Summary The results described here demonstrate that THC-induced catalepsy in mice can be substantially inhibited by the prior administration of 1-THC-7-oic acid, the major metabolite of THC in most species including humans. This raises the possibility that the intensity and duration of action of THC may depend to a large degree on the levels of this metabolite at the sites of action.We thank the National Institute on Drug Abuse for supporting this project by grants DA-02043 and DA-02052 and for supplying all of the cannabinoids. Sex in Caenorhabditis elegans is determined by a regulatory cascade of seven interacting autosomal genes controlled by three X-linked genes in response to the X chromosome-to-autosome (X/A) ratio. XX animals (high X/A) develop as self-fertile hermaphrodites, and XO animals (low X/A) develop as males. The activity of the first gene in the sex-determining cascade, her-1, is required for male sexual development. XO her-1 loss-of-function mutants develop as self-fertile hermaphrodites, whereas XX her-1 gain-of-function mutants develop as masculinized intersexes. By genetic mosaic analysis using a fused free duplication linking her-1 to a cell-autonomous marker gene, we show here that her-1 expression in a sexually dimorphic cell is neither necessary nor sufficient for that cell to adopt a male fate. Our results suggest that her-1 is expressed in many, possibly all, cells and that its gene product can function non-autonomously through cell interactions to determine male sexual development. 相似文献

Structure of an adenine-cytosine base pair in DNA and its implications for mismatch repair 总被引:30,自引:0,他引:30
Mutational pathways rely on introducing changes in the DNA double helix. This may be achieved by the incorporation of a noncomplementary base on replication or during genetic recombination, leading to substitution mutation. In vivo studies have shown that most combinations of base-pair mismatches can be accommodated in the DNA double helix, albeit with varying efficiencies. Fidelity of replication requires the recognition and excision of mismatched bases by proofreading enzymes and post-replicative mismatch repair systems. Rates of excision vary with the type of mismatch and there is some evidence that these are influenced by the nature of the neighbouring sequences. However, there is little experimental information about the molecular structure of mismatches and their effect on the DNA double helix. We have recently determined the crystal structures of several DNA fragments with guanine X thymine and adenine X guanine mismatches in a full turn of a B-DNA helix and now report the nature of the base pairing between adenine and cytosine in an isomorphous fragment. The base pair found in the present study is novel and we believe has not previously been demonstrated. Our results suggest that the enzymatic recognition of mismatches is likely to occur at the level of the base pairs and that the efficiency of repair can be correlated with structural features. 相似文献

The neuropathological hallmarks of Alzheimer's disease and other tauopathies include senile plaques and/or neurofibrillary tangles. Although mouse models have been created by overexpressing specific proteins including beta-amyloid precursor protein, presenilin and tau, no model has been generated by gene knockout. Phosphorylation of tau and other proteins on serine or threonine residues preceding proline seems to precede tangle formation and neurodegeneration in Alzheimer's disease. Notably, these phospho(Ser/Thr)-Pro motifs exist in two distinct conformations, whose conversion in some proteins is catalysed by the Pin1 prolyl isomerase. Pin1 activity can directly restore the conformation and function of phosphorylated tau or it can do so indirectly by promoting its dephosphorylation, which suggests that Pin1 is involved in neurodegeneration; however, genetic evidence is lacking. Here we show that Pin1 expression is inversely correlated with predicted neuronal vulnerability and actual neurofibrillary degeneration in Alzheimer's disease. Pin1 knockout in mice causes progressive age-dependent neuropathy characterized by motor and behavioural deficits, tau hyperphosphorylation, tau filament formation and neuronal degeneration. Thus, Pin1 is pivotal in protecting against age-dependent neurodegeneration, providing insight into the pathogenesis and treatment of Alzheimer's disease and other tauopathies. 相似文献

Transient offsite effects of this construction were apparent as a reduced density of annual plants within 100 m downwind of the edge of the cleared area. Schismus arabicus populations recovered within four years, but Erodium cicutarium populations did not. The effects appeared to be related more to an interaction of seed-dispersal mechanisms with the large, open space than to the obvious sand movement and deposition associated with the clearing operations. 相似文献

Extra-pair copulation and sperm competition in the zebra finch 总被引:2,自引:0,他引:2
Most birds are monogamous, but recent studies have shown that extra-pair copulations (EPCs) occur frequently despite a range of paternity guards, including mate-guarding and frequent copulation. Although EPCs are known to result in extra-pair paternity, there are no previous quantitative estimates of the success of EPCs in fertilizing eggs. We present here estimates of the likelihood of success of extra-pair copulations in a monogamous passerine, the zebra finch Poephila guttata. We show that (1) EPCs occurring under semi-natural conditions in captivity result in extra-pair paternity, (2) sperm from the last male to mate has precedence over previous matings: a single EPC occurring last is disproportionately successful in fertilizing eggs, but EPCs followed by further pair copulations have a low probability of success. Most ion-channels have several kinetically distinct open and closed states. However, a number of groups have reported the presence of sub-conductance states within the open states. It is not clear whether these sub-conductance states represent a partially-occluded open state (with restricted diffusion), or whether they indicate the presence of parallel conductive units. Here we present evidence for the presence of four parallel, equally-conductive subunits that make up a K-selective channel in the apical membrane of early distal tubule in the kidney of Amphiuma. The overall open probability of the channel is controlled by a main gate, the kinetics of which can be described by a simple two-state (open or closed) model. This main gate acts either to occlude or to expose the four subunits in unison. The subunits seem to gate independently both of the main gate and of one another, their open probability obeying the binomial distribution. 相似文献
