The anaerobic oxidation of methane (AOM) with sulphate, an area currently generating great interest in microbiology, is accomplished by consortia of methanotrophic archaea (ANME) and sulphate-reducing bacteria. The enzyme activating methane in methanotrophic archaea has tentatively been identified as a homologue of methyl-coenzyme M reductase (MCR) that catalyses the methane-forming step in methanogenic archaea. Here we report an X-ray structure of the 280?kDa heterohexameric ANME-1 MCR complex. It was crystallized uniquely from a protein ensemble purified from consortia of microorganisms collected with a submersible from a Black Sea mat catalysing AOM with sulphate. Crystals grown from the heterogeneous sample diffract to 2.1?? resolution and consist of a single ANME-1 MCR population, demonstrating the strong selective power of crystallization. The structure revealed ANME-1 MCR in complex with coenzyme M and coenzyme B, indicating the same substrates for MCR from methanotrophic and methanogenic archaea. Differences between the highly similar structures of ANME-1 MCR and methanogenic MCR include a F(430) modification, a cysteine-rich patch and an altered post-translational amino acid modification pattern, which may tune the enzymes for their functions in different biological contexts.  相似文献

Data on plasma sheet crossing measured by Cluster/HIA and Cluster/FGM during the period from July to October in 2001 -2003 are analyzed. Based on previous work on the characteristic features of continuous lobe reconnection (CLR) described in reference, two case studies and a statistical analysis were carried out on correlation between CLR in the mid magnetotail and substorm expansion onset for the events occurring during this period. It is found that almost all CLR events are in close connection with substorms. The beginning of CLR is almost always a few minutes ahead of substorm activities seen in the near Earth magnetotail and on the ground-based stations. This provides a clear indication that CLR is the virtual cause of substorm expansion onset during the period of continuous southward interplanetary magnetic field.  相似文献

Multiple self-healing squamous epithelioma (MSSE), also known as Ferguson-Smith disease (FSD), is an autosomal-dominant skin cancer condition characterized by multiple squamous-carcinoma-like locally invasive skin tumors that grow rapidly for a few weeks before spontaneously regressing, leaving scars. High-throughput genomic sequencing of a conservative estimate (24.2 Mb) of the disease locus on chromosome 9 using exon array capture identified independent mutations in TGFBR1 in three unrelated families. Subsequent dideoxy sequencing of TGFBR1 identified 11 distinct monoallelic mutations in 18 affected families, firmly establishing TGFBR1 as the causative gene. The nature of the sequence variants, which include mutations in the extracellular ligand-binding domain and a series of truncating mutations in the kinase domain, indicates a clear genotype-phenotype correlation between loss-of-function TGFBR1 mutations and MSSE. This distinguishes MSSE from the Marfan syndrome-related disorders in which missense mutations in TGFBR1 lead to developmental defects with vascular involvement but no reported predisposition to cancer.  相似文献

The paper examines Wesley Salmon’s claim that the primary role of plausibility arguments in the history of science is to impose constraints on the prior probability of hypotheses (in the language of Bayesian confirmation theory). A detailed look at Copernicanism and Darwinism and, more briefly, Rutherford’s discovery of the atomic nucleus reveals a further and arguably more important role of plausibility arguments. It resides in the consideration of likelihoods, which state how likely a given hypothesis makes a given piece of evidence. In each case the likelihoods raise the probability of one of the competing hypotheses and diminish the credibility of its rival, and this may happen either on the basis of ‘old’ or ‘new’ evidence.  相似文献

Falsificationism has dominated 20th century philosophy of science. It seemed to have eclipsed all forms of inductivism. Yet recent debates have revived a specific form of eliminative inductivism, the basic ideas of which go back to F. Bacon and J.S. Mill. These modern endorsements of eliminative inductivism claim to show that progressive problem solving is possible using induction, rather than falsification as a method of justification. But this common ground between falsificationism and eliminative inductivism has not led to a detailed investigation into the relationship, if any, which may exist between these two methodologies. This paper reviews several versions of eliminative inductivism, establishes a natural relation between eliminative inductivism and falsificationism, which derives from the distinction between models and theories, and carries out this investigation against a case study of the construction of atom models. The result of the investigation is that falsificationism is a form of eliminative inductivism in the limit of certain constraints.  相似文献
