Myelin-associated glycoprotein (MAG) is a quantitatively minor component in both peripheral and central myelin sheaths that is thought to have a role in cell-cell interactions within the nervous system. We show here that a mouse monoclonal antibody, HNK-1, which is directed against human natural killer cells also recognizes an antigenic determinant of human central and peripheral nervous system white matter by immunoperoxidase staining of tissue sections. Immunoblot analysis of myelin proteins and purified extracted MAG indicates that the antigen recognized is MAG.  相似文献

Caloric restriction extends lifespan in numerous species. In the budding yeast Saccharomyces cerevisiae this effect requires Sir2 (ref. 1), a member of the sirtuin family of NAD+-dependent deacetylases. Sirtuin activating compounds (STACs) can promote the survival of human cells and extend the replicative lifespan of yeast. Here we show that resveratrol and other STACs activate sirtuins from Caenorhabditis elegans and Drosophila melanogaster, and extend the lifespan of these animals without reducing fecundity. Lifespan extension is dependent on functional Sir2, and is not observed when nutrients are restricted. Together these data indicate that STACs slow metazoan ageing by mechanisms that may be related to caloric restriction.  相似文献

The mechanism of tumour cell destruction by natural killer (NK) cells or other lymphocytes is not understood. NK cells appear to represent a primitive anti-tumour surveillance system more analogous to macrophages than lymphocytes. Free oxygen radicals (O-2, OH) and H2O2 are thought to be involved in cell destruction by macrophages and therefore we looked for similar cytocidal intermediates of oxygen in NK cells. These highly reactive molecular species can easily be detected in the presence of luminol by the emission of light. We show here that highly enriched human NK cells respond to NK-sensitive but not NK-insensitive tumour cells with a rapid burst of oxygen metabolites as detected both by chemiluminescence and cytochrome c reduction. Agents which can prevent chemiluminescence and cytochrome c reduction, such as superoxide dismutase (SOD), reduced NK-mediated cytolysis and agents which increased chemiluminescence, such as interferon, also increased NK-mediated cytolysis. These results suggest that the production of oxygen species may be the earliest event to occur in the NK cell following tumour cell contact, and these products are involved in NK-mediated cytolysis.  相似文献

Anomalous X-ray pulsars (AXPs) are slowly rotating neutron stars with very bright and highly variable X-ray emission that are believed to be powered by ultra-strong magnetic fields of >10(14) G, according to the 'magnetar' model. The radio pulsations that have been observed from more than 1,700 neutron stars with weaker magnetic fields have never been detected from any of the dozen known magnetars. The X-ray pulsar XTE J1810-197 was revealed (in 2003) as the first AXP with transient emission when its luminosity increased 100-fold from the quiescent level; a coincident radio source of unknown origin was detected one year later. Here we show that XTE J1810-197 emits bright, narrow, highly linearly polarized radio pulses, observed at every rotation, thereby establishing that magnetars can be radio pulsars. There is no evidence of radio emission before the 2003 X-ray outburst (unlike ordinary pulsars, which emit radio pulses all the time), and the flux varies from day to day. The flux at all radio frequencies is approximately equal--and at >20 GHz XTE J1810-197 is currently the brightest neutron star known. These observations link magnetars to ordinary radio pulsars, rule out alternative accretion models for AXPs, and provide a new window into the coronae of magnetars.  相似文献
