In 1997-98, fires associated with an exceptional drought caused by the El Ni?o/Southern Oscillation (ENSO) devastated large areas of tropical rain forests worldwide. Evidence suggests that in tropical rainforest environments selective logging may lead to an increased susceptibility of forests to fire. We investigated whether this was true in the Indonesian fires, the largest fire disaster ever observed. We performed a multiscale analysis using coarse- and high-resolution optical and radar satellite imagery assisted by ground and aerial surveys to assess the extent of the fire-damaged area and the effect on vegetation in East Kalimantan on the island of Borneo. A total of 5.2 +/- 0.3 million hectares including 2.6 million hectares of forest was burned with varying degrees of damage. Forest fires primarily affected recently logged forests; primary forests or those logged long ago were less affected. These results support the hypothesis of positive feedback between logging and fire occurrence. The fires severely damaged the remaining forests and significantly increased the risk of recurrent fire disasters by leaving huge amounts of dead flammable wood. 相似文献

When the human immunodeficiency virus transactivating gene under the control of the viral regulatory region is introduced into the germline of mice, skin lesions are induced that resemble Kaposi's sarcoma seen in AIDS. Our findings indicate that HIV could play a direct part in causing cancer. 相似文献

The X-ray crystal structure of the subtilisin-type enzyme proteinase K at 1.5 A resolution shows that is has two binding sites for Ca2+. Scatchard analysis indicates that one Ca2+ binds tightly, with pK 7.6 x 10(-8) M-1, and the other only weakly. Although Ca2+ is not directly involved in the catalytic mechanism and is 16.6 A away from the alpha-carbon atoms of the catalytic triad Asp 39-His 69-Ser 224, the activity of proteinase K towards the synthetic substrate succinyl-Ala-Ala-Ala-p-nitroanilide drops slowly to approximately 20% of its original value when it is depleted of Ca2+. This is not due to autolysis of the enzyme. The X-ray crystal structure of Ca2+-free proteinase K shows that removal of Ca2+ from the tight binding site triggers a concerted domino-like movement of five peripheral loops and of two alpha-helices. At a distance of 25 A from this calcium-binding site, the geometry of both the secondary substrate binding site and of the catalytic triad is affected by this movement thereby reducing the activity of the enzyme. 相似文献

Large amounts of methane are produced in marine sediments but are then consumed before contacting aerobic waters or the atmosphere. Although no organism that can consume methane anaerobically has ever been isolated, biogeochemical evidence indicates that the overall process involves a transfer of electrons from methane to sulphate and is probably mediated by several organisms, including a methanogen (operating in reverse) and a sulphate-reducer (using an unknown intermediate substrate). Here we describe studies of sediments related to a decomposing methane hydrate. These provide strong evidence that methane is being consumed by archaebacteria that are phylogenetically distinct from known methanogens. Specifically, lipid biomarkers that are commonly characteristic of archaea are so strongly depleted in carbon-13 that methane must be the carbon source, rather than the metabolic product, for the organisms that have produced them. Parallel gene surveys of small-subunit ribosomal RNA (16S rRNA) indicate the predominance of a new archael group which is peripherally related to the methanogenic orders Methanomicrobiales and Methanosarcinales. 相似文献

Human T-cell leukaemia virus 1 (HTLV-1) is a retrovirus aetiologically associated with adult T-cell leukaemia (ATL), tropical spastic paraparesis (TSP) and possibly multiple sclerosis (MS) in humans. Three founder lines of transgenic mice containing the HTLV-1 tax gene under the control of the viral long terminal repeat (LTR) have previously been shown to develop neurofibromas. Further analysis of these animals has now revealed that they also develop an exocrinopathy involving the salivary and lachrymal glands. This pathology resembles Sj?gren's syndrome, a disease of presumed autoimmune aetiology, features of which are sometimes reported in HTLV-1 associated conditions. Mice with an HTLV-1 tax transgene might be a useful model for studying the development of Sj?gren-syndrome-like pathology. 相似文献

Significant contribution of Archaea to extant biomass in marine subsurface sediments 总被引:1,自引:0,他引:1
Deep drilling into the marine sea floor has uncovered a vast sedimentary ecosystem of microbial cells. Extrapolation of direct counts of stained microbial cells to the total volume of habitable marine subsurface sediments suggests that between 56 Pg (ref. 1) and 303 Pg (ref. 3) of cellular carbon could be stored in this largely unexplored habitat. From recent studies using various culture-independent techniques, no clear picture has yet emerged as to whether Archaea or Bacteria are more abundant in this extensive ecosystem. Here we show that in subsurface sediments buried deeper than 1 m in a wide range of oceanographic settings at least 87% of intact polar membrane lipids, biomarkers for the presence of live cells, are attributable to archaeal membranes, suggesting that Archaea constitute a major fraction of the biomass. Results obtained from modified quantitative polymerase chain reaction and slot-blot hybridization protocols support the lipid-based evidence and indicate that these techniques have previously underestimated archaeal biomass. The lipid concentrations are proportional to those of total organic carbon. On the basis of this relationship, we derived an independent estimate of amounts of cellular carbon in the global marine subsurface biosphere. Our estimate of 90 Pg of cellular carbon is consistent, within an order of magnitude, with previous estimates, and underscores the importance of marine subsurface habitats for global biomass budgets. 相似文献

Idiopathic infantile arterial calcification (IIAC; OMIM 208000) is characterized by calcification of the internal elastic lamina of muscular arteries and stenosis due to myointimal proliferation. We analyzed affected individuals from 11 unrelated kindreds and found that IIAC was associated with mutations that inactivated ecto-nucleotide pyrophosphatase/phosphodiesterase 1 (ENPP1). This cell surface enzyme generates inorganic pyrophosphate (PP(i)), a solute that regulates cell differentiation and serves as an essential physiologic inhibitor of calcification. 相似文献
