Liquid crystal phase transitions in suspensions of polydisperse plate-like particles

Colloidal suspensions that form periodic self-assembling structures on sub-micrometre scales are of potential technological interest; for example, three-dimensional arrangements of spheres in colloidal crystals might serve as photonic materials, intended to manipulate light. Colloidal particles with non-spherical shapes (such as rods and plates) are of particular interest because of their ability to form liquid crystals. Nematic liquid crystals possess orientational order; smectic and columnar liquid crystals additionally exhibit positional order (in one or two dimensions respectively). However, such positional ordering may be inhibited in polydisperse colloidal suspensions. Here we describe a suspension of plate-like colloids that shows isotropic, nematic and columnar phases on increasing the particle concentration. We find that the columnar two-dimensional crystal persists for a polydispersity of up to 25%, with a cross-over to smectic-like ordering at very high particle concentrations. Our results imply that liquid crystalline order in synthetic mesoscopic materials may be easier to achieve than previously thought.     

Cardiolipin, the heart of mitochondrial metabolism

Cardiolipin is a unique phospholipid, which is almost exclusively localized in the mitochondrial inner membrane where it is synthesized from phosphatidylglycerol and cytidinediphosphate-diacylglycerol. After primary synthesis, the mature acyl chain composition of cardiolipin is achieved by at least two remodeling mechanisms. In the mitochondrial membrane cardiolipin plays an important role in energy metabolism, mainly by providing stability for the individual enzymes and enzyme complexes involved in energy production. Moreover, cardiolipin is involved in different stages of the mitochondrial apoptotic process and in mitochondrial membrane dynamics. Cardiolipin alterations have been described in various pathological conditions. Patients suffering from Barth syndrome have an altered cardiolipin homeostasis caused by a primary deficiency in cardiolipin remodeling. Alterations in cardiolipin content or composition have also been reported in more frequent diseases such as diabetes and heart failure. In this review we provide an overview of cardiolipin metabolism, function and its role in different pathological states. Circadian rhythmicity is involved in photoperiodic time measurement in the aphidMegoura viciae

The photoperiodic clock in the vetch aphidMegoura viciae is generally accepted to be based on a non-circadian mechanism or hourglass, as no evidence has been found for the involvement of the circadian system in the photoperiodic response. By using a recently-devised protocol which discriminates between single and repeated night length measurement, we demonstrate here that long-night measurement inMegoura is executed in a repetitive way, and thus that its photoperiodic clock is based on a circadian oscillator after all. However, it is also apparent that the determination of short nights is not repetitive.Dedicated to our dear friend, the late Professor A. D. (tony) Lees who, sadly, died before we had a chance to discuss the data reported here.     

Natural history of the fireflies of the Serra dos Órgãos mountain range (Brazil: Rio de Janeiro) – one of the ‘hottest’ firefly spots on Earth,with a key to genera (Coleoptera: Lampyridae)

ABSTRACT

Fireflies (Coleoptera: Lampyridae) are charismatic insects that have been fruitful model systems in biotechnology. However, lack of information about firefly taxonomy and ecology renders species identification a hard task, especially in the Neotropical region, where fireflies are most diverse. A major gap in the literature on Neotropical fireflies is the lack of knowledge on species’ ecological niches and habitats, which are fundamental aspects for understanding their biology. Here, we provide an annotated checklist of the firefly fauna of the Serra dos Órgãos mountain range (Rio de Janeiro State, Southeastern Brazil), with information on the natural history of each species. We assembled data in three ways: monthly sampling with Malaise traps and active search along an elevational transect from 130 to 2,170 m, over 2 years (2014–2016), extensive field observations, and extracted from historical species records for the Serra dos Órgãos from museum specimens in key collections in Brazil and Europe. We provide a taxonomic key to the genera recorded in the region, and a differential diagnosis for each species, highlighting key references for each taxon. We report 58 species representing 21 genera, making the Serra dos Órgãos one of the richest firefly hotspots on Earth. Most species are restricted to one or two habitat types and/or just one of the regional seasons (warm or cool), and many were only collected either by malaise traps or active search, underlining the importance of sampling different habitat types and seasons, and using different sampling methods when surveying fireflies. Out of the 51 species observed in the field, 49 were active either during the day or the night, although two species – Photuris elliptica and Pyrogaster nigrolineatus – were active in both periods, which is rare in fireflies.     

Mutations in the phospholipid remodeling gene SERAC1 impair mitochondrial function and intracellular cholesterol trafficking and cause dystonia and deafness

Using exome sequencing, we identify SERAC1 mutations as the cause of MEGDEL syndrome, a recessive disorder of dystonia and deafness with Leigh-like syndrome, impaired oxidative phosphorylation and 3-methylglutaconic aciduria. We localized SERAC1 at the interface between the mitochondria and the endoplasmic reticulum in the mitochondria-associated membrane fraction that is essential for phospholipid exchange. A phospholipid analysis in patient fibroblasts showed elevated concentrations of phosphatidylglycerol-34:1 (where the species nomenclature denotes the number of carbon atoms in the two acyl chains:number of double bonds in the two acyl groups) and decreased concentrations of phosphatidylglycerol-36:1 species, resulting in an altered cardiolipin subspecies composition. We also detected low concentrations of bis(monoacyl-glycerol)-phosphate, leading to the accumulation of free cholesterol, as shown by abnormal filipin staining. Complementation of patient fibroblasts with wild-type human SERAC1 by lentiviral infection led to a decrease and partial normalization of the mean ratio of phosphatidylglycerol-34:1 to phosphatidylglycerol-36:1. Our data identify SERAC1 as a key player in the phosphatidylglycerol remodeling that is essential for both mitochondrial function and intracellular cholesterol trafficking.     

Spin-engineering magnetic media

The explosion in demand for increased data-storage density is driving the exploration of new magnetic media. Here we describe a new type of magnetic medium in which the spin configurations are engineered in chemically homogeneous magnetic films: regularly arranged in-plane and out-of-plane spin configurations are defined by altering the magnetic anisotropy. These spin-engineered media not only maintain the surface planarity but also the homogeneity of the magnetic materials, and our method is likely to find immediate application on account of its simplicity and ease of integration.
