用双极性膜电渗析法分离混合氨基酸   总被引:6,自引:0,他引:6  
为开发新的混合氨基酸分离技术,提出用双极性膜电渗析分离混合氨基酸的方法.在对混合氨基酸溶液中离子成分进行分析的基础上,预测了使用该法的可行性,并通过三室双极性膜电渗析法实验验证了理论计算的结果.对于等电点相差很大的谷氨酸和精氨酸以及等电点相差比较大的谷氨酸和甘氨酸以及精氨酸和甘氨酸两元混合物,可以实现很彻底的分离,得到纯度96%以上的产品,电流密度可以达到5~10 mA·cm-2. 对于等电点相差很小的丝氨酸和脯氨酸,则分离难度较大,电流密度在1 mA·cm-2以下.该法不需使用缓冲溶液,可提高分离过程的效率.  相似文献

The ε subunit of the chloroplast ATP synthase and the truncated ε mutants which lack some amino acid residues from the N-terminus or C-terminus were overexpressed in E. coil When the ε subunit or the truncated ε proteins was added to the spinach chloroplast suspension, both the intensity of the fast phase of millisecond delayed light emission (ms-DLE) and the cyclic and noncyclic photophosphorylation activity of chloroplast were enhanced. With an increase in the number of residues deleted from the N-terminus, the enhancement effect of the N-terminal truncated proteins decreased gradually. For the C-terminal truncated proteins, the enhancement effect increased gradually with an increase in the number of residues deleted from the C-terminus. Besides, the ATP synthesis activity of ε-deficient membrane reconstituted with the ε subunit or the truncated ε proteins was compared. The ATP synthesis activity of reconstituted membrane with the N-terminal truncated proteins decreased gradually as the number of residues deleted from the N-terminus increased. For the C-terminal truncated proteins, the ATP synthesis activity of reconstituted membrane increased gradually with an increase in the number of residues deleted from the C-terminus, but was still lower than that of the wild type ε protein. These results suggested that: (a) the N-terminal domain of the ε subunit of the chloroplast ATP synthase could affect the ATP synthesis activity of ATP synthase by regulating the efficiency of blocking proton leakage of ε subunit; and (b) the C-terminal domain of the ε subunit of the chloroplast ATP synthase had a subtle function in modulating the ATP synthesis ability of ATP synthase.  相似文献

电致化学发光是通过电极上直接或间接发生的电化学反应而产生的一种化学发光,因此电致化学发光检测是在化学发光和电化学基础上发展起来的一种新的分析技术。电致化学发光检测技术不但保留了化学发光分析和电化学分析固有的优点,同时还具有其自身的优点,如所发生的化学发光反应易于控制;方法更灵敏,更具有选择性;可以获得更多的化学信息;扩大了化学发光方法可检测的范围;更易于与现代分离技术联用。生物体中很多生物活性物质具有电活性,因此用现代电化学技术研究其电化学行为具有重要的理论意义和实际应用价值。生物体中的生物活性物质通常浓度很低,并且成分复杂,因此分离检测生物体中生物活性物质非常困难。由于电致化学发光检测具有灵敏度高,选择性好的特点,无疑是检测生物体中生物活性物质的强有力工具,如果它能与HPLC、CE及FIA等现代分离技术相结合,将表现出更加强大的生命力。  相似文献

A stochastic optimal control strategy for partially observable nonlinear quasi Hamiltonian systems is proposed. The optimal control forces consist of two parts. The first part is determined by the conditions under which the stochastic optimal control problem of a partially observable nonlinear system is converted into that of a completely observable linear system. The second part is determined by solving the dynamical programming equation derived by applying the stochastic averaging method and stochastic dynamical programming principle to the completely observable linear control system. The response of the optimally controlled quasi Hamiltonian system is predicted by solving the averaged Fokker-Planck-Kolmogorov equation associated with the optimally controlled completely observable linear system and solving the Riccati equation for the estimated error of system states. An example is given to illustrate the procedure and effectiveness of the proposed control strategy.  相似文献

The salt-gradient operation mode used in ion-exchange simulated moving bed chromatography (SMBC) can improve the efficiency of protein separations. A detailed model that takes into account any kind of adsorption/ion-exchange equilibrium, salt gradient, size exclusion, mass transfer resistance, and port periodic switching mechanism, was developed to simulate the complex dynamics. The model predictions were verified by the experimental data on upward and downward gradients for protein separations reported in the literature. All design and operating parameters (number, configuration, length and diameter of columns, particle size, switching period, flow rates of feed, raffinate, desorbent and extract, protein concentrations in feed, different salt concentrations in desorbent and feed) can be chosen correctly by numerical simulation. This model can facilitate the design, operation, optimization, control and scale-up of salt-gradient ion-exchange SMBC for protein separations.  相似文献

本文提出应用具有灵敏度高、分辨率好、峰高易于准确测量等特点的汞膜电极阳极溶出半微 分电析法进行各种纯水和一些常见支持电解质中痕量重金属杂质的测定方法,并探索通过巯基棉 等分离方法对试剂进行提纯的效果. 实验表明:对于水样只要加入几滴盐酸,使其pH=2.5左右即可进行测定,对于一些纯试剂 可直接进行测定,不必加入其他支持电解质,避免沾污。测定方法十分简便。 利用巯基棉除去一些试剂中痕量铜铅等杂质,进行试剂提纯,方法简便,效果良好。  相似文献

用射频磁控溅射方法制备了一系列颗粒型 N i/Pb不连续多层膜样品 ,并采用 X射线衍射方法表征了样品的微观结构。在铁磁共振实验中 ,观察到了一致进动模 (uniform m ode)和高场光学模 (optical mode)的共振吸收峰。主共振峰和光学模的峰位依赖于外磁场的角度及薄膜厚度。高场光学模的发现表明 ,颗粒型 N i/Pb不连续多层膜存在较强的反铁磁耦合 ,耦合强度随Pb层厚度以指数衰减形式变化。这与面内磁滞回线得到的饱和场随 Pb层厚度的变化关系定性上一致。  相似文献

出力不够是20世纪90年代初投运的第一代中国产循环流化床锅炉的主要问题。为改善75t/h平面流分离循环床锅炉的性能,在原分离器的位置空间采用入口带有加速段的方型分离器代替平面流分离器,适当调整受热面,保留现有炉型和钢构架。改造后的运行实践证明,锅炉出力可以稳定达到额定值,物料循环系统工作正常,主循环回路温度分布合理,改造是成功的。  相似文献

A new method based on the combining of the wavelet theory with the fractal theory and named wavelet fractal peak position method (WFPPM) is introduced to extract the number of the components and the relevant peak positions from overlapping signals in chemistry .The overlapping signal is first transformed into continuous wavelet transform value of time domain in certain dilation range via continuous wavelet transform (CWT),and then changed into capacity dimensions (Dc),The number of the components and the relevant positions of overlapping peaks can be identified easily according to the change of Dc.An investigation concerning the influence of different dilation ranges on the peak positions extracted by WFPPM is also provided .Studies show that the WFPPM is an efficient tool for extracting the peak positions and identifying the number of peaks from unresoloved signals,even when this kind of overlapping is significantly serious,Relative errors of lless than 1.0% in peak positions are found when WFPPM is used in the processing of the cadmium (Ⅱ)-indium(Ⅲ) mixture system .The analytical results demonstrate that the desired peak positions can be extracted conveniently ,accurately and rapidly from an unresoled signal via WFPPM,Tremendous developing and applications based on currently reported WFPPM in extracting overlapping signals would be expected in the near future.  相似文献
