Overexpression and export of Vibrio anguillarum metalloprotease in Escherichia coli

Vibrio anguillarum metalloprotease, an extracellular zinc metalloprotease involved in the virulence mechanism of Vibrio anguillarum, is synthesized from the empA gene as a 611-residue precursor and naturally secreted via Sec secretion pathway in Vibrio anguillarum. In this study, heterologous expression of the empA gene encoding metalloprotease and export of the recombinant metalloprotease in Escherichia coli were examined. The empA gene was subcloned into pBAD24 with arabinose promoter and sequenced. The sequence encoded a polypeptide (611 amino acids) consisting of four domains: a signal peptide, an N-terminal propeptide, a mature region and a C-terminal propeptide. The empA gene inserted in plasmid pBAD24 was overexpressed in TOP10 strain of E. coli after arabinose induction. The 36kDa polypeptide of the recombinant metalloprotease as the mature protease was further confirmed by SDS-PAGE and immunoblotting. It was found that recombinant metalloprotease with the EmpA activity and antigenicity was exported into the periplasm of Escherichia coli cells via Sec translocation pathway, whereas it was secreted into extracellular environments in V. anguillarum. The results imply that the expression, export and processing mechanism of the protein in E. coli are similar to those in V. anguillarum.     

近50年内蒙古多伦县气候变化特征分析

以1961～2005年多伦县气象站气象记录为主要数据来源,选取气温、降水量、蒸发量、干湿度等指标,采用常规统计分析方法,进行了年均和季度变化的时间序列分析.结果表明:多伦县年均气温趋于上升趋势,降水量波动较大,并且夏季降水量对全年降水量贡献最大.年均蒸发量总趋势是下降的,整体上比较稳定.干湿度趋于上升,呈现变湿润趋势.近50年来多伦县气候呈现增温增湿的趋势.     

Problems in wavelet analysis of hydrologic series and some suggestions on improvement

Applying the wavelet theory and methods to investigate the hydrologic processes such as precipitation and runoff is a hot field. However, several aspects in research are usually ignored: the effect of admissible condition of wavelet functions and the disturbance of noises for the detection of periods, the effect of the length of a hydrologic time-series on the final result, and the choice between the anomaly and the original time series for wavelet analysis. In this paper, these issues are fully discussed. Precipitation data from Lanzhou Precipitation Station are taken for case study. The result indicates that in the wavelet analysis of hydrologic series, denoise methods should be used to eliminate the influence of noises. The MexHat wavelet function satisfies the admissible condition, which ensures that the periodic properties of hydrologic processes can be well represented by using the MexHat wavelet for decomposition. The affected range of hydro-logic series which should be discarded before analysis is given. It is also suggested that the anomaly series should be used to highlight the actual undulation of the hydrologic series.     

化工基础实验教学改革实践

化工基础实验在理科化学专业的工程技术教育中是一个重要的实践性教学环节。为提高理科学生学习的积极性,提高教学效果,武汉大学从两个方面对化工基础实验进行改革实践:将实验内容尽可能和学院科研成果相结合,尽可能引入化工新技术实验,并开设一些与生活相关的实验,让学生感觉到化工实验不是枯燥的计算与设备;改革教学方法,提高学生主动学习的积极性。通过数年的实践,取得明显教学效果。     

内镜下氩离子凝固术治疗食管贲门失弛缓症疗效观察

目的：探讨氩离子凝固术治疗贲门失弛缓症的疗效。方法：选择2010年1月至2012年1月经食道吞钡及内镜检查诊断的贲门失弛缓症12例患者进行内镜下环行点状治疗后临床观察,治疗单次启动延续时间1～3 s,启动次数视局部组织凝固的深度而定,一般环行凝固4个点,深度0.2~0.3 cm为宜,术中局部喷1∶5去甲肾上腺素盐水10 mL,术后加用止血及抗感染药物及进流质饮食3 d,观察1周出院,所有病例1~3月内复查食道吞钡及胃镜检查。结果：12例患者中2例失访,其余10例患者均复查,临床症状消失,食道吞钡、胃镜下未见复发。结论：氩离子凝固术治疗是治疗贲门失弛缓症的新方法,操作方便,疗效较好,值得推广应用。     

基于Delphi的深海物探设备管理系统设计

在深海石油勘探船队的设备管理中,设备信息的及时交流和沟通成为影响船队正常生产的关键因素,在这其中,网络和计算机技术发挥了重要的作用。通过对船队设备管理流程的研究和分析,结合船队设备管理的实际操作,利用Delphi7和SQL Server 2000并采用了三层C/S结构设计一种深海作业船队设备管理系统。系统通过用户管理、设备管理、文件管理、供应商信息管理、维修商信息管理这五个管理模块,有效的解决了设备管理效率低下,实现了企业设备管理系统的简单化、规范化、合理化、科学化,改变了传统的纸笔以及windows office单机管理模式,很大程度上提高了企业设备管理、使用及维修的效率。
