions remained at low concentration (2%) under 186―290 nm excitation and then increased gradually with energy of incoming excitation photon when excited at 130―186 nm. This dependence should be involved in their excitation mechanisms and quenching pathway in particular excitation region.     

美国初中级教育中对自然科学教师的培养及职业标准

对自然科学教师的培养及职业标准的研究是当代世界各国教育改革的焦点之一。本文试就美国对初中级教育中对自然科学教师的职业准备及其培养标准加以介绍和描述,特别是对美国七十年代以来对自然科学教育由重视到不重视再到重视的具体过程分析,以此为我国自然科学教师的培养提供借鉴作用。     

Selective protection of cells against X-irradiation. Isoproterenol protects only those cells that possesβ-adrenoreceptors

Summary In mixed culture of Chinese hamster fibroblatst, clone 431, and transformed murine L fibroblasts, clone B-82, isoproterenol was found to protect only 431 cells against ionizing radiation. It was shown that 431 cells, in contrast to B-82 cells, possess-adrenoreceptors, and the radioprotective effect of isoproterenol can be realized only if this agent interacts with-adrenoreceptors coupled with the cAMP system. Since malignization often causes the disappearance of-adrenergic and other hormone receptors, the combined culturing and irradiation of the cells studied can be regarded as a model of the growth of malignant cells (B-82) among normal tissue cells (431 cells) under conditions of radiation therapy. A possibility of selective protection against radiation damage of normal tissue cells, with retention of the former radiosensitivity of tumor cells, is discussed.     

Structure of the human CLC-7/Ostm1 complex reveals a novel state

CLC-7 functions as a Cl?/H+ exchanger in lysosomes. Defects in CLC-7 and its β-subunit, Ostm1, result in osteopetrosis and neurodegeneration. Here, we present the cryogenic electron microscopy (cryo-EM) structure of the human CLC-7/Ostm1 complex (HsCLC-7/Ostm1) at a resolution of 3.6 ?. Our structure reveals a new state of the CLC-7/Ostm1 heterotetramer, in which the cytoplasmic domain of CLC-7 is absent, likely due to high flexibility. The disordered cytoplasmic domain is probably not able to restrain CLC-7 subunits and thus allow their relative movements. The movements result in an approximately half smaller interface between the CLC-7 transmembrane domains than that in a previously reported CLC-7/Ostm1 structure with a well-folded cytoplasmic domain. Key interactions involving multiple osteopetrosis-related residues are affected by the interface change.
