Immunogenicity of a highly attenuated MVA smallpox vaccine and protection against monkeypox

The potential use of smallpox as a biological weapon has led to the production and stockpiling of smallpox vaccine and the immunization of some healthcare workers. Another public health goal is the licensing of a safer vaccine that could benefit the millions of people advised not to take the current one because they or their contacts have increased susceptibility to severe vaccine side effects. As vaccines can no longer be tested for their ability to prevent smallpox, licensing will necessarily include comparative immunogenicity and protection studies in non-human primates. Here we compare the highly attenuated modified vaccinia virus Ankara (MVA) with the licensed Dryvax vaccine in a monkey model. After two doses of MVA or one dose of MVA followed by Dryvax, antibody binding and neutralizing titres and T-cell responses were equivalent or higher than those induced by Dryvax alone. After challenge with monkeypox virus, unimmunized animals developed more than 500 pustular skin lesions and became gravely ill or died, whereas vaccinated animals were healthy and asymptomatic, except for a small number of transient skin lesions in animals immunized only with MVA.     

Ectopic beta-chain of ATP synthase is an apolipoprotein A-I receptor in hepatic HDL endocytosis

The effect of high-density lipoprotein (HDL) in protecting against atherosclerosis is usually attributed to its role in 'reverse cholesterol transport'. In this process, HDL particles mediate the efflux and the transport of cholesterol from peripheral cells to the liver for further metabolism and bile excretion. Thus, cell-surface receptors for HDL on hepatocytes are chief partners in the regulation of cholesterol homeostasis. A high-affinity HDL receptor for apolipoprotein A-I (apoA-I) was previously identified on the surface of hepatocytes. Here we show that this receptor is identical to the beta-chain of ATP synthase, a principal protein complex of the mitochondrial inner membrane. Different experimental approaches confirm this ectopic localization of components of the ATP synthase complex and the presence of ATP hydrolase activity at the hepatocyte cell surface. Receptor stimulation by apoA-I triggers the endocytosis of holo-HDL particles (protein plus lipid) by a mechanism that depends strictly on the generation of ADP. We confirm this effect on endocytosis in perfused rat liver ex vivo by using a specific inhibitor of ATP synthase. Thus, membrane-bound ATP synthase has a previously unsuspected role in modulating the concentrations of extracellular ADP and is regulated by a principal plasma apolipoprotein.     

Metamorphic core complex formation by density inversion and lower-crust extrusion.

Metamorphic core complexes are domal uplifts of metamorphic and plutonic rocks bounded by shear zones that separate them from unmetamorphosed cover rocks. Interpretations of how these features form are varied and controversial, and include models involving extension on low-angle normal faults, plutonic intrusions and flexural rotation of initially high-angle normal faults. The D'Entrecasteaux islands of Papua New Guinea are actively forming metamorphic core complexes located within a continental rift that laterally evolves to sea-floor spreading. The continental rifting is recent (since approximately 6 Myr ago), seismogenic and occurring at a rapid rate ( approximately 25 mm yr-1). Here we present evidence-based on isostatic modelling, geological data and heat-flow measurements-that the D'Entrecasteaux core complexes accommodate extension through the vertical extrusion of ductile lower-crust material, driven by a crustal density inversion. Although buoyant extrusion is accentuated in this region by the geological structure present-which consists of dense ophiolite overlaying less-dense continental crust-this mechanism may be generally applicable to regions where thermal expansion lowers crustal density with depth.     

Homology of proteasome subunits to a major histocompatibility complex-linked LMP gene.

The class II region of the major histocompatibility complex (MHC) contains genes encoding at least two subunits of a large, intracellular protein complex (the low molecular mass polypeptide, or LMP, complex). This complex is biochemically similar to the proteasome, an abundant and well conserved protein complex having multiple proteolytic activities. Here we report the isolation of a complementary DNA corresponding to one of the subunits of the LMP complex, LMP-2. The protein predicted from this cDNA sequence closely matches the amino-terminal peptide sequence of a rat proteasome subunit, confirming that the proteasome and the LMP complex share polypeptide subunits. The LMP-2 gene is tightly linked to HAM1, a gene thought to be required for translocating peptide fragments of endogenous antigens into the endoplasmic reticulum for association with MHC class I molecules. These observations suggest that the LMP complex may be responsible for generating peptides from cytoplasmic antigen during antigen processing.     

Evidence for a different site of action of phenoxybenzamine and desmethylimipramine on the catecholamine uptake system

Resumen El tratamiento con fenoxibenzamina o desmetilimipramina da lugar a una potenciación irreversible de las respuestas a noradrenalina, en la aurícula aislada de cobayo. La presencia de tiramina durante el tratamiento previene dicha potenciación cuando se utiliza fenoxibenzamina, mientras que no la modifica en el caso de la desmetilimipramina     

Differential effects of prostaglandins and isoproterenol on cAMP content and Na, K pump activity in rat submandibular acini

The beta-adrenergic agonist isoproterenol and prostaglandins E1 and E2 (but not F2 alpha) increased the cAMP content of rat submandibular acini in vitro, but only isoproterenol enhanced ouabain-sensitive 86Rb (K) uptake. These findings suggest that cAMP is not involved in the activation of the Na, K pump in salivary cells.
